The course of decline of vitality with age due to the process of senescence, when not complicated by the process of growth, follows a simple exponential law; that is the degree of vitality or of senescence (defining vitality as the reciprocal of senescence) at any moment is, regardless of age, a constant percentage of the degree of vitality or senescence of the preceding moment. This exponential law is the same as the law of monomolecular change in chemistry. During the actively growing period of life the index of vitality rises, due to the process of growth and the course of vitality in the case when the growing period is included in the vitality curve, follows a rising and falling course. This rising and falling course may often be represented by an equation containing two exponential terms which is practically the equation used to represent the course of accumulation and disappearance of a substance as the result of two simultaneous consecutive monomolecular chemical reactions.  相似文献

Catalase activity is demonstrated histochemically in the microbodies of aerated cultures of Euglena gracilis strain Z grown on inorganic media supplemented with acetate or glucose. Although this enzyme can also be assayed photometrically in cell-free extracts of acetate-supplemented cells, it is below the level of detectability in extracts of glucose-supplemented cells, there being an order of magnitude fewer microbodies in the latter than the former. Even acetate-supplemented cultures (dark-grown, greening, or continuously light-grown) fail to exhibit detectable catalase activity when CO2 is removed from the air by Ascarite.Negative results were obtained with histochemical techniques considered optimal for the demonstration of cytochrome oxidase; under other conditions, however, a KCN-sensitive enzyme was revealed in the mitochondrial matrix. This (unidentified) enzyme is first observed in mitochondria after 20–24 hr of greening, reaches a maximum intensity at about 48 hr, and becomes undetectable by 72 hr of greening. Poisoning of photosynthesis by 3-(3,4-dichlorophenyl)-1, 1-dimethylurea (DCMU) results in loss of activity of this mitochondrial enzyme.  相似文献

A morphological mutant (col-2) of Neurospora, which is partially deficient in glucose-6-phosphate dehydrogenase (G-6-PD) activity and has lower levels of reduced nicotinamide adenine dinucleotide phosphate (NADPH), accumulated three-fold more triglycerides during log-phase growth than the wild-type strain. Increased lipid deposition was not found in other strains that included slow-growing morphological mutants, NADPH-deficient strains, G-6-PD-deficient mutants, wild-type revertants from col-2, and a cel, col-2 double mutant. The cel, col-2 strain was supplemented with an exogenous source of fatty acids because it cannot synthesize these lipid moieties. The observed normal lipid content of this strain suggests that the lipid deposition in col-2 on glucose is due to an overstimulation of fatty acid synthesis and not a deficiency in fatty acid breakdown. The neutral lipid levels in both wild type and col-2 were decreased to identical levels when grown on glutamate as a carbon source. This effect was not due to changes in glutamic dehydrogenase levels. The omission of citrate from the glutamate medium reduced wild-type neutral lipid levels even further, but had no effect on col-2. The variations with time in the neutral lipid levels of col-2 upon changes in these carbon sources are presented, as well as a discussion of the possible types of regulatory effects unique to the col-2 mutation which might affect fatty acid synthesis.  相似文献

Purely mechanical factors involved in the dynamics of the thin-walled, spherical viscus may be elucidated by studying the behavior of an artificial viscus. An equation is developed on theoretical grounds which expresses the pressure-distention characteristics of such a model and apparently corrects the errors contained in similar equations developed by other investigators. A precise definition for wall “tone” is offered. On this basis the mathematics of “tone”-distention relationships have been developed. Experimental investigation of the artificial viscus (spherical rubber balloon) permits the differences in behavior between the ideal and the actual model to be composed, and confirms the validity of our theoretical developments.
