Abstract: The structure determination of the glutamate dehydrogenase from the hyperthermophile Pyrococcus furiosus has been completed at 2.2 Å resolution. The structure has been compared with the glutamate dehydrogenases from the mesophiles Clostridium symbiosum, Escherichia coli and Neurospora crassa . This comparison has revealed that the hyperthermophilic enzyme contains a striking series of networks of ion-pairs which are formed by regions of the protein which contain a high density of charged residues. Such regions are not found in the mesophilic enzymes and the number and extent of ion-pair formation is much more limited. The ion-pair networks are clustered at both inter domain and inter subunit interfaces and may well represent a major stabilising feature associated with the adaptation of enzymes to extreme temperatures.  相似文献

Glutamate dehydrogenase from Clostridium symbiosum displays unusual kinetic behaviour at high pH when compared with other members of this enzyme family. Structural and sequence comparisons with GDHs from other organisms have indicated that the Asp residue at position 114 in the clostridial enzyme may account for these differences. By replacing this residue by Asn, a mutant protein has been created with altered functional properties at high pH. This mutant protein can be efficiently overexpressed in Escherichia coli, and several criteria, including mobility in non-denaturing electrophoresis, circular dichroism (CD) spectra and initial crystallisation studies, suggest a folding and an assembly comparable to those of the wild-type protein. The D114N mutant enzyme shows a higher optimum pH for activity than the wild-type enzyme, and both CD data and activity measurements show that the distinctive time-dependent reversible conformational inactivation seen at high pH in the wild-type enzyme is abolished in the mutant.  相似文献

The antioxidant efficacy of alpha-carotene and comparison with beta-carotene in multilamellar liposomes prepared from egg yolk phosphatidyl choline (EYPC) exposed to the lipid soluble 2,2'-azobis (2,4-dimethyl valeronitrile) (AMVN) was investigated. Lipid peroxidation was measured as thiobarbituric acid reacting substances (TBARS) at 532 nm or as hydroperoxide formation at 234 nm after separation of phosphatidyl choline hydroperoxide (PCOOH) by high-pressure liquid chromatography (HPLC). Lutein and zeaxanthin, the hydroxyl derivatives of alpha- and beta-carotenes, and the chain breaking antioxidant alpha-tocopherol were also included in the study. AMVN being a lipid soluble, non polar azo initiator penetrates into the hydrophobic interior of the phospholipid bilayer, forming peroxyl radicals which peroxidate the phospholipid leading to PCOOH accumulation. All the carotenoids tested at 1 mol% relative to EYPC significantly suppressed the formation of PCOOH compared to control samples. In this system, alpha-carotene retarded PCOOH formation better than beta-carotene. Similarly, lutein was a better antioxidant than is zeaxanthin. But lutein and zeaxanthin were more effective antioxidants than alpha- and beta-carotenes, respectively. After 1 h of incubation of the carotenoid with AMVN, alpha-, beta-carotene, lutein and zeaxanthin limited PCOOH formation by 77%, 68%, 85% and 82%, respectively, while alpha-tocopherol elicited 90% reduction. AMVN incubated with EYPC for 2 h induced the formation of TBARS compared to control (P < 0.001). alpha-Carotene significantly suppressed the TBARS formation by 78% whilst beta-carotene, lutein, zeaxanthin and alpha-tocopherol elicited 60%, 91% and 80% reductions, respectively. Increasing the concentration of the carotenoid > 1 mol% to EYPC did not significantly increase protection of the membrane against free radical attack. Our findings suggest that alpha-carotene is a better antioxidant than is beta-carotene in phosphatidyl choline vesicles. It may, therefore, be useful in limiting free radical mediated peroxidative damage against membrane phospholipids in vivo.  相似文献

We develop a method to estimate the expected time of survival of a predator population as a function of the size of the habitat island on which it lives and the dynamic parameters of the population and its prey. The model may be thought of either as a patch occupancy model for a structured population or as a model of metapopulation type. The method is applied to a keystone predator species, the neotropical army ant Eciton burchelli. Predictions are made as to how many of the islands and habitat islands in and around Gatun Lake in the Panama Canal, most of which were formed when the canal was dug, can be expected to support such a population today, and these are compared with data.  相似文献

Temperate regions of Australia have extensive commercial plantations of Monterey pine, Pinus radiata D. Don. Replacement of indigenous forests by P. radiata is likely to have significant effects on assemblages of native Lepidoptera, and has been considered a major threat to native fauna through displacing native species. However, many species of Lepidoptera, including ennomine geometrid moths in the genus Chlenias Guenèe, have adopted P. radiata as a larval host. Comparisons were made of oviposition preferences and nutritional ecology of Chlenias auctaria Guenée on P. radiata and two native host plants [Acacia mearnsii De Wild. (Mimosaceae) and Eucalyptus obliqua LHérit (Myrtaceae)]. Females showed no significant oviposition preference for any of the three hosts. Growth of sixth instar larvae was significantly less on P. radiata than on the native hosts. Pupal weights were significantly lower, suggesting that the fitness of moths reared on P. radiata will be significantly reduced. The life history strategy of C. auctaria, which includes dispersal of first instar larvae by ballooning, may predispose this species to feed on poor quality hosts, and this may be common to other polyphagous species found feeding on P. radiata in southern hemisphere plantations. The impact of P. radiata may have a long term effect on lepidopteran communities, beyond the simple replacement of indigenous host plants leading to extirpations of feeding specialists.  相似文献

Allocation of organic carbon (OC) to primary energetic pathways was estimated under seasonal and artificially elevated ambient temperatures for a field population of a freshwater pulmonate snail, Physella virgata. Allocation to respiration increased with temperature. Snails allocated most assimilated OC to reproduction within their natural temperature range (15 degrees -35 degrees C), where assimilation efficiencies remained relatively stable at 25%-35%. However, in artificially heated waters exceeding 35 degrees C, declining assimilation rates and increasing respiratory demands inhibited allocation to reproduction and growth. At the species' 40 degrees C upper thermal limit, assimilation efficiencies fell below 10%, while average consumption levels more than doubled relative to snails unaffected by the thermal effluent. Ambient temperature substantially influenced OC allocation over P. virgata's natural temperature range and negatively affected growth and reproduction at temperatures approaching or exceeding maximum natural levels.  相似文献

The decision-making process that determines when an animal should switch between tasks is a fundamental issue in the study of animal behaviour. We investigated, for the first time, such task switching in terms of the dynamics of worker populations in ant colonies. During colony emigration in the ant Leptothorax albipennis, the colony has to carry out the following three tasks: (1) transport of brood and nestmates to the new nest; (2) sorting of the brood into its characteristic pattern; and (3) building the nest wall. At the beginning of the emigration, the stimuli for all three tasks increase simultaneously but the tasks are performed sequentially by populations of workers in the colony. The issue here is how decision making at the colony level is based on the behaviour of individual workers. We used a mathematical model to explore the hypothesis that such colony level task switching is based on tasks competing for workers. The essential feature of this model is that the sequence of tasks performed by an individual worker need not match the sequence of tasks on which the colony concentrates. We base the parameterization of our model on our detailed experimental study of eight emigrations, one for each of eight L. albipennis colonies. Recent experiments have demonstrated the importance of numerical assessment in animal contests. Nevertheless, few attempts have been made to model explicitly the relationship between the relative number of combatants on each side and the costs and benefits of entering a contest. One framework that may be especially suitable for making such explicit predictions is Lanchester's theory of combat, which has proved useful for understanding combat strategies in humans and several species of ants. We show, with data from a recent series of playback experiments, that a model derived from Lanchester's 'square law' predicts willingness to enter intergroup contests in wild chimpanzees (Pan troglodytes). Furthermore, the model predicts that, in contests with multiple individuals on each side, chimpanzees in this population should be willing to enter a contest only if they outnumber the opposing side by a factor of 1.5. We evaluate these results for intergroup encounters in chimpanzees and also discuss potential applications of Lanchester's square and linear laws for understanding combat strategies in other species.  相似文献

Structural maintenance of chromosomes (SMC) proteins are found in nearly all organisms. Members of this protein family are involved in chromosome condensation and sister chromatid cohesion. Bacillus subtilis SMC protein (BsSMC) plays a role in chromosome organization and partitioning. To better understand the function of BsSMC, we studied the effects of an smc null mutation on DNA supercoiling in vivo. We found that an smc null mutant was hypersensitive to the DNA gyrase inhibitors coumermycin A1 and norfloxacin. Furthermore, depleting cells of topoisomerase I substantially suppressed the partitioning defect of an smc null mutant. Plasmid DNA isolated from an smc null mutant was more negatively supercoiled than that from wild-type cells. In vivo cross-linking experiments indicated that BsSMC was bound to the plasmid. Our results indicate that BsSMC affects supercoiling in vivo, most likely by constraining positive supercoils, an activity which contributes to chromosome compaction and organization.  相似文献
