Administration of 4-acetylaminofluorene (4AAF) to rats by oral gavage (1000 mg/kg) produces a wave of S-phase activity in the liver 36 h later, followed by a wave of mitoses at 48 h. These events were monitored by autoradiography of isolated hepatocytes and by histopathology, respectively. DNA-labelling was shown to occur following both in vivo and in vitro radiolabelling. The level of S-phases observed approached that reported following partial hepatectomy. These effects were not accompanied by unscheduled DNA synthesis (UDS) nor was any frank histopathological damage to the liver evident. 2-Acetylaminofluorene (2AAF) elicited a very weak S-phase response at a dose level of 50 mg/kg, but gave marked UDS between 12 and 48 h.  相似文献

Abstract Restriction enzyme analysis typing with Hin fI, Hae III and Pst I was performed on Moraxella (Branhamella) catarrhalis strains consecutively collected from children suspected of respiratory tract infection and the type strain. Use of Hin fI gave the most distinct patterns. Great polymorphism was seen among strains.  相似文献

Glutamate excretion due to amino acid starvation was investigated in “stringent” and “relaxed” strains ofEscherichia coli. The observed excretion process isrelA-dependent, carrier-mediated, and glutamate-specific. After induction, excretion was detected within less than 2 min and continued for more than 5h with a rate of 7–10 nmol (mg dry weight)−1 min−1. Using carbonyl cyanidem-chlorophenylhydrazone or polymyxin B nonapeptide, together with valinomycin, it was shown that glutamate excretion is driven by the membrane potential.  相似文献

Isolabelling only occurs in CHO cells that have been allowed to replicate for more than 2 but less than 3 cell divisions in the presence of BrdU. The isolabelling is confined to late replicating regions of the chromosomes. The staining patterns obtained indicate that BrdU was incorporated three times in these regions and that the isolabelling did not come from the segregation of label in polynemic chromosomes.  相似文献

Ethanol (68.2 mM) did not appreciably affect the growth of Dipodascus aggregatus with glucose (55.5 mM] as carbon source. Growth with fructose was inhibited whereas growth with galactose was stimulated by ethanol in this concentration. The fungus could grow with ethanol as the sole carbon source. D. aggregatus did not grown with maltose as the sole carbon source. Growth with maltose + ethanol started much earlier than growth with ethanol alone. Nonanal (80 μ) in ethanolic solution stimulated the growth of Dipodascus aggregatus with fructose (55.5 mM) as carbon source (inoculum grown with fructose or glucose). If the inoculum had been grown with galactose, neither growth with galactose nor growth with glucose was affected by nonanal. If the inoculum had been grown with glucose, growth with galactose was weakly. stimulated. —Growth with galactose (galactose-grown inoculum) was strongly stimulated by nonanal if xylose at a low concentration (0.53 mM) was added. — The oxygen uptake of glucose grown cells with glucose as substrate was stimulated by 200 μM nonanal in the absence of ethanol. The respiratory activity of galactose-grown cells was also stimulated with galactose as well as with glucose as substrate. In the absence of exogenous substrate the oxygen uptake of glucose-grown cells was weakly stimulated by nonanal whereas that of galactose-grown cells was strongly stimulated.  相似文献

The interactions between field layer vascular plants andSphagnum mosses in peat-forming systems are discussed in terms of differences in growth strategies, access to light, acquisition of mineral nutrients and water and the processes involved in the formation of the micro-topographical structures characteristic for these systems. To keep pace with the vertical growth ofSphagnum, the co-occurring vascular plants require a growth strategy involving continuous movement of the growing point and meristematic tissue upwards and a frequent formation of adventitious roots. The growth form and architecture of the vascular plants determine the occurrence and distribution of the structural units on a mire, the hummocks, lawns and hollows. Dwarf shrubs and other vascular plants with an orthotropic growth pattern characterise hummocks, where they form a firm matrix which reinforces and supports the spongy biomass ofSphagnum. In a similar way, clonal herbs stabilise the lawns because of the predominantly plagiotropic, or only weakly orthotropic, growth pattern of the rhizomes and coarse roots in the upper, oxic layers. Extended periods of drought often may have deleterious effects on the mosses but smaller impacts on the vascular plants because of their more efficient water conducting system. Different sources of mineral nutrients are used bySphagnum (atmospheric deposition) and the vascular plants (mineralisation of the organic matter). The presence ofSphagnum, therefore, reduces the supply of nutrient resources to the vascular plants.Sphagnum thereby gains a competitive advantage. A high rate of mineralisation would be beneficial to the vascular plants by increasing their growth rates causing shading of theSphagnum mosses and covering the moss layer by the above-ground litter fall. However, the slow decomposition ofSphagnum litter keeps the system in balance as it will reduce the nutrient supply to the vascular plants.  相似文献

The genotoxic activity of the benzidine-derived azo dye, Direct Black 38 (DB38), was studied in vivo, using two different genetic end-points: unscheduled DNA synthesis in liver (UDS) and bone marrow micronucleus (MN). Exposure times were 12, 24 or 36 h. Both assays were performed in the same rat, except for the 24-h exposure when only MN was investigated. For the liver UDS assay, the rat hepatocarcinogen, 6-dimethylaminophenylazobenzthiazole (6BT), was used as positive control and for the MN assay, cyclophosphamide (CP). In agreement with earlier results, 6BT gave rise to a dose-related increase in liver UDS after 12-h exposure to 25 or 50 mg/kg bw. After 36-h exposure, there was still an indication of a weak dose-response effect between 0 and 5 net nuclear grains (NG). DB38 induced liver UDS at the higher dose levels used (500 and 1000 mg/kg), and after both 12- and 36-h exposure. With the longer exposure time, a weak induction of UDS was also observed at 100 mg/kg. The strongest UDS induction (12.2 NG), was obtained in one rat after 36-h exposure to 500 mg/kg. DB38 also had a weak effect on the MN induction, which was statistically significant at the higher concentrations used. A dose-related response was observed at all exposure times used.  相似文献

The recent literature on the bioavailability of different forms of P in freshwater systems is reviewed. Bioavailable P is defined as the sum of immediately available P and the P that can be transformed into an available form by naturally occurring processes. Methods used to estimate the bioavailable P pool, which vary between studies largely depending on the time perspective applied, are critically evaluated. Most studies on particulate P aim to determine the potentially available P pool. Potential bioavailability of particulate P is normally analysed in bioassays with algal yield determinations and the available P fraction is characterized from interpretations of results of sequential chemical extractions. NaOH-extractable P is in most studies the most algal-available P fraction. For soil samples and tributary water particulate matter, NaOH-P has often been found to be equal to algal extractable P. In other studies depletions of NaOH-P have accounted for the algal P uptake, but only a minor proportion of the fraction has been utilized. Organic P in lake water particulate matter and bed sediments of eutrophic lakes can also be algal-available to a significant extent.Studies on the bioavailability of dissolved P have often been concerned with immediate availability, or the minimum amount of available P. Such studies need other types of experimental design and normally assays with radiotracers are used. Immediately available P is frequently found to be less than P chemically assessed as dissolved reactive P (DRP) at low (< 10 µg DRP·l-1) concentrations. However, immediate availability may also approach or exceed DRP concentrations, especially at higher concentrations. Potential bioavailability, assayed as for particulate P, may generally render higher bioavailability than P assayed as immediately available. Large fractions of dissolved P remain unutilized and are primarily found in the high molecular weight fraction of dissolved P.  相似文献
