The nature of the modification of yeast ribosomes by the recombinant form of the ricin A chain has been examined. Evidence is presented that the 26S rRNA molecule is depurinated at a specific site and that the activity is inhibited by antibody raised to ricin A chain. It thus appears that the recombinant form of this toxin retains the depurination activity of the native molecule. These results are consistent with the model that the site of depurination is in a highly conserved sequence forming a loop on the surface of the ribosome, a domain involved in elongation factor-dependent binding of aminoacyl-tRNA.  相似文献

(5E)-6a-Carbaprostaglandin I2 (carbacyclin) was oxidized to (5E)-15-dehydro-6a-carbaprostaglandin I2 (15-dehydrocarbacyclin) by partially purified rhesus monkey lung prostaglandin dehydrogenase (PGDH). The (5E)-15-dehydro-6a-carbaprostaglandin I2 was isolated by preparative thin-layer chromatography and identified by gas chromatography-mass spectrometry. A Lineweaver-Burke plot gave an apparent Km value of 2.9 μM and a Vmax of 35.7 nmoles carbacyclin oxidized/mg protein/min. These values are similar to previously reported Km and Kmax values for PGI2 and PGE1.  相似文献

The interaction of the purified C1q component of human C with synchronized, quiescent human gingival fibroblasts was investigated, and the presence of a specific binding site was demonstrated. Quantitative binding studies with radioiodinated C1q showed that binding was specific, saturable, and reversible upon addition of unlabeled C1q or by increasing the salt concentration. Scatchard plot analysis of the data yielded an affinity constant of 2 X 10(7) M-1 for all cell strains examined. The capacity for C1q binding varied among the eight cell strains examined. The number of binding sites per cell ranged from 2.6 to 17.7 X 10(6) with an average of 8.4 X 10(6). The receptor was insensitive to trypsin digestion, and it bound the collagen-like portion of the C1q molecule. Specific immunofluorescence staining showed that virtually all the viable cultured fibroblasts were able to bind added C1q. Flow cytometric analysis demonstrated a spectrum of fluorescence intensity among the cell strains, and there was a positive correlation between fluorescence intensity and the number of binding sites detected by using radiolabeled C1q.  相似文献

Between 1975 and 1983, adult female vervet monkeys (Cercopithecus aethiops sabaeus) over 3.5 years of age, living in two undisturbed social groups in a captive colony in Sepulveda, California, have averaged 1.0 births per female year with a mean interbirth interval of 10.7 months. Increased fecundity did not result in decreased survival rates of offspring in this population. Fecundity was influenced by the mother's age and dominance rank. The primary factor in the age-fecundity relationship was the age at first birth, which varied from three to five years. High-ranking females contributed the most to the high rate of fecundity, with significantly shorter interbirth intervals, more births per female year, and more surviving infants compared to low-ranking females.  相似文献

Among nonhuman primates the composition of social groups influences the interactions of group members. We assessed the effects of acute changes in social composition on behavior among 15 adult male vervet monkeys (Cercopithecus aethiops sabaeus). Subjects were observed in their basal social groups which comprised 3 adult males, 2–4 adult females, and offspring; and in two subgroups consisting of either two or three adult males. Agonism and vigilance increased in smaller groups relative to basal conditions, while subjects in two-male groups displayed more aggression than those in three-male groups. These findings suggest that, among male vervet monkeys, acute disruption of stable social groups increases aggressive behavior, and that the amount of agonism is influenced by the composition of the consequent subgroups. © 1992 Wiley-Liss, Inc.  相似文献
