The hemlock woolly adelgid, Adelges tsugae Annand is an invasive insect that frequently causes hemlock (Tsuga spp.) mortality in the eastern United States. Studies have shown that once healthy hemlocks become infested by the adelgid, nutrients are depleted from the tree, leading to both tree decline and a reduction of the adelgid population. Since A. tsugae is dependent on hemlock for nutrients, feeding on trees in poor health may affect the ability of the insect to obtain necessary nutrients and may consequently affect their physiological and population health. Trees were categorized as lightly or moderately impacted by A. tsugae based on quantitative and qualitative tree health measurements. Population health of A. tsugae on each tree was determined by measuring insect density and peak mean fecundity; A. tsugae physiological health was determined by measuring insect biomass, total carbon, carbohydrate, total nitrogen, and amino nitrogen levels. Adelges tsugae from moderately impacted trees exhibited significantly greater fecundity than from lightly impacted trees. However, A. tsugae from lightly impacted hemlocks contained significantly greater levels of carbohydrates, total nitrogen, and amino nitrogen. While the results of the physiological analysis generally support our hypothesis that A. tsugae on lightly impacted trees are healthier than those on moderately impacted trees, this was not reflected in the population health measurements. Adelges tsugae egg health in response to tree health should be verified. This study provides the first examination of A. tsugae physiological health in relation to standard A. tsugae population health measures on hemlocks of different health levels.  相似文献

The endoplasmic reticulum (ER) has the ability to signal organelle dysfunction via a complex signaling network known as the unfolded protein response (UPR). In this work, hamster fibroblast cells exhibiting moderate levels of ER stress were compared to those exhibiting severe ER stress. Inhibition of N-linked glycosylation was accomplished via a temperature-sensitive mutation in the Dad1 subunit of the oligosaccharyltransferase (OST) complex or by direct inhibition with tunicamycin (Tm). Temperature shift (TS) treatment generated weak activation of ER stress signaling when compared to doses of Tm that are typically used in ER stress studies (500–1000 nM). A dose-response analysis of key ER stress signaling mediators, inositol-requiring enzyme 1 (IRE1) and protein kinase R (PKR)-like endoplasmic reticulum kinase (PERK), revealed 20–40 nM of Tm to generate activation intensity similar to TS treatment. In parental BHK21 cells, moderate (20–40 nM) and high doses (200–1000 nM) of Tm were compared to identify physiological and signaling-based differences in stress response. Inhibition of ER Ca2+ release via ITPR activity with 2-aminoethoxydiphenyl borate (2-APB) or Xestospongin C (XeC) was sufficient to protect against apoptosis induced by moderate but not higher doses of Tm. Analysis of kinase activation over a range of Tm exposures revealed the p38 stress-activated protein kinase (SAPK) to display increasing activation with Tm dosage. Interestingly, Tm induced the extracellular regulated kinases (Erk1/2) only at moderate doses of Tm. Inhibition of ER transmembrane stress sensors (IRE1, PERK) or cytosolic signaling mediators (p38, Jnk1, Erk1/2) was used to evaluate pathways involved in apoptosis activation during ER stress. Inhibition of either PERK or p38 was sufficient to reduce cell death and apoptosis induced by moderate, but not high, doses of Tm. During ER stress, cells exhibited a rapid decline in anti-apoptotic Mcl-1 and survivin proteins. Inhibition of PERK was sufficient to block this affect. This work reveals moderate doses of ER stress to generate patterns of stress signaling that are distinct from higher doses and that apoptosis activation at moderate levels of stress are dependent upon PERK and p38 signaling. Studies exploring ER stress signaling should recognize that this signaling acts as a rheostat rather than a simple switch, behaving distinctively in a dose-dependent manner.  相似文献

The first cells probably possessed rudimentary metabolic networks, built using a handful of multifunctional enzymes. The promiscuous activities of modern enzymes are often assumed to be relics of this primordial era; however, by definition these activities are no longer physiological. There are many fewer examples of enzymes using a single active site to catalyze multiple physiologically‐relevant reactions. Previously, we characterized the promiscuous alanine racemase (ALR) activity of Escherichia coli cystathionine β‐lyase (CBL). Now we have discovered that several bacteria with reduced genomes lack alr, but contain metC (encoding CBL). We characterized the CBL enzymes from three of these: Pelagibacter ubique, the Wolbachia endosymbiont of Drosophila melanogaster (wMel) and Thermotoga maritima. Each is a multifunctional CBL/ALR. However, we also show that CBL activity is no longer required in these bacteria. Instead, the wMel and T. maritima enzymes are physiologically bi‐functional alanine/glutamate racemases. They are not highly active, but they are clearly sufficient. Given the abundance of the microorganisms using them, we suggest that much of the planet's biochemistry is carried out by enzymes that are quite different from the highly‐active exemplars usually found in textbooks. Instead, primordial‐like enzymes may be an essential part of the adaptive strategy associated with streamlining.  相似文献

Californian Salt-Marsh Vegetation: An Improved Model of Spatial Pattern   总被引:6,自引:0,他引:6  
Although tidal wetland vegetation patterns are typically related to elevation, we hypothesized that the vertical range of a species may shift where the topography is more heterogeneous. We examined plant species occurrences in relation to elevation, proximity to the bay, and proximity to tidal creeks at a near-pristine wetland in San Quintín Bay, Baja California, Mexico. At the whole-wetland scale, most species occurred primarily within a 30-cm elevation band (the marsh plain). However, Spartina foliosa occurred only at the bayward margin, even though “suitable” elevations were present further inland. A similar pattern was found in San Diego Bay. At the microtopographic scale, three species on the marsh plain were strongly influenced by elevation, whereas four species responded to both elevation and proximity to tidal creeks. The latter species tended to “avoid” the lower 10 cm of the marsh plain except near a tidal creek. Species richness was thus greater (by 0.6 species at the lowest 10-cm class) at the tidal creek margin. Better drainage near creeks is the hypothesized cause. Our results help explain why species that are transplanted to constructed wetlands do not always grow at the full range of elevations they occupy in natural wetlands. We recommend that species be introduced to their modal elevation (determined from nearby reference marshes) and that salt-marsh construction designs include topographic heterogeneity (complex tidal creek networks). The analysis of broad-scale and fine-scale patterns of occurrence also suggests new habitat nomenclature. Elevation-based terms (“low,”“middle,” and “high” marsh) should be replaced by a system that considers elevation, landscape position, and conspicuous species. We suggest three habitat designations: (a) the high marsh—a 30- to 70-cm elevation range with Salicornia subterminalis; (b) the marsh plain—a 30-cm elevation range with heterogeneous topography and up to nine common species; and (c) cordgrass habitat—the bayward portion of the marsh plain and lower elevations, all occupied by Spartina foliosa. Although these habitats do not have discrete boundaries, separate terms are needed for wetland restoration plans and these designations will improve recognition that vegetation patterns respond to horizontal, as well as vertical, position.  相似文献

Non-Indigenous Woody Invasive Plants in a Rural New England Town   总被引:3,自引:0,他引:3  
We investigated the abundance of non-indigenous woody invasive plants in Farmington, Maine, a rural New England town in a forested landscape. We found 12 invasive species and more than 7 patches per km from surveys on 33 transects (54.3 km) along field edges, abandoned railroad right-of-ways, roadsides, and riparian zones. Invasive abundance was apparently lower than for more developed areas of the northeastern US, where, in contrast to western Maine, invasives have extensively penetrated forest interiors. Invasive abundance increased with the amount of landscaping and proximity to town, suggesting a close association between local horticulture and the spread of woody invasives. Invasive abundance and diversity were highest in riparian areas, probably due to relatively high levels of propagule pressure. Species differed in the extent of invasiveness, ranging from those still dependent on planted parent trees to fully invasive populations. The invasive species recorded in this study have caused environmental and economic damage elsewhere. The lower levels of invasiveness in Farmington are likely a result of the isolation, small human population, and forested landscape rather than low levels of invasibility. This suggests the potential for future risks, and the importance of intervention while populations can still be eradicated or controlled.  相似文献

ABSTRACT.   Recent advances in the methods used to estimate detection probability during point counts suggest that the detection process is shaped by the types of cues available to observers. For example, models of the detection process based on distance-sampling or time-of-detection methods may yield different results for auditory versus visual cues because of differences in the factors that affect the transmission of these cues from a bird to an observer or differences in an observer's ability to localize cues. Previous studies suggest that auditory detections predominate in forested habitats, but it is not clear how often observers hear birds prior to detecting them visually. We hypothesized that auditory cues might be even more important than previously reported, so we conducted an experiment in a forested habitat in North Carolina that allowed us to better separate auditory and visual detections. Three teams of three observers each performed simultaneous 3-min unlimited-radius point counts at 30 points in a mixed-hardwood forest. One team member could see, but not hear birds, one could hear, but not see, and the third was nonhandicapped. Of the total number of birds detected, 2.9% were detected by deafened observers, 75.1% by blinded observers, and 78.2% by nonhandicapped observers. Detections by blinded and nonhandicapped observers were the same only 54% of the time. Our results suggest that the detection of birds in forest habitats is almost entirely by auditory cues. Because many factors affect the probability that observers will detect auditory cues, the accuracy and precision of avian point count estimates are likely lower than assumed by most field ornithologists.  相似文献

Background  

Y-chromosomal haplogroup (Y-HG) Q is suggested to originate in Asia and represent recent founder paternal Native American radiation into the Americas. This group is delineated into Q1, Q2 and Q3 subgroups defined by biallelic markers M120, M25/M143 and M3, respectively. Recently, a novel subgroup Q4 has been identified which is defined by bi-allelic marker M346, representing HG Q (0.41%, 3/728) in Indian population. With scanty details of HG Q in Asia, especially India, it was pertinent to explore the status of the Y-HG Q in Indian population to gather an insight to determine the extent of diversity within this region.  相似文献

The literature concerning eye migration and cranial development during flatfish metamorphosis is briefly reviewed and found to give rise to five hypotheses to account for the phenomenon. The cranial morphology of specimens of Soleidae, Pleuronectidae, Scophthalmidae and Bothidae in pre-metamorphic, metamorphic and post-metamorphic stages are described. The mechanisms by which eye migration and cranial development occur are discussed and some aspects of pleuronectiform relationships are briefly commented on.  相似文献
