In periodontitis, polymorphonuclear leucocytes (PMNs) are activated. They entrap and eliminate pathogens by releasing neutrophil extracellular traps (NETs). Abnormal NET degradation is part of a pro-inflammatory status, affecting co-morbidities such as cardiovascular disease. We aimed to investigate the ex vivo NET degradation capacity of plasma from periodontitis patients compared to controls (part 1) and to quantify NET degradation before and after periodontal therapy (part 2). Fresh NETs were obtained by stimulating blood-derived PMNs with phorbol 12-myristate 13-acetate. Plasma samples from untreated periodontitis patients and controls were incubated for 3 h onto freshly generated NETs (part 1). Similarly, for part 2, NET degradation was studied for 91 patients before and 3, 6 and 12 mo after non-surgical periodontal therapy with and without adjunctive systemic antibiotics. Finally, NET degradation was fluorospectrometrically quantified. NET degradation levels did not differ between periodontitis patients and controls, irrespective of subject-related background characteristics. NET degradation significantly increased from 65.6 ± 1.7% before periodontal treatment to 75.7 ± 1.2% at 3 mo post periodontal therapy, and this improvement was maintained at 6 and 12 mo, irrespective of systemic usage of antibiotics. Improved NET degradation after periodontitis treatment is another systemic biomarker reflecting a decreased pro-inflammatory status, which also contributes to an improved cardiovascular condition.  相似文献

Humid tropical forests play a dominant role in many global biogeochemical cycles, yet long-term records of tropical stream chemistry and its response to disturbance events such as severe storms and droughts are rare. Here we document the long-term variability in chemistry of two streams in the Luquillo Mountains, Puerto Rico over a period of 27 years. Our two focal study watersheds, the Río Icacos and Quebrada Sonadora, both drain several hundred hectares of tropical wet forests, and each received direct hits from Hurricanes Hugo (1989) and Georges (1998). They differ primarily in lithology (granitic vs. volcaniclastic) and elevation. Changes in major cations, anions, silica, and dissolved organic carbon were minimal over the study period, but the concentrations of nitrate show a strong response to hurricane disturbance and the longest time to recovery. Potassium also showed a large, although less consistent, response to disturbance. In the granitic terrain, nitrate concentrations exceeded long-term pre-hurricane background levels for over a decade, but were elevated in the volcaniclastic terrain for only 1–2 years. Lithology appears to be the primary driver explaining the different response trajectories of the two watersheds. In the granitic terrain, which showed slow recovery to pre-hurricane conditions, the quartz diorite bedrock weathers to produce coarser soils, deeper groundwater flow paths, and riparian zones with sharp spatial variation in redox conditions and very high nitrogen levels immediately adjacent to the stream. Groundwater flow paths are shallow and the levels of N in streamside groundwater are much lower in the volcaniclastic terrain. The recovery of vegetation following hurricane disturbance appears similar in the two watersheds, suggesting that the extent of structural damage to canopy trees determines the magnitude of NO3 increases, but that the duration of elevated concentrations in stream water is a function of lithology.  相似文献

Cell–cell junction remodeling is associated with dramatic actin reorganizations. Several actin regulatory systems have been implicated in actin remodeling events as cell–cell contacts are assembled and disassembled, including zyxin/LPP–VASP complexes. These complexes facilitate strong cell–cell adhesion by maintaining actin-membrane connections. It has been proposed that zyxin and LPP localize to cell–cell junctions via a well-defined interaction with alpha-actinin. This was recently confirmed for LPP, but zyxin localization at cell–cell contacts occurs independently of alpha-actinin binding. Here we seek to map the zyxin sequence responsible for localization to cell–cell contacts and identify the protein that docks zyxin at this cellular location. Previous results have shown that a zyxin fragment excluding the alpha-actin binding site and the LIM domains (amino acids 51–392) can independently localize to cell–cell contacts. Here, expression of smaller zyxin fragments show that zyxin localization requires amino acids 230–280. A yeast-two-hybrid screen, using the central region of zyxin as bait, resulted in the identification of the cell–cell adhesion receptor nectin-4 as a zyxin binding partner. Further demonstrating zyxin–nectin interactions, zyxin binds the intracellular domain of nectin-2 in vitro. Depletion of nectin-2 from L cells expressing E-cadherin results in a loss of zyxin localization to cell–cell contacts, demonstrating that the zyxin–nectin interaction plays a critical role in zyxin targeting to these sites.  相似文献

Escherichia coli heat-labile enterotoxin (LT) is a powerful mucosal adjuvant; however, it is associated with toxic effects when delivered intranasally, and its mechanism of action is poorly understood. In this article, we demonstrate that LT acts as a highly effective adjuvant when administered parenterally, promoting Ag-specific IL-17, as well as IFN-γ, IL-4, and IL-10 production in response to coadministered Ags. We found that the adjuvant activity of LT was mediated in part by inducing dendritic cell (DC) activation; LT promoted CD80 and CD86 expression by DCs and enhanced IL-1α, IL-1β, and IL-23 production. An LT mutant, LTK63, that lacks enzyme activity was less effective than the wild-type toxin in promoting DC maturation and the development of Ag-specific Th17 cells. LT enhanced IL-23 and IL-1α production from DCs via activation of ERK MAPK and IL-1β secretion through activation of caspase-1 and the NLRP3 inflammasome. These cytokines played a major role in promoting Th17 responses by LT and LTK63. The induction of Th17 cells in vivo in response to LT and LTK63 as adjuvants was significantly reduced in IL-1RI-deficient mice. Finally, using a murine respiratory infection model, we demonstrated that LT can act as a highly effective adjuvant for a pertussis vaccine, promoting Ag-specific Th17 cells and protection against Bordetella pertussis challenge, which was significantly reduced in IL-17-defective mice. Our findings provide clear evidence that LT can promote protective immune responses in part through induction of innate IL-1 and, consequently, Th17 cells.  相似文献

Lagoon of Islands was a rare wetland of high conservation value, supporting floating islands of vegetation before it was dammed to supply water for consumptive use downstream. After damming, water quality declined to such an extent that in 2013, the dam wall was removed and a wetland restoration project was commenced, resulting in immediate improvements in water quality and, within five years, regeneration of a plant community largely composed of dominant species of the original wetland.  相似文献

Many butterfly species that were historically common throughout the UK are thought to have declined due to land use intensification, whilst others have increased regionally due to climate change. Population trends in these species are inadequately assessed by current monitoring programmes: the UK Butterfly Monitoring Scheme (UKBMS) mainly samples semi-natural habitats; coarse-scale distribution recording under-estimates declines in population abundance. We designed a scheme (the wider countryside butterfly survey—WCBS) to provide unbiased abundance trends for widespread butterfly species across the whole countryside. The WCBS has been developed over a 3 year pilot study, culminating in a UK-wide roll out in 2009. After testing a range of survey methods, the scheme involves visiting randomly selected 1-km squares at least twice during July–August and walking a linear transect route, counting the number of individual butterflies seen within 5 m. A method for volunteer recorders to collect abundance data on non-lepidopteran insects was also tested. The results of the first year of the WCBS and future plans for the scheme will be described.  相似文献

This paper considers the measurement of voice quality variations relating to three different performance styles of early music singing by a trained soprano. In particular, the various outputs available following the collection of electrolaryngographic data are presented along with acoustic analyses based on the acoustic pressure waveform to enable a comparison of the three different singing styles. Voice quality analysis is based on the output from the electrolaryngograph, that enables a measurement of fundamental period to 1 μs accuracy to be made, enabling fundamental frequency variations including vibrato to be explored. In addition, the percentage of each cycle for which the vocal folds are in contact is measured and varies between the three voice qualities providing insights into the nature of vocal fold vibration in each case. Acoustic output spectral measures are made that are not only based on traditional wide- and narrow-band but also hearing modelling spectrography. These provide a clear indication as to which portions of the spectrum are most prominent in terms of perception of the three different voice qualities. A number of features resulting from the various analyses distinguish the singing styles, thereby providing evidence that the methods used are appropriate for objective characterisation and differentiation of singing styles in early music.  相似文献

Climate warming threatens the survival of species at their warm, trailing‐edge range boundaries but also provides opportunities for the ecological release of populations at the cool, leading edges of their distributions. Thus, as the climate warms, leading‐edge populations are expected to utilize an increased range of habitat types, leading to larger population sizes and range expansion. Here, we test the hypothesis that the habitat associations of British butterflies have expanded over three decades of climate warming. We characterize the habitat breadth of 27 southerly distributed species from 77 monitoring transects between 1977 and 2007 by considering changes in densities of butterflies across 11 habitat types. Contrary to expectation, we find that 20 of 27 (74%) butterfly species showed long‐term contractions in their habitat associations, despite some short‐term expansions in habitat breadth in warmer‐than‐usual years. Thus, we conclude that climatic warming has ameliorated habitat contractions caused by other environmental drivers to some extent, but that habitat degradation continues to be a major driver of reductions in habitat breadth and population density of butterflies.  相似文献

Ecological responses to climate change may depend on complex patterns of variability in weather and local microclimate that overlay global increases in mean temperature. Here, we show that high‐resolution temporal and spatial variability in temperature drives the dynamics of range expansion for an exemplar species, the butterfly Hesperia comma. Using fine‐resolution (5 m) models of vegetation surface microclimate, we estimate the thermal suitability of 906 habitat patches at the species' range margin for 27 years. Population and metapopulation models that incorporate this dynamic microclimate surface improve predictions of observed annual changes to population density and patch occupancy dynamics during the species' range expansion from 1982 to 2009. Our findings reveal how fine‐scale, short‐term environmental variability drives rates and patterns of range expansion through spatially localised, intermittent episodes of expansion and contraction. Incorporating dynamic microclimates can thus improve models of species range shifts at spatial and temporal scales relevant to conservation interventions.  相似文献
