In colonies of the queenless ant Streblognathus peetersi, dominance interactions produce a reproductive hierarchy in which one individual, the alpha, is capable of producing offspring
while her subordinates remain infertile. Based on differences between behaviour and cuticular hydrocarbon profiles, the subordinates
can be further divided into high and low ranking workers. Although it had been shown previously that alphas treated with a
juvenile hormone analog lose their reproductive status, little was known of the endocrinological basis of dominance in this
species. To elucidate the underlying endocrinology of these three ranks, we measured the individual in vitro rate of juvenile
hormone (JH) production of excised corpora allata, and the ecdysteroid titer of pooled hemolymph samples. Production of JH
was highest in low-ranking workers, intermediate in high rankers, and almost undetectable in alphas. Ecdysteroid titers were
low for low rankers, but were more than twice as high for both high rankers and alphas. The results support the hypothesis
that JH suppresses ovarian function in these queenless ants, and suggest that ecdysteroids may be responsible for stimulating
vitellogenin production. The possible role of these hormones as behavioural modulators is also discussed.

Crystal structures of protein-tyrosine phosphatase 1B in complex with compounds bearing a novel isothiazolidinone (IZD) heterocyclic phosphonate mimetic reveal that the heterocycle is highly complementary to the catalytic pocket of the protein. The heterocycle participates in an extensive network of hydrogen bonds with the backbone of the phosphate-binding loop, Phe(182) of the flap, and the side chain of Arg(221). When substituted with a phenol, the small inhibitor induces the closed conformation of the protein and displaces all waters in the catalytic pocket. Saturated IZD-containing peptides are more potent inhibitors than unsaturated analogs because the IZD heterocycle and phenyl ring directly attached to it bind in a nearly orthogonal orientation with respect to each other, a conformation that is close to the energy minimum of the saturated IZD-phenyl moiety. These results explain why the heterocycle is a potent phosphonate mimetic and an ideal starting point for designing small nonpeptidic inhibitors. 相似文献

The Notch signaling pathway is an evolutionarily conserved intercellular signaling mechanism, and mutations in its components disrupt embryonic development in many organisms and cause inherited diseases in humans. We previously described construction and analysis of a hypomorphic allele of the Notch2 gene. Homozygosity for this allele leads to embryonic and perinatal lethality due to cardiovascular and kidney defects. We report here novel Notch2 mutant alleles generated by gene targeting in embryonic stem cells, including a conditional null allele in which exon 3 of the Notch2 gene is flanked by loxP sequences. These new Notch2 mutant alleles expand the set of tools available for studying the myriad roles of the Notch pathway during mammalian development and will enable analysis of Notch2 function at additional stages of embryogenesis and in adult mice. 相似文献

Neural signatures of economic preferences for risk and ambiguity 总被引:9,自引:0,他引:9
People often prefer the known over the unknown, sometimes sacrificing potential rewards for the sake of surety. Overcoming impulsive preferences for certainty in order to exploit uncertain but potentially lucrative options may require specialized neural mechanisms. Here, we demonstrate by functional magnetic resonance imaging (fMRI) that individuals' preferences for risk (uncertainty with known probabilities) and ambiguity (uncertainty with unknown probabilities) predict brain activation associated with decision making. Activation within the lateral prefrontal cortex was predicted by ambiguity preference and was also negatively correlated with an independent clinical measure of behavioral impulsiveness, suggesting that this region implements contextual analysis and inhibits impulsive responses. In contrast, activation of the posterior parietal cortex was predicted by risk preference. Together, this novel double dissociation indicates that decision making under ambiguity does not represent a special, more complex case of risky decision making; instead, these two forms of uncertainty are supported by distinct mechanisms. 相似文献

Neuromuscular training protocols that include both plyometrics and dynamic balance exercises can significantly improve biomechanics and neuromuscular performance and reduce anterior cruciate ligament injury risk in female athletes. The purpose of this study was to compare the effects of plyometrics (PLYO) versus dynamic stabilization and balance training (BAL) on power, balance, strength, and landing force in female athletes. Either PLYO or BAL were included as a component of a dynamic neuromuscular training regimen that reduced measures related to ACL injury and increased measures of performance. Nineteen high school female athletes participated in training 3 times a week for 7 weeks. The PLYO (n = 8) group did not receive any dynamic balance exercises and the BAL (n = 11) group did not receive any maximum effort jumps during training. Pretraining vs. posttraining measures of impact force and standard deviation of center of pressure (COP) were recorded during a single leg hop and hold. Subjects were also tested for training effects in strength (isokinetic and isoinertial) and power (vertical jump). The percent change from pretest to posttest in vertical ground reaction force was significantly different between the BAL and PLYO groups on the dominant side (p < 0.05). Both groups decreased their standard deviation of center of pressure (COP) during hop landings in the medial/lateral direction on their dominant side, which equalized pretested side to side differences. Both groups increased hamstrings strength and vertical jump. The results of this study suggest that both PLYO and BAL training are effective at increasing measures of neuromuscular power and control. A combination of PLYO and BAL training may further maximize the effectiveness of preseason training for female athletes. 相似文献

CD8+ T cells are crucial for control of a number of medically important protozoan parasites, including Trypanosoma cruzi, the agent of human Chagas disease. Yet, in contrast to the wealth of information from viral and bacterial infections, little is known about the antigen specificity or the general development of effector and memory T-cell responses in hosts infected with protozoans. In this study we report on a wide-scale screen for the dominant parasite peptides recognized by CD8+ T cells in T. cruzi-infected mice and humans. This analysis demonstrates that in both hosts the CD8+ T-cell response is highly focused on epitopes encoded by members of the large trans-sialidase family of genes. Responses to a restricted set of immunodominant peptides were especially pronounced in T. cruzi-infected mice, with more than 30% of the CD8+ T-cell response at the peak of infection specific for two major groups of trans-sialidase peptides. Experimental models also demonstrated that the dominance patterns vary depending on the infective strain of T. cruzi, suggesting that immune evasion may be occurring at a population rather than single-parasite level. 相似文献

Cribellar prey capture threads found in primitive, horizontal orb-webs reflect more light, including ultraviolet wavelengths, than viscous threads found in more derived, vertical orb-webs. Low web visibility and vertical orientation are each thought to increase prey interception and may represent key innovations that contributed to the greater diversity of modern, araneoid orb-weaving spiders. This study compares prey interception rates of cribellate orb-webs constructed by Uloborus glomosus (Uloboridae) with viscous orb-webs constructed by Leucauge venusta (Tetragnathidae) and Micrathena gracilis (Araneidae). We placed sectors of cribellar and viscous threads side by side in frames that were oriented either horizontally or vertically. The webs of both U. glomosus and L. venusta intercepted more prey when vertically oriented. In each orientation L. venusta webs intercepted more insects than did U. glomosus. Although this is consistent with the greater visibility of cribellar threads, the more closely spaced capture spirals of L. venusta may have contributed to this difference. Micrathena gracilis webs intercepted more prey than did U. glomosus webs, although web orientation did not affect the performance of this araneoid species. The stickier and more closely spaced capture spirals of M. gracilis may have enhanced the interception rates of this species and accounted for the greater number of smaller dipterans retained in its webs. The tendency for these slow, weak flight insects to be blown into both horizontal and vertical webs may account for similar interception rates of horizontal and vertical M. gracilis webs. These observations support the enhanced prey interception of vertically oriented orb-webs, but offer only qualified support for the contributions of lower visibility viscous capture threads. 相似文献
