The viable bacterial particle size distribution in the atmosphere at the Hanford Nuclear Reservation, Richland, WA during two, 1-week periods in June 1992, was observed at three intervals during the day (morning, midday and evening) and at three heights (2, 4, and 8 m) above ground level. The distributions were significantly different (P=0.01) between the two, 1-week sampling periods and between morning, midday, and evening observations, but not between the three heights. Approximately 30 to 50% fell into the largest particle size category; 7.0µm aerodynamic diameter. All particle size categories were at their minimum bacterial concentration at around noon, with the lowest concentrations in the smaller size categories (<2.1µm aerodynamic diameter). This suggests, that at this high desert location, solar radiation (SR) damage to airborne bacteria is particle size discriminatory. There is a relatively greater effect on the smaller size categories at midday and a relatively lesser effect in the morning and evening.  相似文献

Among the South American Pleistocene Glyptodontidae (Xenarthra, Cingulata), one of the most scarcely represented genus in the fossil record is Neuryurus Ameghino, in which the only well characterized species, until now, is Neuryurus rudis (Gervais), coming from the Ensenadan (early-middle Pleistocene) of the Pampean region, Argentina. In this contribution, we report and describe a new species of Neuryurus, coming from the Arroyo Feliciano Formation (early Lujanian; ca. 130 ka), Entre Ríos province, Argentina, in a particular palaebiogeographic context, more associated with the western sector of Uruguay and southern Brazil than the Pampean region. From a morphological perspective, this new taxon is characterized by: (a) the remarkable development of the foramina at the exposed surface of the osteoderms of the dorsal carapace, showing un aspect almost spongy; (b) the large diameter of the perforations observed in the ventral surface of the osteoderms, resembling to those present in the dorsal surface of the osteoderms of the Glyptodontidae Doedicurinae; (c) the evident thickness of the osteoderms, as in Glyptodon Owen; (d) area of contact and articulation of adjacent osteoderms very denticulate, with deep osseous interdigitating projections, as in Glyptodon and Glyptotherium Osborn, but here more evident; (e) larger osteoderms of the dorsal carapace showing, in the dorsal surface, many little conical tubercles, having some resemblance to Panochthus tuberculatus Burmeister. The presence of this new species in the early late Pleistocene of the Mesopotamic region is another element that adds to the biogeographical characterization of this particular area, especially evident during the warm and humid pulses of the late Pleistocene. In turn, this species represents the most complete record of the genus outside the Pampean region and partially complete the stratigraphic and geographic distribution of the genus.  相似文献

Summary Quiescent normal human mesothelial (NHM) cells will undergo one round of DNA synthesis when they are incubated in a defined medium consisting of LHC basal medium supplemented with hydrocortisone, insulin, transferrin, and one of the following peptide mitogens: epidermal growth factor; transforming growth factor beta (1 or 2); platelet derived growth factor (a,b heterodimer or b,b homodimer); fibroblast growth factor (acid or basic forms); interleukin 1 (alpha or beta forms); interleukin 2; interferon gamma; interferon beta; or cholera toxin. However, sustained cell multiplication does not occur unless the medium contains hydrocortisone, insulin, transferrin, any one of the above-listed peptide growth factors and high density lipoproteins. Growth can be increased twofold if the medium contains certain combinations of these mitogens and high density lipoproteins. The finding that NHM cells can respond to a broad spectrum of growth factors supports the possibility that an autocrine mechanism may be part of the mechanism that leads to transformation of these cells by asbestos.  相似文献

Two-dimensional (2D) gel electrophoresis of immunoprecipitated HLA-DR antigens from eight homozygous typing cells (HTC) expressing the HLA-DRw8 specificity revealed a clustering of polymorphic chain patterns into distinct electrophoretic variants. The variant patterns correlate with three discrete HLA-D clusters that are defined in the mixed leukocyte culture reaction (MLR) using DRw8-positive HTC. These HLA-D clusters have been provisionally designated Dw8.1, detected primarily in Caucasoids, Dw8.2, detected primarily in American Indians, and Dw8.3, detected predominantly in Orientals. All three HLA-Dw8.1 cell lines express a single DR-locus product as defined by immunoprecipitation with a DR-specific monoclonal antibody, P4.1. This DR chain is identical among the Dw8.1 cell lines and different from the DR chains of the Dw8.2 and Dw8.3 cell lines. Two separate Dw8.2 HTC express a shared DR chain that is slightly more basic than the 8.1 DR molecule; interestingly, one of these lines also expresses an additional DR-like chain not found in the other cells. Thus, the two lines defining the Dw8.2 cluster share one distinct class 11 molecule, but differ in another and therefore are not biochemically HLA-identical. Cells from the Dw8.3 cluster are likewise distinct from all other Dw8 clusters. One additional DRw8-positive HTC has been analyzed and found to be distinct from the Dw8.1, 8.2 and 8.3 clusters by both MLR and 2D gels. lmmunoprecipitates using monoclonal antibody 1B5 [anti-DR and anti-DQ(DS)] identify additional polymorphic class II variants among the cell lines tested. These data indicate that HLA-DRw8 is a public serologic specificity present on class II molecules expressed on multiple distinct haplotypes. These haplotypes differ from each other in expression of polymorphic class II molecules encoded by at least two HLA loci. They also differ in HLA-D, even though they all type as HLA-DRw8 homozygous. In Dw8.2, variation in expressed chains is not reflected in variation in HLA-D, indicating that MLR, as well as serologic typing, does not detect the full degree of allelic polymorphism within HLA.  相似文献

The PII protein has been considered pivotal to the dual cascade regulating ammonia assimilation through glutamine synthetase activity. Here we show that PII, encoded by the glnB gene, is not always essential; for instance upon ammonia deprivation of a glnB deletion strain, glutamine synthetase can be deadenylylated as effectively as in the wild-type strain. We describe a new operon, glnK amtB , which encodes a homologue of PII and a putative ammonia transporter. We cloned and overexpressed glnK and found that the expressed protein had almost the same molecular weight as PII, reacted with polyclonal PII antibody, and was 67% identical in terms of amino acid sequence with Escherichia coli PII. Like PII, purified GlnK can activate the adenylylation of glutamine synthetase in vitro , and, in vivo , the GlnK protein is uridylylated in a glnD -dependent fashion. Unlike PII, however, the expression of glnK depends on the presence of UTase, nitrogen regulator I (NRI), and absence of ammonia. Because of a NRI and a σN54) RNA polymerase-binding consensus sequence upstream from the glnK gene, this suggests that glnK is regulated through the NRI/NRII two-component regulatory system. Indeed, in cells grown in the presence of ammonia, glutamine synthetase deadenylylation upon ammonia depletion depended on PII. Possible regulatory implications of this conditional redundancy of PII are discussed.  相似文献

Assay of centromere function using a human artificial chromosome   总被引:8,自引:0,他引:8  
In order to define a functional human centromere sequence, an artificial chromosome was constructed as a reproducible DNA molecule. Mammalian telomere repeats and a selectable marker were introduced into yeast artificial chromosomes (YACs) containing alphoid DNA from the centromere region of human chromosome 21 in a recombination-deficient yeast host. When these modified YACs were introduced into cultured human cells, a YAC with the alphoid DNA from the α21-I locus, containing CENP-B boxes at a high frequency and a regular repeat array, efficiently formed minichromosomes that were maintained stably in the absence of selection and bound CENP-A, CENP-B, CENP-C and CENP-E. The minichromosomes, 1–5 Mb in size and composed of multimers of the introduced YAC DNA, aligned at metaphase plates and segregated to opposite poles correctly in anaphase. Extensive cytological analyses strongly suggested that the minichromosomes had not acquired host sequences and were formed in all cases by a de novo mechanism. In contrast, minichromosomes were never produced with a modified YAC containing alphoid DNA from the α21-II locus, which contains no CENP-B boxes and has a less regular sequence arrangement. We conclude that α21-I alphoid DNA can induce de novo assembly of active centromere/kinetochore structures on minichromosomes. Received: 22 August 1998 / Accepted: 28 August 1998  相似文献

RING (Really Interesting New Gene)‐in‐between‐RING (RBR) enzymes are a distinct class of E3 ubiquitin ligases possessing a cluster of three zinc‐binding domains that cooperate to catalyse ubiquitin transfer. The regulation and biological function for most members of the RBR ligases is not known, and all RBR E3s characterized to date are auto‐inhibited for in vitro ubiquitylation. Here, we show that TRIAD1 and HHARI, two members of the Ariadne subfamily ligases, associate with distinct neddylated Cullin‐RING ligase (CRL) complexes. In comparison to the modest E3 ligase activity displayed by isolated TRIAD1 or HHARI, binding of the cognate neddylated CRL to TRIAD1 or HHARI greatly stimulates RBR ligase activity in vitro, as determined by auto‐ubiquitylation, their ability to stimulate dissociation of a thioester‐linked UBCH7~ubiquitin intermediate, and reactivity with ubiquitin‐vinyl methyl ester. Moreover, genetic evidence shows that RBR ligase activity impacts both the levels and activities of neddylated CRLs in vivo. Cumulatively, our work proposes a conserved mechanism of CRL‐induced Ariadne RBR ligase activation and further suggests a reciprocal role of this special class of RBRs as regulators of distinct CRLs.  相似文献

Previously starved urchins, Lytechinus variegatus, (36.0 ± 0.8 (SE) mm test diameter) were held in replicated (3) 10-L aquaria with artificial seawater at 22 ± 2  °C and 32‰ salinity and fed three diet treatments. Urchins were fed diets containing 9 : 35, 20 : 23 or 31 : 12% dry protein: % dry carbohydrate (P : C) ad libitum for a 65-day period. Gonads from urchins fed the 9 : 35 P : C diet had similar organic, lower ash, and lower water content than urchins fed the 31 : 12 P : C protein diet. Water content varied with both diet and nutritional history; consequently, water content may have limited value as a predictor of gonad nutritional status. Protein and carbohydrate concentrations in the gonad were directly related to the dietary composition of these nutrients; gonad lipids did not vary with diet. Excess carbohydrates are frequently stored as fats in fish and mammals but this does not appear to be the case in L. variegatus. Test carbohydrate storage and gut protein storage also reflected dietary composition. Image analysis of ovaries indicated decreased nutritive phagocyte volume, increased germinal epithelium volume and larger oocyte diameters in urchins fed high protein, low carbohydrate diets. Analysis of testes also indicated decreased nutritive phagocyte volume and increased gamete volume with urchins fed high protein, low carbohydrate diets, but differences among treatments were less obvious than in ovaries. This study suggests that high protein, low carbohydrate diets promote gamete growth and development. In addition, the biochemical and gametic composition of gonads can be altered by manipulating dietary composition. This could affect the quality and value of sea urchin roe for human consumption.  相似文献

This study provides the first quantitative analysis of the littoral and supralittoral insect and mite assemblages of sub-Antarctic Marion Island. Seventeen mite species (126,203 individuals) from 11 families were found on the shore at Macaroni Bay. Three families dominated the assemblages in both abundance and diversity: the Hyadesiidae, Ameronothridae and Halacaridae. Six insect species from three orders were found on the shore. Species richness increased from one in the littoral, to four and two species in the Mastodia and Caloplaca zones, respectively. The littoral chironomid midge Telmatogeton amphibius was the most abundant insect species, constituting 80% of all insects counted. Arthropod assemblages corresponded closely to the cryptogam-dominated zonation patterns previously identified for the Marion Island shore. This clear association between arthropod and cryptogam zonation patterns provided a clear indication of habitat specificity in many of the species, and a quantitative analysis of habitat specificity on a species by species basis supported this idea. The specificity of most species to the shore, which forms part of the epilithic biotope, is most likely a consequence of the considerable age of this biotope compared to the younger, post-glacial vascular vegetation. Tourist species, i.e. species transient to an assemblage, inflated species richness in zones and the distribution ranges of species across zones. It is suggested that previous qualitative analyses of shoreline arthropod communities may have overestimated species ranges and richnesses because of the inclusion of tourist species. It is suggested that if a sound understanding of patterns in, and processes underlying Antarctic arthropod assemblages is to be achieved, quantitative analyses must be expanded in the region.
