To study the specificity of gastric lipases on carotenoid mono- and diesters, an enzymatic assay was applied. Digestions were carried out in phosphate buffer at pH 7.4 and 37 °C. As substrates we employed oleoresins from marigold (Tagetes erecta L.; lutein diesters), red paprika (Capsicum annuum L., mainly capsanthin diesters), papaya (Carica papaya L.; β-cryptoxanthin esters), and loquat (Eriobotrya japonica Lindl.; β-cryptoxanthin esters) as well as retinyl palmitate. These were reacted with porcine pancreatic lipase, porcine pancreatin, porcine cholesterol esterase, and human pancreatic lipase. As reference enzyme a yeast lipase from Candida rugosa was applied. A high turnover could be observed with porcine pancreatic lipase and porcine cholesterol esterase, indicating cholesterol esterase to be a plausible candidate for generation of free carotenoids in the gut. Human pancreatic lipase accepted only retinyl palmitate as substrate, carotenoid mono- and diesters were not hydrolyzed. The assay permits an approach for calculation of enzymatic activities towards carotenoid esters as substrates for the first time, which is based on the amount of enzyme formulation, present in the assay (U/mg solid). Furthermore, these studies provide deeper insight into carotenoid ester bioaccessibility.  相似文献

The amino acid residue tryptophan 27 of 6,7-dimethyl-8-ribityllumazine synthase of the yeast Schizosaccharomyces pombe was replaced by tyrosine. The structures of the W27Y mutant protein in complex with riboflavin, the substrate analogue 5-nitroso-6-ribitylamino-2,4(1H,3H)-pyrimidinedione, and the product analogue 6-carboxyethyl-7-oxo-8-ribityllumazine, were determined by X-ray crystallography at resolutions of 2.7-2.8 A. Whereas the indole system of W27 forms a coplanar pi-complex with riboflavin, the corresponding phenyl ring in the W27Y mutant establishes only peripheral contact with the heterocyclic ring system of the bound riboflavin. These findings provide an explanation for the absence of the long wavelength shift in optical absorption spectra of riboflavin bound to the mutant enzyme. The structures of the mutants are important tools for the interpretation of the unusual physical properties of riboflavin in complex with lumazine synthase.  相似文献

Desferrioxamine B is widely used as therapeutic agent for removal of excess body iron and, more recently, for removal of aluminium. A HPLC-based method for direct sensitive and reliable determination of ferrioxamine, desferrioxamine, aluminoxamine and related metabolites has been developed for use in pharmacokinetic studies. The method consists of complete separation of the analytes by an optimized mobile phase avoiding conversion of desferrioxamine to ferrioxamine by the analytical system and overcoming problems due to peak tailing properties of desferrioxamine. A post-column derivatization reaction with colourless fluoro-complexed iron converts all iron free species to ferrioxamine and allows quantification at 430 nm avoiding interference of UV-absorbing coelutes. This reaction might be of interest for other analytical procedures concerning iron chelators. The influence of the post-column reaction system on the column plate number is characterized. As the reaction rate of desferrioxamine and aluminoxamine with iron(III) is of second-order kinetics, a quadratic calibration function is observed, resulting from a compromise between residence time and peak broadening. A solid-phase extraction procedure is presented for extraction of the analytes from plasma. Limits of detection (S/N ratio of 3) were determined as 1.95 ng for ferrioxamine, 3.9 ng for aluminoxamine and 15.7 ng for desferrioxamine, expressed as on-column load. A new iron-free metabolite was identified with fast atom bombardment-mass spectrometry as N-hydroxylated desferrioxamine.  相似文献

Reversibly blindfolded crayfish (Procambarus clarkii) react to small swimming fish (Astyanax fasciatus mexicanus) approaching or passing nearby with antennal and cheliped movements and body turns (Fig. 3). We studied the accuracy and dynamics of crayfish orientation responses to the previously analyzed hydrodynamic disturbances caused by the fish, mostly produced by tail flicks.Antennal and cheliped movements started slightly before the onset of turning responses (Fig. 4). Antennal sweeps were performed most rapidly. 50% of the appendage sweeps resulted in contacts with the fish (Fig. 5).Most turns were directed toward the stimulus (Fig. 6). Response amplitudes increased with increasing stimulus angle. Turns were accurate for small stimulus angles, but smaller than expected for larger ones. Sweeps of ipsilateral antennae and chelipeds were generally directed backwards, while those of contralateral appendages were smaller and directed forwards. The amplitudes of appendage sweeps first increased with increasing stimulus angle and then decreased again for more caudal stimulus directions. Lateral stimuli (60°–120°) from opposite sides were usually significantly distinguished. The amplitudes of the different elements of orientation behaviour were highly correlated with each other, indicating that they were directed by the same sensory input.  相似文献

Summary In the present study the time course and spectral-amplitude distribution of hydrodynamic flow fields caused by moving fish, frogs, and crustaceans were investigated with the aid of laser-Doppler-anemometry. In the vicinity of a hovering fish sinusoidal water movements can be recorded whose velocity spectra peak below 10 Hz (Fig. 2). Single strokes during startle responses or during steady swimming of fish, frogs, and crustaceans cause short-lasting, low-frequency (<10 Hz), transient water movements (Fig. 3). Low-frequency transients also occur if a frog approaches and passes a velocity-sensitive hydrodynamic sensor. In contrast, transient water movements caused by a rapidly struggling or startled fish or water motions measured in the wake of a slowly swimming (47 cm/s) trout can be broadbanded, i.e., these water movements can contain frequency components up to at least 100 Hz (Figs. 4, 5A, 6). High-frequency hydrodynamic events can also be measured behind obstacles submerged in running water (Fig. 5C). The possible biological advantage of the ability to detect high-frequency hydroynamic events is discussed with respect to the natural occurrence of high frequencies and its potential role in orientation and predator-prey interactions of aquatic animals.Abbreviations LDA laser Doppler anemometer  相似文献

Inducible Co-stimulator (ICOS) plays a critical role in mediating T cell differentiation and function and is considered a key player in balancing T effector and T regulatory (Treg) cell responses. Here we show that activation of the ICOS signalling pathway during acute influenza A virus (IAV) infection by application of an agonistic ICOS antibody reduced the frequency of CD8+ T cells in the respiratory tract of IAV infected animals and delayed pathogen elimination. In line with this, immune-mediated influenza pneumonia was significantly ameliorated in mice that received ICOS agonist as indicated by significantly reduced alveolar infiltrations and bronchointerstitial pneumonia, while at the same time virus-related pathology remained unaffected. Importantly, ICOS agonist treatment resulted in expansion of CD4+Foxp3+ Tregs in IAV infected mice, which was associated with elevated levels of the immunosuppressive cytokine IL-10 in the alveolar space. Together, our findings suggest a prominent role of ICOS signaling during acute IAV infection by increasing the Treg/CD8+ T cell ratio with beneficial outcome on immune-mediated pneumonia and underline the suitability of ICOS as potential therapeutic target for immune intervention in those infectious conditions characterized by strong immunopathology rather than virus-mediated cytopathic effects.  相似文献

Public lands in the Rocky Mountain West are home to an abundance of vertebrate paleontological resources. These fossils typically are found in badlands terrain and at some distance from convenient transportation. These and other factors often make conventional surveying and mapping techniques time-consuming and problematic. Even obtaining quality images, at useful scales with limited distortions, can be difficult. In order to preserve the value of these unique paleontological resources, an integrated approach to close-range photogrammetry and high-accuracy ground-control surveying was developed in northern Wyoming for the documentation of tracks at the Red Gulch Dinosaur Tracksite. At this site, several methods for taking high-resolution, low-distortion photographs of localities were investigated. These methods included using tripods of various heights, remote-controlled airplanes, and an Aerial Camera Blimp System. In addition, the use of a variety of ground-control collection methods, including high accuracy GPS and Light Detection and Ranging, have also been investigated. These various field data collection methods were successfully integrated using soft copy photogrammetry to produce digital terrain models, which can represent the surface to a precision of 1 cm or less. The three-dimensional data were brought into GIS software where they are displayed, combined with photographs, and rotated for viewing from different perspectives. As a result of the success of these technologies in Wyoming, studies of other dinosaur tracksites in Colorado, Utah, and Wyoming (as well as bonebeds) have utilized these methods. The information gained from these sites is aiding in our understanding of community dynamics and preservational history of dinosaur populations in the Rocky Mountain West.  相似文献

Consistent inter-individual differences in daily activity rhythms (i.e., chronotypes) can have ecological consequences in determining access to food resources and avoidance of predators. The most common measure to characterize chronotypes in animals as well as humans is the onset of activity (i.e., early or late chronotypes). However, daily activity rhythms may also differ in the relative amount of activity displayed at particular time periods. Moreover, chronotypes may also be linked to other consistent inter-individual differences in behavior (i.e., personality), such as the propensity to take risks. Here, we used the signal crayfish Pacifastacus leniusculus to test the presence of chronotypes and risk-taking personality traits and a potential behavioral syndrome between these traits. We first exposed crayfish to 5 days of light–darkness to measure daily activity rhythms and then we applied a visual predator-simulating stimulus in 2 different contexts (neutral and food). Our results showed consistent (i.e., across 5 days) inter-individual differences in the relative nocturnal activity displayed in the early and middle, but not in the late part of darkness hours. Moreover, while crayfish displayed inter-individual differences in risk-taking behavior, these were not found to be consistent across 2 contexts. Therefore, we were not able to formally test a behavioral syndrome between these 2 traits. In conclusion, our study provides the first evidence of chronotypes in the relative amount of activity displayed at particular time periods. This could be a valuable information for applied ecological aspects related to the signal crayfish, which is a major invasive species of freshwater ecosystems.  相似文献
