Mexico is the main producer, consumer and exporter of avocado in the world, being Michoacan the main producer state contributing more than 80% of the national production. There are phytopathogens that decimate the production causing the death of the tree. Root samples were collected in avocado trees that showed the characteristic symptomatology of the disease known as avocado sadness, the sampling was carried out in four of the main avocado producing towns, in the state of Michoacan, Mexico. The isolation consisted in sowing root tissue in Petri dishes with V8®-PARPH culture medium, subsequently they were identified morphologically and for species level it was determined by molecular biology, with the PCR-ITS technique. Pathogenicity tests were performed in triplicate with avocado seedlings with more than six leaves. After 24 hours, the inoculated plants expressed decay in the apical part, after 120 hours the leaves showed yellowing and after 15 days there was a generalized wilt on the stem and leaves, re-isolating the phytopathogen Phytopythium vexans. This study confirms the first report of the oomycete P. vexans affecting avocado trees in the most important producing region of the Mexican Republic.  相似文献

Corresponding with the increasing global resource demand, harvesting now affects millions of hectares of boreal forest each year, and yet our understanding of harvesting impacts on boreal carbon (C) dynamics relative to wildfire remains unclear. We provide a direct comparison of C stocks following clearcut harvesting and fire over a 27-year chronosequence in the boreal forest of central Canada. Whereas many past studies have lacked measurement of all major C pools, we attempt to provide complete C pool coverage, including live biomass, deadwood, forest floor, and mineral soil C pools. The relative contribution of each C pool to total ecosystem C varied considerably between disturbance types. Live biomass C was significantly higher following harvesting compared with fire because of residual live trees and advanced regeneration. Conversely, most live biomass was killed following fire, and thus post-fire stands contained higher stocks of deadwood C. Snag and stump C mass peaked immediately following fire, but dramatically decreased 8 years after fire as dead trees began to fall over, contributing to the downed woody debris C pool. Forest floor C mass was substantially lower shortly after fire than harvesting, but this pool converged 8 years after fire and harvesting. When total ecosystem C stocks were examined, we found no significant difference during early stand development between harvesting and fire. Maximum total ecosystem C occurred at age 27 years, 185.1 ± 18.2 and 163.6 ± 8.0 Mg C ha?1 for harvesting and fire, respectively. Our results indicate strong differences in individual C pools, but similar total ecosystem C after fire and clearcutting in boreal forests, and shall help improve modeling terrestrial C flux after stand-replacing disturbances.  相似文献

Proteolysis of extracellular matrix components and the production of cryptic bioactive factors play key roles in vascular remodeling. We showed previously that extracellular matrix proteolysis is triggered by the apoptosis of endothelial cells (EC), resulting in the release of an anti-apoptotic C-terminal fragment of endorepellin (LG3). Here, we characterize the endorepellin-cleaving proteases released by apoptotic EC using a multifaceted proteomics strategy. Cathepsin L (CathL), a cysteine protease known to be associated with cardiovascular disease progression in animal models and humans, was isolated from medium conditioned by apoptotic EC. CathL cleaved recombinant endorepellin in vitro, leading to LG3 release. Inhibition of CathL activity in EC exposed to pro-apoptotic stimuli prevented LG3 release without modulating the development of apoptosis in EC. Inhibition of caspase-3 activation in EC with the biochemical inhibitor DEVD-fluoromethyl ketone or small interfering RNAs concomitantly prevented CathL release by EC, LG3 production, and the development of paracrine anti-apoptotic activity. These data demonstrate that caspase-3 activation is a novel pathway of importance for triggering extracellular CathL release and the cleavage of extracellular matrix components.  相似文献

Background  

Milk contains too little arginine for normal growth, but its precursors proline and glutamine are abundant; the small intestine of rodents and piglets produces arginine from proline during the suckling period; and parenterally fed premature human neonates frequently suffer from hypoargininemia. These findings raise the question whether the neonatal human small intestine also expresses the enzymes that enable the synthesis of arginine from proline and/or glutamine. Carbamoylphosphate synthetase (CPS), ornithine aminotransferase (OAT), argininosuccinate synthetase (ASS), arginase-1 (ARG1), arginase-2 (ARG2), and nitric-oxide synthase (NOS) were visualized by semiquantitative immunohistochemistry in 89 small-intestinal specimens.  相似文献

Coarse woody debris (CWD) volume and diversity are vital attributes of forest ecosystems. However, despite their importance, their long-term dynamics associated with fire- or logging-origin and overstory type have not been examined in boreal forest. We hypothesize that (1) CWD compositional diversity increases with stand development whereas CWD volume follows a U-shaped pattern. Furthermore, we attempted to test if (2) CWD volume and compositional diversity converge for postlogged and postfire stands through stand development, and (3) mixedwoods have more CWD volume and greater compositional diversity than conifer or broadleaf overstory types. We sampled 72 stands ranging in age from 7 to 201 years in fire-origin stands and 7–31 years in managed stands with conifer, mixedwood, and broadleaf overstory types in central boreal Canada. For fire-origin stands, snag volume was 100–260 m3/ha in 7-year-old stands, 5–20 m3/ha in 25-year-old stands, and 25–60 m3/ha in older stands; downed woody debris (DWD) volume decreased from 7 to 72–90 year-old stands, increased in 124- to 139-year-old stands, then either decreased or increased in 201-year-old stands depending on overstory type. CWD diversity increased from 25 to 124–139 year-old and plateaued, but in 7-year-old stands, CWD diversity was as high as that in the 124 and up year-old age classes. Logging resulted in a smaller amount and lower size variability of CWD in 7-year-old stands, with a larger portion being fast-decomposing Betula papyrifera. Most CWD characteristics had not converged by approximately 30 years since disturbance between the two stand origins. More diverse CWD occurred in mixedwoods, but conifer stands contained the greatest CWD volume except in 7 year-old postfire stands. The effect of solR inactivation on the metabolism of Clostridium acetobutylicum was examined using fermentation characterization and metabolic flux analysis. The solR-inactivated strain (SolRH) of this study had a higher rate of glucose utilization and produced higher solvent concentrations (by 25%, 14%, and 81%, respectively, for butanol, acetone, and ethanol) compared to the wild type. Strain SolRH(pTAAD), carrying a plasmid-encoded copy of the bifunctional alcohol/aldehyde dehydrogenase gene (aad) used in butanol production, produced even higher concentrations of solvents (by 21%, 45%, and 62%, respectively, for butanol, acetone, and ethanol) than strain SolRH. Clarithromycin used for strain SolRH maintenance during SolRH(pTAAD) fermentations did not alter product formation; however, tetracycline used for pTAAD maintenance resulted in 90% lower solvent production. BackgroundPreviously, we demonstrated that OVA-loaded macrophages (OVA-Mφ) partially suppress OVA-induced airway manifestations of asthma in BALB/c mice. In vitro studies showed that OVA-Mφ start to produce IL-10 upon interaction with allergen-specific T cells, which might mediate their immunosuppressive effects. Herein, we examined whether IL-10 is essential for the immunosuppressive effects of OVA-Mφ in vivo, and whether ex vivo stimulation of the IL-10 production by OVA-Mφ could enhance these effects.MethodsPeritoneal Mφ were loaded with OVA and stimulated with LPS or immunostimulatory sequence oligodeoxynucleotide (ISS-ODN) in vitro. The increase of IL-10 production was examined and, subsequently, ex vivo stimulated OVA-Mφ were used to treat (i.v.) OVA-sensitized mice. To further explore whether Mφ-derived IL-10 mediates the immunosuppressive effects, Mφ isolated from IL-10-/- mice were used for treatment.ResultsWe found that stimulation with LPS or ISS-ODN highly increased the IL-10 production by OVA-Mφ (2.5-fold and 4.5-fold increase, respectively). ISS-ODN stimulation of OVA-Mφ significantly potentiated the suppressive effects on allergic airway inflammation. Compared to sham-treatment, ISS-ODN-stimulated OVA-Mφ suppressed the airway eosinophilia by 85% (vs. 30% by unstimulated OVA-Mφ), IL-5 levels in bronchoalveolar lavage fluid by 80% (vs. 50%) and serum OVA-specific IgE levels by 60% (vs. 30%). Importantly, IL-10-/-Mφ that were loaded with OVA and stimulated with ISS-ODN ex vivo, failed to suppress OVA-induced airway inflammation.ConclusionsThese results demonstrate that Mφ-derived IL-10 mediates anti-inflammatory responses in a mouse model of allergic asthma, which both can be potentiated by stimulation with ISS-ODN.  相似文献

Background

Lymphocyte function-associated antigen-1 (LFA-1, CD11a/CD18, alpha L beta 2) is required for many cellular adhesive interactions during the immune response.

Results

The Capra hircus CD11a-encoding cDNA was sequenced and compared with its human, murine, rat, bovine and ovine counterparts. Despite some focal differences, it shares all the main characteristics of its known mammalian homologues.

Conclusion

Therefore, along with the caprine CD18-encoding cDNA, which has been available for a few months, the sequence data revealed here will allow the Capra hircus LFA-1 expression in vitro as a tool to explore the specificities of inflammation in the caprine species.

Factors were defined for elongation, rooting and acclimatization of micropropagated shoots ofLarix x eurolepis Henry initiated from short shoot buds of plagiotropic stecklings serially propagated for 9 years from an 8-year-old tree. Initiation and multiplication were on Schenk and Hildebrandt (SH) medium supplemented with 5 M 6-benzyladenine (BA) and 1 M indole-butyric acid (IBA). Stem elongation was obtained in 36% of the shoots on SH medium containing 0.5 M BA and 63% of the remaining non-elongated shoots initiated stem elongation after transfer on SH medium devoid of growth regulators. Rooting involved 2 steps: root induction on Campbell and Durzan mineral salts and Murashige and Skoog organic elements, both half-strength (CD-MS/2), supplemented with 1 M of both naphthaleneacetic acid (NAA) and IBA, and root elongation following transfer to CD-MS/2 medium devoid of growth regulators. Repeating this 2-step sequence yielded up to 67% rooted shoots. Acclimatization of plantlets ranged from 83% to 100%. Over 300 plants were transferred to the greenhouse; some showed plagiotropic growth.  相似文献
