Arrestins are proteins that arrest the activity of G protein-coupled receptors (GPCRs). While it is well established that normal inactivation of photoexcited rhodopsin, the GPCR of rod phototransduction, requires arrestin (Arr1), it has been controversial whether the same requirement holds for cone opsin inactivation. Mouse cone photoreceptors express two distinct visual arrestins: Arr1 and Arr4. By means of recordings from cones of mice with one or both arrestins knocked out, this investigation establishes that a visual arrestin is required for normal cone inactivation. Arrestin-independent inactivation is 70-fold more rapid in cones than in rods, however. Dual arrestin expression in cones could be a holdover from ancient genome duplication events that led to multiple isoforms of arrestin, allowing evolutionary specialization of one form while the other maintains the basic function. 相似文献

The liver is a target for toxic chemicals such as cadmium (Cd). When the liver is damaged, hepatic stellate cells (HSC) are activated and transformed into myofibroblast-like cells, which are responsible for liver fibrosis. Curcuma longa has been reported to exert a hepato-protective effect under various pathological conditions. We investigated the effects of C. longa administration on HSC activation in response to Cd induced hepatotoxicity. Forty adult male albino rats were divided into: group 1 (control), group 2 (Cd treated), group 3 (C. longa treated) and group 4 (Cd and C. longa treated). After 6 weeks, liver specimens were prepared for light and electron microscopy examination of histological changes and immunohistochemical localization of alpha smooth muscle actin (αSMA) as a specific marker for activated HSC. Activated HSC with a positive αSMA immune reaction were not detected in groups 1 and 3. Large numbers of activated HSC with αSMA immune reactions were observed in group 2 in addition to Cd induced hepatotoxic changes including excess collagen deposition in thickened portal triads, interlobular septa with hepatic lobulation, inflammatory cell infiltration, a significant increase in Kupffer cells and degenerated hepatocytes. In group 4, we observed a significant decrease in HSC that expressed αSMA with amelioration of the hepatotoxic changes. C. longa administration decreased HSC activation and ameliorated hepatotoxic changes caused by Cd in adult rats. 相似文献

We investigated the origins of greater clot rigidity associated with FXIIIa-dependent cross-linking. Fibrin clots were examined in which cross-linking was controlled through the use of two inhibitors: a highly specific active-center-directed synthetic inhibitor of FXIIIa, 1,3-dimethyl-4,5-diphenyl-2[2(oxopropyl)thio]imidazolium trifluoromethylsulfonate, and a patient-derived immunoglobulin directed mainly against the thrombin-activated catalytic A subunits of thrombin-activated FXIII. Cross-linked fibrin chains were identified and quantified by one- and two-dimensional gel electrophoresis and immunostaining with antibodies specific for the alpha- and gamma-chains of fibrin. Gamma-dimers, gamma-multimers, alpha(n)-polymers, and alpha(p)gamma(q)-hybrids were detected. The synthetic inhibitor was highly effective in preventing the production of all cross-linked species. In contrast, the autoimmune antibody of the patient caused primarily an inhibition of alpha-chain cross-linking. Clot rigidities (storage moduli, G') were measured with a cone and plate rheometer and correlated with the distributions of the various cross-linked species found in the clots. Our findings indicate that the FXIIIa-induced dimeric cross-linking of gamma-chains by itself is not sufficient to stiffen the fibrin networks. Instead, the augmentation of clot rigidity was more strongly correlated with the formation of gamma-multimers, alpha(n)-polymers, and alpha(p)gamma(q)-hybrid cross-links. Sex ratio theory attempts to explain observed variation in offspring sex ratio at both the population and the brood levels. In the context of low‐fecundity organisms producing high‐investment offspring, the drivers of adaptive variation in sex ratio are incompletely understood. For raptors that display reverse sexual dimorphism (RSD), preferential allocation of resources to the putatively cheaper sex (male) may be a response to environmental, social or demographic stressors. To assess the extent of skew in offspring sex ratios and to evaluate possible dietary, environmental and demographic correlates of such skew to long‐lived RSD avian species, we evaluated the offspring sex ratio of 219 chicks from 119 broods in 30 territories of Eastern Imperial Eagles Aquila heliaca across 7 years and four regions at a nature reserve in Kazakhstan. Only in one region in 1 year of our study did the offspring sex ratio differ significantly from parity (10 males : 1 female in 11 territories). Whereas offspring sex ratios were independent of dietary diversity, precipitation, temperature and productivity, we found that year had a moderate effect on brood sex ratio within territories. Our results provide limited evidence of brood sex manipulation in these populations of Eastern Imperial Eagles, and no mechanistic insight into predictions associated with it. Stochastic variation is likely to explain much of the fluctuation we observed in sex ratios, but our observations are also consistent with the hypothesis that sex‐ratio manipulation may occur irregularly, in concurrence with atypical environmental or demographic conditions that fluctuate at a time scale longer than that of our 7‐year study. 相似文献

The plant alkaloids vinblastine and colchicine are known to arrest cells in mitosis by virtue of their binding to spindle protein. These drugs are also capable of binding to microtubule protein and causing these structures to disaggregate into nonfunctional subunits (1, 2). Microtubular structures are thought to be involved in the secretory process of a number of proteins including insulin (7), collagen (4), and thyroid hormone (12). This study compared evoked potentials (EP) recorded from the neocortical vibrissae representation region of adult rats (the barrelfield) with those occurring in neocortical grafts 4–5 months after transplant. Epidural recording showed that displacing the vibrassae led to shaping of EP in the intact barrelfield beginning with a positive followed by a negative component. The EP recorded from the surface of the graft occurred with the same latency but consisted of a negative component only. In conclusion, coordinated "population" discharges of grafted neurons do generate local field poentials, but owing to the diffuse neuronal organization of the grafts, these potentials are difficult to distinguish from EP resulting from passive current from adjacent neocortical areas and subcortical structures of the host brain.Institute of Biological Physics, Academy of Sciences of the USSR, Pushchino, Moscow Region.
