An ultrastructural and cytochemical study of normal human mammary epithelial cells cultured from post-weaning breast fluids is described. Cells were examined at the time of plating and at intervals up to 28 days in culture. Three different stages in the morphological differentiation of these cells in vitro were observed: (1) the first stage was the formation of a monolayer of single cells, which occurred between days 1 and 10 in culture. The cells in this stage were not interconnected by junctional complexes and lacked Mg++- dependent ATPase activity in the plasma membranes, but did contain a large quantity of lipid and exhibited some secretory characteristics. (2) The second stage, occurring at 10 to 16 days in culture, was characterized by the formation of junctional complexes, the appearance of Mg++-dependent ATPase in the plasma membrane and a decrease in the number of dense bodies with peroxidase activity. (3) The third stage, occurring at 16 to 28 days in culture, was characterized by the formation of stratified layers of epithelial cells, which were interconnected by a larger number of desmosomes with numerous pleomorphic microfilaments. The Mg++-dependent ATPase activity in the plasma membrane was retained and the dense bodies with peroxidase activity were rarely observed at this stage. During the last seven days were prominent in the cells of the stratified layer. After 28 days in the culture, the cells began to round up and slough off the culture plate. 相似文献

Even though significant breakthroughs with over 18% power conversion efficiencies (PCEs) in polymer:non‐fullerene acceptor (NFA) bulk heterojunction organic solar cells (OSCs) have been achieved, not many studies have focused on acquiring a comprehensive understanding of the underlying mechanisms governing these systems. This is because it can be challenging to delineate device photophysics in polymer:NFA blends comprehensively, and even more complicated to trace the origins of the differences in device photophysics to the subtle differences in energetics and morphology. Here, a systematic study of a series of polymer:NFA blends is conducted to unify and correlate the cumulative effects of i) voltage losses, ii) charge generation efficiencies, iii) non‐geminate recombination and extraction dynamics, and iv) nuanced morphological differences with device performances. Most importantly, a deconvolution of the major loss processes in polymer:NFA blends and their connections to the complex BHJ morphology and energetics are established. An extension to advanced morphological techniques, such as solid‐state NMR (for atomic level insights on the local ordering and donor:acceptor π? π interactions) and resonant soft X‐ray scattering (for donor and acceptor interfacial area and domain spacings), provide detailed insights on how efficient charge generation, transport, and extraction processes can outweigh increased voltage losses to yield high PCEs. 相似文献
Temperatures in mountain areas are increasing at a higher rate than the Northern Hemisphere land average, but how fauna may respond, in particular in terms of phenology, remains poorly understood. The aim of this study was to assess how elevation could modify the relationships between climate variability (air temperature and snow melt‐out date), the timing of plant phenology and egg‐laying date of the coal tit (Periparus ater). We collected 9 years (2011–2019) of data on egg‐laying date, spring air temperature, snow melt‐out date, and larch budburst date at two elevations (~1,300 m and ~1,900 m asl) on a slope located in the Mont‐Blanc Massif in the French Alps. We found that at low elevation, larch budburst date had a direct influence on egg‐laying date, while at high‐altitude snow melt‐out date was the limiting factor. At both elevations, air temperature had a similar effect on egg‐laying date, but was a poorer predictor than larch budburst or snowmelt date. Our results shed light on proximate drivers of breeding phenology responses to interannual climate variability in mountain areas and suggest that factors directly influencing species phenology vary at different elevations. Predicting the future responses of species in a climate change context will require testing the transferability of models and accounting for nonstationary relationships between environmental predictors and the timing of phenological events. 相似文献
Cat's claw creeper, Dolichandra unguis-cati (Bignoniaceae), a perennial woody vine native to tropical America, is a target for biological control in Australia and South Africa. The cat's claw creeper leaf-tying moth Hypocosmia pyrochroma (Lepidoptera: Pyralidae) from tropical South America was released as a biological control agent for cat's claw creeper in Australia from 2007 to 2010. A total of 2,277 adults, 837 pupae and 77,250 larvae were released at 40 sites in Queensland and New South Wales. Releases were made mostly in open fields (85%), and at limited sites (15%) in insect-proof cages erected over naturally occurring cat's claw creeper infestations in the field. Sampling was conducted annually in spring and autumn to monitor the establishment and dispersal of H. pyrochroma. Establishment of H. pyrochroma was first noticed in 2012 at three release sites and since then the number of established sites has increased to 80 in 2020. Establishment was evident on both ‘short-pod’ and ‘long-pod’ forms of cat's claw creeper and was more widespread in sites where releases were made within insect-proof field cages (50%) than in sites with open field releases (9%). The moth was active from late spring to late autumn with peak larval activity in late summer. To date, all field establishments have been in areas predicted by a CLIMEX model as climatically suitable but restricted mostly to riparian environment (93% of establishment), where the moth has continued to spread from 1.5 to 23 km from release sites. In contrast, there is the only limited establishment and spread in non-riparian corridors, highlighting the role of microclimate (riparian) as a limiting factor for establishment and spread. Future efforts will focus on redistribution of the agent to river/creek systems where the moth is currently not present. 相似文献
Information on plant community assembly mechanisms is limited on forest reclamation sites after mining in the Canadian boreal forest. We assessed the change in plant community composition after Year 2 and Year 5 on species-rich forest floor mineral mix (FFMM) and species-poor peat mineral mix (PMM) reclamation soils by examining assembly mechanisms, i.e., seed bank, seed rain, biotic dispersal, vegetative expansion, and competition. Initial plant cover and diversity were greater on FFMM due to non-native species originating from the seed bank, which had 5× more seeds in the FFMM. By Year 5, both soil types had approximately 40% cover and 80 species richness due to the addition of wind and biotic-dispersed species and were characterized by a shift towards native species. Native forbs using vegetative reproduction expanded up to 2 m from FFMM into PMM. At Year 5 competition does not seem to have a large role in the structuring of the vegetation community. Overall, multiple factors were involved in structuring plant communities on reclamation sites, but we observed a general convergence between plant communities on different soil types in a relatively short period of time.
