Plague, caused by the bacterium Yersinia pestis, is a mammalian vector-borne disease, transmitted by fleas that serve as the vector between rodent hosts. For many pathogens, including Y. pestis, there are strong evolutionary pressures that lead to a reduction in ‘useless genes'', with only those retained that reflect function in the specific environment inhabited by the pathogen. Genetic traits critical for survival and transmission between two environments, the rodent and the flea, are conserved in epizootic/epidemic plague strains. However, there are genes that remain conserved for which no function in the flea–rodent cycle has yet been observed, indicating an additional environment may exist in the transmission cycle of plague. Here, we present evidence for highly conserved genes that suggests a role in the persistence of Y. pestis after death of its host. Furthermore, maintenance of these genes points to Y. pestis traversing a post-mortem path between, and possibly within, epizootic periods and offering insight into mechanisms that may allow Y. pestis an alternative route of transmission in the natural environment. 相似文献

Between November 2010, and May 2011, eleven cases of cholera, unrelated to a concurrent outbreak on the island of Hispaniola, were recorded, and the causative agent, Vibrio cholerae serogroup O75, was traced to oysters harvested from Apalachicola Bay, Florida. From the 11 diagnosed cases, eight isolates of V. cholerae were isolated and their genomes were sequenced. Genomic analysis demonstrated the presence of a suite of mobile elements previously shown to be involved in the disease process of cholera (ctxAB, VPI-1 and -2, and a VSP-II like variant) and a phylogenomic analysis showed the isolates to be sister taxa to toxigenic V. cholerae V51 serogroup O141, a clinical strain isolated 23 years earlier. Toxigenic V. cholerae O75 has been repeatedly isolated from clinical cases in the southeastern United States and toxigenic V. cholerae O141 isolates have been isolated globally from clinical cases over several decades. Comparative genomics, phenotypic analyses, and a Caenorhabditis elegans model of infection for the isolates were conducted. This analysis coupled with isolation data of V. cholerae O75 and O141 suggests these strains may represent an underappreciated clade of cholera-causing strains responsible for significant disease burden globally. 相似文献

This paper explores the potential of a new method of reconstructing historical vegetation change in the Australian rangelands. Historical monitoring of rangeland vegetation has been so deficient that it is not possible to determine whether a long‐term trend toward degradation has occurred (as is often assumed) or, indeed, if it is continuing to occur. Because long‐term records are unavailable any attempt to monitor vegetation retrospectively must be based on proxy measures rather than direct observation. Where historical data are lacking an integration of palaeoecological, archaeological and ecological methods is required to reconstruct the past. Our research is based on a detailed analysis of sheep faeces deposited near a shearing shed in the semiarid rangelands of south‐west Queensland between the late 1930s and the mid‐1990s. The faeces in these deposits represent the diet of sheep in the days leading up to the property’s annual shearing and as such are a potentially useful index to changes in vegetation. Results indicate significant changes in the diet of sheep since the late 1940s. The potential of this method, and its limitations, are discussed. Long‐term records are critical in understanding issues of sustainability in land management and it is intended that this paper will stimulate further research into historical vegetation change in rangelands. 相似文献
