Autonomous selfing can provide reproductive assurance (RA) for flowering plants that are unattractive to pollinators or in environments that are pollen limited. Pollen limitation may result from the breakdown of once-continuous habitat into smaller, more isolated patches (habitat fragmentation) if fragmentation negatively impacts pollinator populations. Here we quantify the levels of pollen limitation and RA among large and small populations of Collinsia parviflora, a wildflower with inter-population variation in flower size. We found that none of the populations were pollen limited, as pollen-supplemented and intact flowers did not differ in seed production. There was a significant effect of flower size on RA; intact flowers (can self) produced significantly more seeds than emasculated flowers (require pollen delivery) in small-flowered plants but not large-flowered plants. Population size nested within flower size did not significantly affect RA, but there was a large difference between our two replicate populations for large-flowered, small populations and small-flowered, large populations that appears related to a more variable pollination environment under these conditions. In fact, levels of RA were strongly negatively correlated with rates of pollinator visitation, whereby infrequent visitation by pollinators yielded high levels of RA via autonomous selfing, but there was no benefit of autonomous selfing when visitation rates were high. These results suggest that autonomous selfing may be adaptive in fragmented habitats or other ecological circumstances that affect pollinator visitation rates.  相似文献

Foliar fungal pathogens from the genus Mycosphaerella affect eucalyptus in natural forests and plantations worldwide. QTL analysis was conducted to dissect the genetic control of resistance in Eucalyptus globulus to a natural infection by Mycosphaerella leaf disease, using a clonally replicated outbred F2 family (112 genotypes) planted in a field trial. Two major QTL, with high LOD support (20.2 and 10.9) and high genomewide significance, explained a large proportion (52%) of the phenotypic variance in the severity of damage by Mycosphaerella cryptica, which may be indicative of oligogenic control. Both QTL were validated in a second F2 family and one was validated in a third F2 family. The mean values of different genotype classes at both major QTL argue for Mendelian inheritance with resistance dominant over susceptibility. There were strong correlations between the levels of Mycosphaerella damage in related genetic material planted in three widely separated locations in Tasmania. These findings together provide evidence that the genes controlling resistance to Mycosphaerella damage are stable in different genetic backgrounds and across different environments.  相似文献

Protein kinase A (PKA) or cAMP-dependent protein kinase (cAPK) mediates the synergistic effects of cAMP- and glucocorticoid (GC)-induced apoptosis in lymphoid cells. Using two human acute lymphoblastic leukemia cell (CEM) clones with respective GC-sensitive and GC-resistant phenotypes, we discovered that the PKA regulatory subunit isoform RII(beta) is preferentially expressed in the GC-sensitive clone C7-14 cells, whereas other intracellular cAMP receptors, including the exchange proteins directly activated by cAMP (Epac), are expressed at similar levels in both GC-sensitive and GC-resistant clones. High RII(beta) expression level in C7-14 cells is associated with elevated total PKA cellular activity and cAMP sensitivity, which consequently lead to an increased basal PKA activity. cAMP analogs that selectively activate type II PKA recapitulate the effects of forskolin of promoting apoptosis and antagonizing AKT/PKB activity in both GC-sensitive and GC-resistant clones, whereas type I PKA-selective agonists do not. Furthermore, down-regulation of RII(beta) leads to increased AKT/PKB activation and enhanced GC resistance in C7-14 cells. These results demonstrate that PKA RII(beta) is responsible for increased GC sensitivity, critical for cAMP-mediated synergistic cell killing in CEM cells, and may represent a novel therapeutic target for GC-resistant lymphoid malignancy.  相似文献

We report the first direct observations of the physical and chemical microstructure of spider dragline, revealed by analytical transmission electron microscopy. Individual crystallites were imaged within the amorphous matrix. They are irregularly shaped, approximately 70–100 nm in diameter, and uniformly distributed throughout the matrix. Electron diffraction determined their space group to be P21. The corresponding orthogonal cell has lattice parameters of a = 13.31 Å (β-sheet repeat), b = 9.44 Å (interchain repeat within β-sheets), and c = 20.88 Å (repeat along polypeptide chain). Electron energy loss spectroscopy indicated compositional variations within the matrix, and between the crystallites and matrix. Most notably, calcium was found exclusively in the crystallites. Attempts to produce synthetic analogues of dragline, which exhibits an unparalleled combination of strength, stiffness, and toughness, cannot depend solely on duplicating the constituent proteins. The complex hierarchical microstructure of the natural material must be taken into account. © 1994 John Wiley & Sons, Inc.  相似文献

Co‐occurring species are rarely considered as a factor influencing habitat selection. However, niche theory predicts that sharing resources, predators, and other interspecific interactions can limit the environmental conditions under which a species may exist. How does the spatial distribution of one species affect that of another within shared landscapes? We tested whether sympatric marten Martes americana and fishers M. pennanti in a mountain landscape in Alberta, Canada exhibit local‐scale spatial segregation, beyond differential habitat selection. We modelled marten and fisher distribution in relation to remotely‐sensed habitat data and species co‐occurrence, using generalized linear models and information‐theoretic model selection. Marten and fishers selected different habitat types and showed different responses to habitat fragmentation. Even after accounting for these differences, the absence of one species significantly explained the occurrence of the other. We conclude that the spatial distribution of marten and fishers influences habitat selection by each other at landscape scales, and hypothesize that this pattern may result from competition in a spatially heterogeneous environment. Species‐habitat models that consider only resources may fail to capture key predictors of species’ occurrence. Reliable prediction and inference requires that ecologists expand from landscapes to also include species‐scapes: a spatial plane of species interactions that combines with resources to drive species’ distributions.  相似文献

To determine if microbial species play an active role in the development of calcium carbonate (CaCO 3 ) deposits (speleothems) in cave environments, we isolated 51 culturable bacteria from a coralloid speleothem and tested their ability to dissolve and precipitate CaCO 3 . The majority of these isolates could precipitate CaCO 3 minerals; scanning electron microscopy and X-ray diffractrometry demonstrated that aragonite, calcite and vaterite were produced in this process. Due to the inability of dead cells to precipitate these minerals, this suggested that calcification requires metabolic activity. Given growth of these species on calcium acetate, but the toxicity of Ca 2+ ions to bacteria, we created a loss-of-function gene knock-out in the Ca 2+ ion efflux protein ChaA. The loss of this protein inhibited growth on media containing calcium, suggesting that the need to remove Ca 2+ ions from the cell may drive calcification. With no carbonate in the media used in the calcification studies, we used stable isotope probing with C 13 O 2 to determine whether atmospheric CO 2 could be the source of these ions. The resultant crystals were significantly enriched in this heavy isotope, suggesting that extracellular CO 2 does indeed contribute to the mineral structure. The physiological adaptation of removing toxic Ca 2+ ions by calcification, while useful in numerous environments, would be particularly beneficial to bacteria in Ca 2+ -rich cave environments. Such activity may also create the initial crystal nucleation sites that contribute to the formation of secondary CaCO 3 deposits within caves.  相似文献

Members of the pentraxin family, including PTX3 and serum amyloid P component (SAP), have been reported to play a role in innate host defence against a range of microbial pathogens, yet little is known regarding their antiviral activities. In this study, we demonstrate that human SAP binds to human influenza A virus (IAV) strains and mediates a range of antiviral activities, including inhibition of IAV-induced hemagglutination (HA), neutralization of virus infectivity and inhibition of the enzymatic activity of the viral neuraminidase (NA). Characterization of the anti-IAV activity of SAP after periodate or bacterial sialidase treatment demonstrated that α(2,6)-linked sialic acid residues on the glycosidic moiety of SAP are critical for recognition by the HA of susceptible IAV strains. Other proteins of the innate immune system, namely human surfactant protein A and porcine surfactant protein D, have been reported to express sialylated glycans which facilitate inhibition of particular IAV strains, yet the specific viral determinants for recognition of these inhibitors have not been defined. Herein, we have selected virus mutants in the presence of human SAP and identified specific residues in the receptor-binding pocket of the viral HA which are critical for recognition and therefore susceptibility to the antiviral activities of SAP. Given the widespread expression of α(2,6)-linked sialic acid in the human respiratory tract, we propose that SAP may act as an effective receptor mimic to limit IAV infection of airway epithelial cells.  相似文献

Demography impacts the observed standing level of genetic diversity present in populations. Distinguishing the relative impacts of demography from selection requires a baseline of expressed gene variation in naturally occurring populations. Six nuclear genes were sequenced to estimate the patterns and levels of genetic diversity in natural Arabidopsis lyrata subsp. petraea populations that differ in demographic histories since the Pleistocene. As expected, northern European populations have genetic signatures of a strong population bottleneck likely due to glaciation during the Pleistocene. Levels of diversity in the northern populations are about half of that in central European populations. Bayesian estimates of historical population size changes indicate that central European populations also have signatures of population size change since the last glacial maxima, suggesting that these populations are not as stable as previously thought. Time since divergence amongst northern European populations is higher than amongst central European populations, suggesting that the northern European populations were established before the Pleistocene and survived glaciation in small separated refugia. Estimates of demography based on expressed genes are complementary to estimates based on microsatellites and transposable elements, elucidating temporal shifts in population dynamics and confirming the importance of marker selection for tests of demography.  相似文献
