The skin provides an important first line of defence and immunological barrier to invasive pathogens, but immune responses must also be regulated to maintain barrier function and ensure tolerance of skin surface commensal organisms. In schistosomiasis-endemic regions, populations can experience repeated percutaneous exposure to schistosome larvae, however little is known about how repeated exposure to pathogens affects immune regulation in the skin. Here, using a murine model of repeated infection with Schistosoma mansoni larvae, we show that the skin infection site becomes rich in regulatory IL-10, whilst in its absence, inflammation, neutrophil recruitment, and local lymphocyte proliferation is increased. Whilst CD4+ T cells are the primary cellular source of regulatory IL-10, they expressed none of the markers conventionally associated with T regulatory (Treg) cells (i.e. FoxP3, Helios, Nrp1, CD223, or CD49b). Nevertheless, these IL-10+ CD4+ T cells in the skin from repeatedly infected mice are functionally suppressive as they reduced proliferation of responsive CD4+ T cells from the skin draining lymph node. Moreover, the skin of infected Rag-/- mice had impaired IL-10 production and increased neutrophil recruitment. Finally, we show that the mechanism behind IL-10 production by CD4+ T cells in the skin is due to a combination of an initial (day 1) response specific to skin commensal bacteria, and then over the following days schistosome-specific CD4+ T cell responses, which together contribute towards limiting inflammation and tissue damage following schistosome infection. We propose CD4+ T cells in the skin that do not express markers of conventional T regulatory cell populations have a significant role in immune regulation after repeated pathogen exposure and speculate that these cells may also help to maintain skin barrier function in the context of repeated percutaneous insult by other skin pathogens.  相似文献

Queens in some species of ant with several queens per colony are believed to inhibit one another's fecundity pheromonally. This paper describes a repeat of an experimental test of this idea in the multiple-queen ant Leptothorax acervorum. The three most fecund queens from a field-collected colony were housed together in an experimental nest. This was then filmed for 10 h per day over 36 consecutive days (i.e. for 360 h in total). Over the first 12 d, all queens were present in the colony. For the middle 12 d, the two most fecund queens were removed, but were returned for the final 12 d. The queen that was observed throughout the experiment failed to show a rise in fecundity when the other queens were removed. Instead, her egg-laying rate fell, although this drop was not significant. These results confirm previous findings that short-term, strong pheromonal inhibition of fecundity does not occur among L. acervorum queens. The phenomena of pheromonal inhibition in other species may be a result of workers treating queens differentially in response to rapid, short-term changes in the numbers of queens and eggs. If so, the results presented here could stem from the relative infrequency of such changes in L. acervorum.  相似文献

Background

Research findings investigating the sociodemographics of autism spectrum disorder (ASD) have been inconsistent and rarely considered the presence of intellectual disability (ID).

Methods

We used population data on Western Australian singletons born from 1984 to 1999 (n = 398,353) to examine the sociodemographic characteristics of children diagnosed with ASD with or without ID, or ID without ASD compared with non-affected children.

Results

The profiles for the four categories examined, mild-moderate ID, severe ID, ASD without ID and ASD with ID varied considerably and we often identified a gradient effect where the risk factors for mild-moderate ID and ASD without ID were at opposite extremes while those for ASD with ID were intermediary. This was demonstrated clearly with increased odds of ASD without ID amongst older mothers aged 35 years and over (odds ratio (OR) = 1.69 [CI: 1.18, 2.43]), first born infants (OR = 2.78; [CI: 1.67, 4.54]), male infants (OR = 6.57 [CI: 4.87, 8.87]) and increasing socioeconomic advantage. In contrast, mild-moderate ID was associated with younger mothers aged less than 20 years (OR = 1.88 [CI: 1.57, 2.25]), paternal age greater than 40 years (OR = 1.59 [CI: 1.36, 1.86]), Australian-born and Aboriginal mothers (OR = 1.60 [CI: 1.41, 1.82]), increasing birth order and increasing social disadvantage (OR = 2.56 [CI: 2.27, 2.97]). Mothers of infants residing in regional or remote areas had consistently lower risk of ASD or ID and may be linked to reduced access to services or under-ascertainment rather than a protective effect of location.

Conclusions

The different risk profiles observed between groups may be related to aetiological differences or ascertainment factors or both. Untangling these pathways is challenging but an urgent public health priority in view of the supposed autism epidemic.  相似文献

Reactive oxygen species (ROS) play key roles in mucosal defense, yet how they are induced and the consequences for pathogens are unclear. We report that ROS generated by epithelial NADPH oxidases (Nox1/Duox2) during Campylobacter jejuni infection impair bacterial capsule formation and virulence by altering bacterial signal transduction. Upon C. jejuni invasion, ROS released from the intestinal mucosa inhibit the bacterial phosphotyrosine network that is regulated by the outer-membrane tyrosine kinase Cjtk (Cj1170/OMP50). ROS-mediated Cjtk inactivation results in an overall decrease in the phosphorylation of C. jejuni outer-membrane/periplasmic proteins, including UDP-GlcNAc/Glc 4-epimerase (Gne), an enzyme required for N-glycosylation and capsule formation. Cjtk positively regulates Gne by phosphorylating an active site tyrosine, while loss of Cjtk or ROS treatment inhibits Gne activity, causing altered polysaccharide synthesis. Thus, epithelial NADPH oxidases are an early antibacterial defense system in the intestinal mucosa that modifies virulence by disrupting bacterial signaling.  相似文献

We present results from a detailed three-dimensional finite element analysis of the cranium and mandible of the Australian dingo (Canis lupus dingo) during a range of feeding activities and compare results with predictions based on two-dimensional methodology [Greaves, W.S., 2000. Location of the vector of jaw muscle force in mammals. Journal of Morphology 243, 293-299]. Greaves showed that the resultant muscle vector intersects the mandible line slightly posterior to the lower third molar (m3). Our work demonstrates that this is qualitatively correct, although the actual point is closer to the jaw joint. We show that it is theoretically possible for the biting side of the mandible to dislocate during unilateral biting; however, the bite point needs to be posterior to m3. Simulations show that reduced muscle activation on the non-biting side can considerably diminish the likelihood of dislocation with only a minor decrease in bite force during unilateral biting. By modulating muscle recruitment the animal may be able to maximise bite force whilst minimising the risk of dislocation.  相似文献

Social insects (ants, bees, wasps and termites) as a group are species rich and ecologically dominant. Many are outstanding "ecological engineers", or providers of "ecosystem services", or potential bioindicator species. Few social insects are currently formally classified as Threatened, but this is almost certainly due to a lack of information on population sizes and trends in scarce species. The main influence that sociality has on threats faced by social insects is in reducing effective population sizes, increasing population genetic subdivision and possibly reducing levels of genetic variation relative to solitary species. The main influence that sociality has on threats from social insects is via its role in the ecological success of invasive species, which frequently pose a major hazard to native biotas. In some cases, social features underpinning ecological success in the original range almost certainly contribute to the success of invasive social insects. However, recent studies show or strongly suggest that, in some of the most notoriously invasive populations of ants, bees and wasps, novel social traits have arisen that greatly enhance the rate of spread and ecological competitiveness of these populations. Sociality can therefore represent either a liability or an asset in its contribution to the persistence of social insect populations.  相似文献
