Endocrine disruption is commonly thought to be restricted to a direct endocrine mode of action i.e. the perturbation of the activation of a given type of hormonal receptor by its natural ligand. Consistent with the WHO definition of an endocrine disrupter, a key issue is the “altered function(s) of the endocrine system”. Such altered functions can result from different chemical interactions, beyond agonistic or antagonistic effect at a given receptor. Based on neuroendocrine disruption by polychlorinated biphenyls and bisphenol A, this paper proposes different mechanistic paradigms that can result in adverse health effects. They are a consequence of altered endocrine function(s) secondary to chemical interaction with different steps in the physiological regulatory processes, thus accounting for a possibly indirect endocrine mode of action. 相似文献

Comparative genomic data among organisms allow the reconstruction of their phylogenies and evolutionary time scales. Molecular timings have been recently used to suggest that environmental global change have shaped the evolutionary history of diverse terrestrial organisms. Living xenarthrans (armadillos, anteaters and sloths) constitute an ideal model for studying the influence of past environmental changes on species diversification. We have optimised an indirect immunoperoxidase technique demonstrating bromodeoxyuridine (BrdU) incorporation into dividing cells for cerebellar tissue sections of four-day-old rats injected with this marker. This permits confident identification of granule-cell precursors engaged in DNA synthesis in the external granular layer of the developing cerebellum. Preservation of BrdU immunoreactivity is attained using methanol/acetic acid fixation and different pretreatments before immunostaining, while unlabeled nuclei can be recognized clearly after Feulgen or hematoxylin counterstaining. We established conditions to ensure satisfactory BrdU uptake without affecting cell-cycle progression during the postlabeling time period. The dose of BrdU employed provides saturation S-phase labeling from at least 1 h after BrdU delivery. Various kinetic parameters and phase durations have been determined in experiments involving a single injection or cumulative labeling sequences, and the cycle time was calculated based on two models of generative behavior: steady-state and exponential growth. The working hypothesis of steadystate kinetics can be adopted successfully if the existence of neuroblasts with different proliferation rates is taken into account. 相似文献

To expand the functional analysis of plant mitochondria, we have undertaken the building of the proteome of pea mitochondria purified from leaves (green and etiolated), roots and seeds. In the first stage, we focused our proteomic exploration on the soluble protein complement of the green leaf mitochondria. We used traditional two-dimensional polyacrylamide gel electrophoresis, in combination with size exclusion chromatography as a third dimension, to identify the major proteins and further resolve their macromolecular complexity. The two-dimensional map of soluble proteins of green leaf mitochondria revealed 433 spots (with Coomassie blue staining) and around 73% of the proteins (in mass) were identified using three different approaches: Edman degradation, matrix-assisted laser desorption/ionization mass spectrometry and electrospray ionization tandem mass spectrometry. Quite a lot of the polypeptides were present in multiforms which indicated the presence of isoforms or the occurrence of post-translational modifications. Among these proteins, we uncovered an abundant family that was identified as aldehyde dehydrogenases, representing approximately 7.5% of the soluble proteins. The comparative analysis of soluble mitochondrial proteomes led to the identification of a number of proteins which were specifically present in root or in seed mitochondria, thus revealing the impact of tissue differentiation at the mitochondrial level. 相似文献

The existence of endogenous LH RH like immunoreactivity is shown in human urines after appropriate extraction, bu the radioimmunoassay of LH RH. In normaly cycling and menopausal women the quantities of endogenous hormone found in urines are greater after acid extraction than those found after extraction at pH 7. Furthermore, the increase observed by extraction in acidified methanol is directly correlated and proportional to the quantity of hormone assayable by extraction at pH 7. A highly abundant satellite DNA comprising 20% of the Meloidogyne fallax
(Nematoda, Tylenchida) genome was cloned and sequenced. The satellite
monomer is 173 bp long and has a high A + T content of 72.3%, with frequent
runs of A's and T's. The sequence variability of the monomers is 2.7%,
mainly due to random distribution of single-point mutations. A search for
evidence of internal repeated subunits in the monomer sequence revealed a
6-bp motif (AAATTT) for which five degenerated repeats, differing by just a
single base pair, could be identified. Pairwise comparison of the M. fallax
satellite with those from the sympatric species Meloidogyne chitwoodi and
Meloidogyne hapla revealed a high sequence similarity (68.39%) with one
satellite DNA subfamily in M. chitwoodi, which indicated an unexpected
close relationship between them. Given the high copy number and the extreme
sequence homogeneity among monomeric units, it may be assumed that the
satellite DNA of M. fallax could have evolved through some recent and
extensive amplification burst in the nematode genome. In this case, its
relatively short life would not yet have allowed the accumulation of random
mutations in independent amplified repeats. Considering the morphological
resemblance between the two species and their ability to produce
interspecific fertile hybrids under controlled conditions, these results
indicate that M. fallax may share a common ancestor with M. chitwoodi, from
which it could have diverged recently. All these data suggest that M.
fallax could be the result of a recent speciation process and show that
Meloidogyne satellite DNAs may be of interest to resolve phylogenetic
relationships among closely related species from this genus.

Morphological analysis of surface immunoglobulin biosynthesis in mouse B lymphoid cells using immunoelectron microscopy 总被引:1,自引:0,他引:1
Immunoelectron microscopic techniques were employed to investigate the biosynthesis of surface immunoglobulin (sIg) molecules in murine splenic B lymphocytes. In addition, cryoultramicrotomy was used due to its unique ability to retain antigenicity and provide good accessibility of external immunolabeling reagents to the cellular interior. In order to induce the biosynthesis of sIg, preexisting surface molecules were first removed completely by treatment with antibody and chymotrypsin. And the cells were then allowed to regenerate the expression of sIg molecules in culture. Our results clearly indicate that cytoplasmic immunoglobulin (cIg) exists in a close association with endoplasmic reticulum, Golgi, and vesicle structures within the cytoplasm as early as 15–30 min after antibody/enzyme treatment. After approximately 60 min, intracellular cIg molecules were observed to be incorporated into the plasma membrane through the fusion of cIg containing vesicles with the surface membrane. Reexpression of sIg reaches about 50% of its original level at 6 hr and 90% of its original level at 20 hr of incubation. Both the intracellular appearance of Ig-positive structures and reexpression of sIg on the cell surface are found to be inhibited by both puromycin and tunicamycin which suggest that both protein synthesis and glycosylation are required for sIg synthesis and reexpression. To our knowledge these studies have provided the first morphological analysis of the biosynthesis of membrane glycoproteins with intact whole cells. 相似文献
