The synthesis and biological activity of a series of aldehyde inhibitors of cathepsin K are reported. This paper aims at providing a better knowledge of factors determining body size (especially the importance of heredity) in Helix aspersa Müller.
A preliminary investigation of snails from Maghreb allowed us to show the importance of heredity in the variability of body size and to produce an efficient design for a subsequent reliable estimation of heritabilities. All traits measured (diameter, height and weight) were highly correlated and weight appeared to be the most convenient measure of size.
The second experiment provided 4150 snails born from known individuals among 500 wild snails. Pedigrees were recorded. Weight and diameter revealed high heritabilities (>0.4), which is relevant for commercial selection since variability of both traits was important. The design also revealed a significant non-genetic maternal effect and also that offspring from pairs where only one animal laid were bigger than offspring from pairs where both animals laid. This surprising observation has to be confirmed and its mechanisms studied. 相似文献

Florida pompano (Trachinotus carolinus) are a species of growing interest for commercial aquaculture. Effective health monitoring is crucial to the successful growout of the species, and prophylactic and therapeutic use of chemicals and antibiotics has been the traditional strategy for promoting stock health. However, concerns about antimicrobial resistance, chemical residues in seafood products and the environment, and resultant immunosuppression have prompted the industry to identify alternative management strategies, including supplementation with prebiotics, probiotics, and combinations of both (synbiotics). The objectives of this study are to determine and compare hematological, plasma biochemical, and plasma protein electrophoresis data of synbiotic-supplemented (β-glucan and Pediococcus acidilactici) and non-supplemented Florida pompano. Reference intervals for blood analytes are provided for both groups and for subgroups (females, males, large, and small fish) where statistically significant results exist. There are no differences between the hematological and plasma biochemistry analytes between the supplemented and control groups, except for blood urea nitrogen and carbon dioxide, indicating a possible effect of synbiotic supplementation on gill function and osmoregulation. Sex-related and size-related differences are observed within each of the control and supplemented groups; however, biometric measurements do not strongly correlate with blood analytes. These data represent baseline hematological and plasma biochemical data in the Florida pompano and indicate the safety of synbiotic supplementation in this commercially important species. This study serves to further the commercialization of Florida pompano by providing blood analyte reference intervals for health monitoring in the aquaculture setting. 相似文献

The effect of biotin deficiency on the metabolism of cholesterol was studied in rats fed cholesterol-free and cholesterol-containing
diet. Biotin deficiency induced by feeding raw egg-white resulted in higher cholesterol in the serum and aorta, and higher
high density lipoprotein cholesterol and low density lipoprotein + very low density lipoprotein cholesterol. In the liver,
cholesterol increased only in the cholesterol diet group but not in the cholesterol-free diet group. Levels of triglycerides
were lower in the biotindeficient, cholesterol-free diet group, but triglycerides were elevated in the cholesterol diet group.

A cytogenetic study has lead us to a stock of fertile heterozygotes for a triple translocation. The chromosomal rearrangement has first been detected in a female resulting from a cross between a normal female and a male submitted to X ray-irradiation. The aberration consists of rearrangements between a chromosome 3, a chromosome 6 and a chromosome 7. Abnormal chromosomes have the following constitution: 7q?: the terminal portion of the long arm is lost and replaced by the end of the short arm of the chromosome 3. 6 q+: the terminal portion of the long arm is lost and replaced by the end of the long arm of the chromosome 7. 3p+: the terminal portion of the short arm is lost and replaced by the end of the long arm of the chromosome 6. On the analogy of the human chromosome standardization, the formula of heterozygotes is 24, t (3p+, 6q+, 7q?). The first meiotic division shows both in the female and in the male 9 bivalents and one hexavalent. The formulae of the gametes are the same in both sexes. When a heterozygote is bred with a normal individual the offspring is composed of phenotypically normal or abnormal animals, depending on their karyotypes. The unbalanced karyotypes are lethal or semilethal. The importance of the malformations depends on the temperature of the water where the animals grow. Gastrointestinal malignancies account for about 20% of all cancers worldwide. It is widely accepted that cancer evolves through several stepwise morphological stages such as the adenoma-carcinoma and hyperplastic polyp-serrated adenoma-carcinoma sequences in colorectal cancers, and the metaplasia-dysplasia-carcinoma sequences in esophageal and gastric cancers. The morphological progression is associated with the accumulation of multiple genetic and epigenetic events. It is now recognized that epigenetic silencing of gene expression by CpG island methylation is an important alternative mechanism of inactivating tumor suppressor genes. Inflammatory conditions of the gastrointestinal and pancreaticobiliary tracts and liver such as Barrett esophagus, Helicobacter pylori gastritis, inflammatory bowel disease and viral hepatitis, are associated with increased frequency of malignancies and CpG methylation. In addition, CpG methylation is present in aberrant crypt foci and pancreatic intraepithelial neoplasia that are considered putative precursors of colon and pancreatic carcinomas, respectively. Understanding of these early genetic and epigenetic changes allows for the discoveries of potential screening, monitoring and therapeutic strategies. Targeting of the epigenetic changes that occur before the development of frank malignancy offers a potential chemopreventive strategy. 相似文献

Angiogenesis plays critical roles in the recovery phase of ischemic heart disease and peripheral vascular disease. An increase in autophagy is protective under hypoxic and chronic ischemic conditions. In the present study we determined the role of autophagy in angiogenesis. 3-Methyladenine (3-MA) and small interfering RNA (siRNA) against ATG5 were used to inhibit autophagy induced by nutrient deprivation of cultured bovine aortic endothelial cells (BAECs). Assays of BAECs tube formation and cell migration revealed that inhibition of autophagy by 3-MA or siRNA against ATG5 reduced angiogenesis. In contrast, induction of autophagy by overexpression of ATG5 increased BAECs tube formation and migration. Additionally, inhibiting autophagy impaired vascular endothelial growth factor (VEGF)-induced angiogenesis. However, inhibition of autophagy did not alter the expression of pro-angiogenesis factors such as VEGF, platelet-derived growth factor, or integrin αV. Furthermore, autophagy increased reactive oxygen species (ROS) formation and activated AKT phosphorylation. Inhibition of autophagy significantly decreased the production of ROS and activation of AKT but not of extracellular regulated kinase, whereas overexpression of ATG5 increased cellular ROS production and AKT activation in BAECs. Inhibition of AKT activation or ROS production significantly decreased the tube formation induced by ATG5 overexpression. Here we report a novel observation that autophagy plays an important role in angiogenesis in BAECs. Induction of autophagy promotes angiogenesis while inhibition of autophagy suppresses angiogenesis, including VEGF-induced angiogenesis. ROS production and AKT activation might be important mechanisms for mediating angiogenesis induced by autophagy. Our findings indicate that targeting autophagy may provide an important new tool for treating cardiovascular disease. 相似文献
