Question: Do severe winter flood events lift the nutrient limitation of biomass production in a river floodplain? How does this affect plant species richness? How long do the effects last? Location: Floodplain grassland on calcareous sandy loam near river Rhine in The Netherlands. Methods: Plots were fertilised with four treatments (control, N, P, N+P) for 21 years; plant species composition, vegetation biomass and tissue nutrient concentrations were determined every year between 1985 and 2005. Results: Fertilisation with N generally increased biomass production and reduced species richness, but these effects varied over time. During the first four years of the experiment, biomass production appeared to be co‐limited by N and P, while N fertilisation dramatically reduced plant species richness; these effects became weaker subsequently. Following two extreme winter floods in 1993–94 and 1994–95 and a drought in spring 1996, the effects of fertilisation disappeared between 1998 and 2001 and then appeared again. Flooding caused an overall reduction in species richness (from c. 24 to 15 species m‐2) and an increase in biomass production, which were only partly reversed after ten years. Conclusions: Long time series are necessary to understand vegetation dynamics and nutrient limitation in river floodplains, since they are influenced by occasional flood and drought events, whose effects may persist for more than ten years. A future increase in flooding frequency might be detrimental to species richness in floodplain grasslands.  相似文献

Climatic change has great impacts on stream catchments and their ecology. Expectations are that more extreme climate events will result in undesired flooding in stream catchments. In the Netherlands, former floodplains with a history of agricultural use are put into use again as flooding areas for the purpose of water retention. This study focuses on the effects of winter flooding on various plant site conditions in the soil, such as redox, nutrient, pH, and base status. We compared the effects on groundwater- and rainwater-dominated floodplains. Water chemistry (pH, EC, HCO3, SO4, Cl, Ca, Mg, Na, K, , NO3, NH4, and PO4) and soil nutrients (Total N and P, and bio-available P) were monitored for one year, including a 3–4 month period with winter flooding. In both floodplains no direct effect of the flood-water chemistry was detected in the pore water, because the soil pores had become saturated with groundwater or rainwater, respectively, just before flooding, flood-water did not penetrate the soil. We found that the increase in pH and ammonium concentration in the rainwater floodplain were due to changes in redox status, resulting from the completely water-filled state of the soil pores during the flooding event. Furthermore, we noticed an increase in soil nutrient contents and a shift in plant species composition in the rainwater floodplain: the vegetation included more plant species characteristic for N-richness. Finally, we conclude that winter flooding has more drastic effects on biogeochemical conditions and vegetation composition in the atmotrophic conditions characteristic for low-order subcatchments than in lithotrophic conditions in the larger, higher-order subcatchments of the stream basin.  相似文献

ObjectivesRheumatoid arthritis (RA) has been identified as an independent cardiovascular risk factor. The importance of risk factors such as hypertension and hyperlipidemia in the generation of atherosclerosis in RA patients is unclear. This study analyzed clinical parameters associated with carotid intima media thickness (cIMT) in patients with RA.MethodsSubjects with RA and healthy controls without RA, both without known cardiovascular disease, were included. Participants underwent a standard physical examination and laboratory measurements including a lipid profile. cIMT was measured semi-automatically by ultrasound.ResultsIn total 243 RA patients and 117 controls were included. The median RA disease duration was 7 years (IQR 2–14 years). The median DAS28 was 2.4 (IQR 1.6–3.2) and 114 (50.4%) of the RA patients were in remission. The presence of RA and cIMT were not associated (univariate analysis). Multivariable regression analysis showed that cIMT in RA patients was associated with age (B = 0.006, P<0.001) and systolic blood pressure (B = 0.003, P = 0.003). In controls, cIMT was associated with age (B = 0.006, P<0.001) and smoking (B = 0.097, P = 0.001).ConclusioncIMT values were similar between RA patients and controls. Hypertension was strongly associated with cIMT in RA patients. After adjustment, no association between cIMT and specific RA disease characteristics was found in this well treated RA cohort.  相似文献

Background

Diets rich in whole grain are associated with several health benefits. Little is known however, about whole grain consumption patterns in Malaysia. The aim of this study was to assess whole grain intakes and dietary source in Malaysian children and adolescents.

Methods

This analysis is from the MyBreakfast study, a national cross sectional study investigating eating habits among primary and secondary school children throughout Malaysia, conducted in 2013. Children (n = 5,165) and adolescents (n = 2,947) who completed two days of dietary assessment using a food record or recall respectively were included. The whole grain content of foods was estimated mainly through the use of quantitative ingredient declarations on food labels. All wholegrain foods were considered irrespective of the amount of whole grain they contained.

Results

Overall, only 25% of children and 19% of adolescents were wholegrain consumers. Mean daily intakes in the total sample were 2.3g/d (SD 5.8g/d) in children and 1.7g/d (SD 4.7g/d) in adolescents and in the consumer’s only sample, mean intakes reached 9.1g/d (SD 8.6) and 9.2g/d (SD 7.1g/d) respectively. Wheat was the main grain source of whole grain while ready to eat breakfast cereals and hot cereals were the main food contributors. Less than 3% of the children and adolescents reached the US quantitative whole grain recommendation of 48g/day.

Conclusion

Whole grain is consumed by only a minority of Malaysian children and adolescents and even among consumers, intakes are well below recommendations. Efforts are needed to firstly understand the barriers to whole grain consumption among Malaysian children in order to design effective health promotion initiatives to promote an increase in whole grain consumption.  相似文献

Summary A gradient-enhanced 3D NOESY-HMQC experiment is presented and applied to15N-labeled Mnt repressor (1–76) in1H2O. Both coherence selection and1H2O suppression are achieved using gradients. A singlescan experiment can be recorded for each time increment, which greatly reduces recording time with respect to conventional methods using phase cycling. This can be of use for kinetic studies and for relatively unstable molecules.  相似文献

The influence of several metabolic inhibitors and pharmacologic agents on macrophage deformation (induced by fluid shear stress) was examined in relationship to changes in ATP content and phagocytosis of latex beads. Two relatively specific inhibitors of glycolysis (iodoacetate [IA], and sodium fluoride [NaF]) and a sulfhydryl-binding agent (N-ethylmaleimide [NEM] markedly inhibited phagocytosis and reduced cell deformability. A microtubule-disrupting agent (vinblastine) and a highly specific inhibitor of glycolysis (2-deoxyglucose) markedly inhibited phagocytosis without influencing cell deformability. An organomercurial sulfhydryl binding agent p-chloromercuribenzene (PCMBS) and a microfilament-disrupting agent (cytochalasin B) inhibited phagocytosis and increased cell deformability. The effects of these agents on phagocytosis and cell deformability bore no consistent relationship to alterations in cellular content of ATP. The observation that 2-deoxyglucose, the most specific inhibitor of glycolysis examined, reduced ATP content to levels far lower (15 percent of control values) than those achieved by any other agent examined and inhibited phagocytosis without altering cell deformability, suggests that alterations in cell deformability induced by NaF, IA, NEM, PCMBS, and cytochalasin B are not due to inhibition of glycolysis per se, but instead result from direct or indirect effects of these agents on cell constituents, possibly contractile proteins, which are determinants of cell deformability. The finding that cytochalasin B, NEM, PCMBS, and IA interfere with phagocytosis and alter cell deformability, together with evidence that these agents interact with isolated actin and myosin, suggests that contractile proteins are important both in phagocytosis and as determinants of cell deformability. The observation that vinblastine, colchicines, and heavy water (D(2)O) did not alter cell deformability, even though vinblastine caused formation of intracellular crystals of microtubular protein, indicates that microtubules are not major determinants of cell deformability. The observations that beads adhered normally to surfaces of cytochalasin B- and of PCMBS-treated cells and that shear-stress induced deformation was increased whereas phagocytosis was markedly inhibited, suggest that deformation of cells around beads associated with ingestion depends on some form of cellular (contractile?) activity, whereas deformation of cells by fluid shear stress is a passive phenomenon.  相似文献

To develop a nisin-producing cheese starter, Lactococcus lactis subsp. cremoris SK110 was conjugated with transposon Tn5276-NI, which codes for nisin immunity but not for nisin production. Cheese made with transconjugant SK110::Tn5276-NI as the starter was bitter. The muropeptide of the transconjugant contained a significantly greater amount of tetrapeptides than the muropeptide of strain SK110, which could have decreased the susceptibility of the cells to lysis and thereby the release of intracellular debittering enzymes.  相似文献

In fibroblasts, stimulation of receptor tyrosine kinases results in the activation of the extracellular signal-regulated kinase 2 (ERK2). The major signalling pathway employed by these receptors involves the activation of p21ras and raf-1 kinase. Here we show that in NIH3T3 and rat-1 fibroblasts, elevation of the intracellular cAMP level results in the inhibition of ERK2 activation induced by PDGF, EGF and insulin treatment. Analysis of various signalling intermediates shows that cAMP interferes at a site downstream of p21ras, but upstream of raf-1 kinase. Inhibition by cAMP depends on both the cAMP concentration and the absolute amount of p21ras molecules bound to GTP, suggesting a mechanism of competitive inhibition. Also TPA-induced, p21ras-independent, activation of raf-1 kinase and ERK2 is inhibited by cAMP. We have used the inhibitory effect of cAMP to investigate whether phosphorylation of mSos, a p21ras nucleotide exchange factor, is dependent on the activity of the raf-1 kinase/ERK2 pathway. We found that phosphorylation of mSos, as monitored by a mobility shift, is delayed with respect to p21ras and ERK2 activation and is inhibited by cAMP in a similar cell type- and concentration-dependent manner as the inactivation of ERK2. These results provide evidence for a model of p21ras-directed signalling towards ERK2 that feeds back on mSos by regulating its phosphorylation status and that can be negatively modulated by protein kinase A and positively modulated by protein kinase C action.  相似文献
