Advanced personalized dosimetry for molecular nuclear therapy has been shown to be feasible in clinical practice. At the same time instrumentation and dosimetric software are still evolving at a high pace. Procedures developed so far differ in approach and sophistication, and standard operating procedures necessary for accurate patient specific dosimetry do not yet exist. Expression of the Myc oncoprotein is downregulated in response to stress signals to allow cells to cease proliferation and escape apoptosis, but the mechanisms involved in this process are poorly understood. Cell cycle arrest in response to DNA damage requires downregulation of Myc via a p53-independent signaling pathway. Here we have used siRNA?screening of the human kinome to identify MAPKAPK5 (MK5, PRAK) as a negative regulator of Myc expression. MK5 regulates translation of Myc, since it is required for expression of miR-34b and miR-34c that bind to the 3'UTR of MYC. MK5 activates miR-34b/c expression via phosphorylation of FoxO3a, thereby promoting nuclear localization of FoxO3a and enabling it to induce miR-34b/c expression and arrest proliferation. Expression of MK5 in turn is directly activated by Myc, forming a negative feedback loop. MK5 is downregulated in colon carcinomas, arguing that this feedback loop is disrupted during colorectal tumorigenesis. 相似文献

Oxygen concentration profiles have been measured, by means of with microelectrodes in sediments of Lake Baikal and Lake Malawi,
along transects allowing to give a survey of two major ancient Rift lakes: Lake Baikal (Eastern Siberia) and Lake Malawi (East
Africa), along depth transects in the constitutive basins of the lakes and/or of relevant depths with regard to oxygen (including
including the deepest point, 1680 m, in Lake Baikal). Sediment oxygen penetration depths (SOPs) display very different patterns,
depending on the lake in the two lakes. In Lake Baikal, SOPs are variable, show no significant relationship with bathymetric
depth and are surprisingly deep on Akademichesky ridge (> 50.0 mm), emphasizing the distinctive feature of this region in
the lake. While the Selenga river is an important source of eutrophication, the similarity of SOP-values in the Selenga shallow
with those of most other sites suggests either a dilution of organic material by allochthonous matter, or a strong south-to-north
transport of particles. In Lake Malawi, available oxygen is restricted to a maximum of three millimetres of the sediment,
and there is a negative relationship with bathymetric depth, as a result of a steady decline of oxygen concentration with
depth through the water column. Amongst the few parameters known to affect SOPs, the oxygen consumption by the sediment seems
the most significant in both lakes. SOP-values furthermore confirm differences in the trophic status of Baikal and Malawi,
respectively. The importance of oxygen as a factor likely to create ecological segregation for benthic organisms is discussed.
Lake Malawi offers possibilities of bathymetric segregation but no vertical segregation in the sediment. In contrast, no bathymetric
segregation related to oxygen is possible in Lake Baikal, but vertical segregation in the sediment is very likely.

The Hedgehog signaling pathway is critical for a significant number of developmental patterning events. In this study, we focus on the defects in pharyngeal arch and cardiovascular patterning present in Sonic hedgehog (Shh) null mouse embryos. Our data indicate that, in the absence of Shh, there is general failure of the pharyngeal arch development leading to cardiac and craniofacial defects. The cardiac phenotype results from arch artery and outflow tract patterning defects, as well as abnormal development of migratory neural crest cells (NCCs). The constellation of cardiovascular defects resembles a severe form of the human birth defect syndrome tetralogy of Fallot with complete pulmonary artery atresia. Previous studies have demonstrated a role for Shh in NCC survival and proliferation at later stages of development. Our data suggest that SHH signaling does not act directly on NCCs as a survival factor, but rather acts to restrict the domains that NCCs can populate during early stages (e8.5-10.5) of cardiovascular and craniofacial development. 相似文献

Earlier findings show seasonality in processes and behaviors such as brain plasticity and depression that in part are regulated by Brain-Derived Neurotrophic Factor (BDNF). Based on this we investigated seasonal variation in serum BDNF concentrations in 2,851 persons who took part in the Netherlands Study of Depression and Anxiety (NESDA). Analyses by month of sampling (monthly n’s >196) showed pronounced seasonal variation in serum BDNF concentrations (P<.0001) with increasing concentrations in the spring-summer period (standardized regression weight (ß) = 0.19, P<.0001) and decreasing concentrations in the autumn-winter period (ß = −0.17, P<.0001). Effect sizes [Cohen’s d] ranged from 0.27 to 0.66 for monthly significant differences. We found similar seasonal variation for both sexes and for persons with a DSM-IV depression diagnosis and healthy control subjects. In explorative analyses we found that the number of sunshine hours (a major trigger to entrain seasonality) in the week of blood withdrawal and the 10 weeks prior to this event positively correlated with serum BDNF concentrations (Pearson’s correlation coefficients ranged: 0.05 – 0.18) and this could partly explain the observed monthly variation. These results provide strong evidence that serum BDNF concentrations systematically vary over the year. This finding is important for our understanding of those factors that regulate BDNF expression and may provide novel avenues to understand seasonal dependent changes in behavior and illness such as depression. Cardiac cushion formation is crucial for both valvular and septal development. Disruption in this process can lead to valvular and septal malformations, which constitute the largest part of congenital heart defects. One of the signaling pathways that is important for cushion formation is the TGFβ superfamily. The involvement of TGFβ and BMP signaling pathways in cardiac cushion formation has been intensively studied using chicken in vitro explant assays and in genetically modified mice. The life-cycle of the ancient asexual ostracod Darwinula stevensoni was studied during 1 year in a eutrophic pond in Belgium. The reproductive period of this species started in March and was effectively completed by September of the same year. All changes in population structure took place during the spring and summer months and a rapid turnover of the instars was observed. The life-cycle of Darwinula stevensoni appears to take one year or less in Belgium and this is considerably shorter than the 4 years which had been reported previously from subarctic populations. The difference to the present study is most likely temperature-related. Maximal densities of D. stevensoni were observed in June and July and attained 105 ind. m–2. During winter, densities were lower with a mean of 104 ind. m–2. Consequently, the calculated population size of each month was high throughout the year. Together with the low mutation rate, such a large population size could effectively counteract the stochastic loss of mutation-free genotypes as predicted by Muller's ratchet. D. stevensoni is a brooder; the maximum number of embryos and juvenile instars (up to third stage) found within a single female was 11. 相似文献
