Two Amerindian populations of French Guiana were investigated for plasma proteins and red-cell enzymes. In the Wayampi tribe, rare variants were identified in 4 systems. The corresponding alleles are designated AK1 3 Wayampi, PGM1 (4/10) Wayampi, PGM2 6 Wayampi, and TfD. In the Emerillon tribe, a variant allele of PGM2, designated PGM2 6 Emerillon, was identified. For three of the systems, PGM1, PGM2, Tf, similar isozymes have been described in other Amerindian populations. These findings suggest that the corresponding alleles may have the same origin. 相似文献

Starting from the interaction of galangin (3,5,7-trihydroxyflavone) with a cytosolic nucleotide-binding domain of P-glycoprotein, a series of flavonol derivatives was synthesized and tested for their binding affinity towards the same target. Over the last decade, bamboo has emerged as an interesting plant for the treatment of various polluted waters using plant-based wastewater treatment systems. In these systems, nitrogen and phosphorous concentrations in wastewater can exceed plant requirements and potentially limit plant growth. The effects of two nutrient rates on the growth of seven bamboo species were assessed in a one-year experiment: Dendrocalamus strictus, Thyrsostachys siamensis, Bambusa tuldoides, Gigantochloa wrayi, Bambusa oldhamii, Bambusa multiplex and Bambusa vulgaris. Nutrient rates were applied with a 20:20:20 NPK fertilizer as 2.6 and 13.2 t.ha.yr?1NPK to three-year-old bamboo planted in 70 L containers. Morphological characters, photosynthetic responses, and NPK content in bamboo tissues were investigated. Under high-nutrient supply rate, the main trend observed was an increase of culm production but the culms’ diameters were reduced. For the seven species, the aboveground biomass yield tended to increase with high-nutrient rate. Increasing in nutrient rates also improved the photosynthetic activity which is consistent with the increase of nitrogen and phosphorus contents measured in plant tissues. All the bamboo species tested appears suitable for wastewater treatment purposes, but the species Bambusa oldhamii and Gigantochloa wrayi showed the higher biomass yield and nutrient removal. 相似文献

A library of halogenated 2-arylindolyl-3-oxocarboxamides was prepared to develop radioligands to visualize cerebral PBR by SPECT and PET imaging. In vitro evaluation showed that most of the synthesized compounds were selective,high-affinity PBR ligands with adequate lipophilicity (log D7.4 in the range of 1.6-2.4). Vinculin regulates both cell-cell and cell-matrix junctions and anchors adhesion complexes to the actin cytoskeleton through its interactions with the vinculin binding sites of alpha-actinin or talin. Activation of vinculin requires a severing of the intramolecular interactions between its N- and C-terminal domains, which is necessary for vinculin to bind to F-actin; yet how this occurs in cells is not resolved. We tested the hypothesis that talin and alpha-actinin activate vinculin through their vinculin binding sites. Indeed, we show that these vinculin binding sites have a high affinity for full-length vinculin, are sufficient to sever the head-tail interactions of vinculin, and they induce conformational changes that allow vinculin to bind to F-actin. Finally, microinjection of these vinculin binding sites specifically targets vinculin in cells, disrupting its interactions with talin and alpha-actinin and disassembling focal adhesions. In their native (inactive) states the vinculin binding sites of talin and alpha-actinin are buried within helical bundles present in their central rod domains. Collectively, these results support a model where the engagement of adhesion receptors first activates talin or alpha-actinin, by provoking structural changes that allow their vinculin binding sites to swing out, which are then sufficient to bind to and activate vinculin. 相似文献

In granulomatous inflammatory lung diseases such as sarcoidosis, the balance of cytokine production by activated T cells in the lungs may influence clinical disease outcome. To investigate the potential of T lymphocytes to produce cytokines and contribute to this process, T cells from bronchoalveolar lavage (BAL) and PB from 19 patients with active lung disease were stimulated, stained, and analysed by flow cytometry for intracellular production of cytokines and expression of the activation marker CD69. Higher proportions of BAL cells expressed CD69 compared with PB, in the absence of in vitro stimulation. The expression of IFN-γ was similar in unstimulated BAL and PB T cells, and there was no association between the expression of CD69 and IFN-γ. Following stimulation, there were increased numbers of IFN-γ+ T cells. A similar trend was found with IL-2+ T cells, but there were lower levels of IL-4+ T cells in BAL compared with PB, and similar levels of IL-10+ T cells. The presence of activated T lymphocytes in BAL samples from patients with sarcoidosis, with the potential to produce Th1 type 1 cytokines may contribute to the inflammatory processes in this granulomatous lung disease. The use of intracellular flow cytometry to investigate cytokine production by BAL T cells could help to indicate potential targets for future therapy. 相似文献
