Taste buds, the sensory organs for taste, have been described as arising solely from the surrounding epithelium, which is in distinction from other sensory receptors that are known to originate from neural precursors, i.e., neural ectoderm that includes neural crest (NC). Our previous study suggested a potential contribution of NC derived cells to early immature fungiform taste buds in late embryonic (E18.5) and young postnatal (P1-10) mice. In the present study we demonstrated the contribution of the underlying connective tissue (CT) to mature taste buds in mouse tongue and soft palate. Three independent mouse models were used for fate mapping of NC and NC derived connective tissue cells: (1) P0-Cre/R26-tdTomato (RFP) to label NC, NC derived Schwann cells and derivatives; (2) Dermo1-Cre/RFP to label mesenchymal cells and derivatives; and (3) Vimentin-CreER/mGFP to label Vimentin-expressing CT cells and derivatives upon tamoxifen treatment. Both P0-Cre/RFP and Dermo1-Cre/RFP labeled cells were abundant in mature taste buds in lingual taste papillae and soft palate, but not in the surrounding epithelial cells. Concurrently, labeled cells were extensively distributed in the underlying CT. RFP signals were seen in the majority of taste buds and all three types (I, II, III) of differentiated taste bud cells, with the neuronal-like type III cells labeled at a greater proportion. Further, Vimentin-CreER labeled cells were found in the taste buds of 3-month-old mice whereas Vimentin immunoreactivity was only seen in the CT. Taken together, our data demonstrate a previously unrecognized origin of taste bud cells from the underlying CT, a conceptually new finding in our knowledge of taste bud cell derivation, i.e., from both the surrounding epithelium and the underlying CT that is primarily derived from NC.  相似文献

Behavior of spin labels in a variety of interdigitated lipid bilayers   总被引:2,自引:0,他引:2  
The behavior of a number of spin labels in several lipid bilayers, shown by X-ray diffraction to be interdigitated, has been compared in order to evaluate the ability of the spin label technique to detect and diagnose the structure of lipid bilayers. The main difference between interdigitated and non-interdigitated gel phase bilayers which can be exploited for determination of their structure using spin labels, is that the former have a much less steep fluidity gradient. Thus long chain spin labels with the nitroxide group near the terminal methyl of the chain, such as 16-doxylstearic acid, its methyl ester, or a phosphatidylglycerol spin label containing 16-doxylstearic acid (PG-SL), are more motionally restricted and/or ordered in the interdigitated bilayer than in the non-interdigitated bilayer. This difference is large enough to be of diagnostic value for all three spin labels in the interdigitated bilayers of dihexadecylphosphatidylcholine, dipalmitoylphosphatidylcholine/ethanol, and 1,3-dipalmitoylphosphatidylcholine. However, it is not large enough to be of diagnostic value at low temperatures. Use of probes with the nitroxide group closer to the apolar/polar interface reveals that these latter interdigitated bilayers are more disordered or less closely packed. As the temperature is increased, however, the motion of the PG-SL does not increase as much in these interdigitated bilayers as in non-interdigitated bilayers. The difference in the motion and/or order of PG-SL between interdigitated and non-interdigitated bilayers is large enough at higher temperatures to be of value in diagnosing the structure of the bilayers. Thus by choice of a suitable spin label and a suitable temperature, this technique should prove useful for detection and diagnosis of lipid bilayer structure with a good degree of reliability. Caution must, of course be exercised, as with any spectroscopic technique. Spin labels will also be invaluable for more detailed studies of known interdigitated bilayers, which would be time- and material-consuming, if carried out using X-ray diffraction solely.  相似文献

Replication-defective vectors derived from reticuloendotheliosis virus were used to transduce exogenous genes into early somatic stem cells of the chicken embryo. One of these vectors transduced and expressed the chicken growth hormone coding sequence. The helper cell line, C3, was used to generate stocks of vector containing about 10(4) transducing units per ml. Injection of 5- to 20-microliters volumes of vector directly beneath the blastoderm of unincubated chicken embryos led to infection of somatic stem cells. Infected embryos and adults contained unrearranged integrated proviral DNAs. Embryos expressed the transduced chicken growth hormone gene and contained high levels of serum growth hormone. Blood, brain, muscle, testis, and semen contained from individuals injected as embryos contained vector DNA. Replication-defective vectors of the reticuloendotheliosis virus transduced exogenous genes into chicken embryonic stem cells in vivo.  相似文献

Health insurance in the United States is failing patients and physicians alike. In this country 37 million uninsured face economic barriers to care, and the health of many suffers as a result. The "corporatization" of medical care threatens professional values with an unprecedented administrative and commercial intrusion into the daily practice of medicine. Competitive strategies have also failed their most ostensible goal--cost control. In contrast, Canada offers a model of a national health insurance plan that provides universal and comprehensive coverage, succeeds at restraining health care inflation, and does little to abrogate the clinical autonomy of physicians in private practice. I propose that American physicians relent in their historical opposition to national health insurance and participate in the development of a universal, public insurance plan responsive to the needs of both patients and physicians.  相似文献

From a single aflatoxin B1 oxime — bovine serum albumin conjugate, polyclonal and monoclonal antibody preparations were produced. The four rabbit polyclonal antisera were specific for aflatoxin Bi in a microtitration plate enzyme — linked immunosorbent assay. The monoclonal antibodies showed a wide range of differing specificities, recognizing, for example, aflatoxins B1, B2, G1 and G2; B1 and B2; B1 and G1; and G1 alone. No antibody preparations reacted with aflatoxin M1. The significance of these results to the strategy of anti-aflatoxin antibody production for use in quantitative enzyme immunoassays is discussed.  相似文献

Summary The fractional concentration of CO2 within the occupied burrows of burrow-dwelling birds has been measured at 0.065 or higher while the fractional concentration of O2 can be 0.14 or lower. The ventilatory responses of the Burrowing Owl (Athene cunicularia) and a non-burrowing bird, the Bobwhite (Colinus virginianus), to the levels of hypercarbia, hypoxia and hypoxic hypercarbia likely encountered by this owl within its burrow were assessed. Ventilatory responses toFi CO 2 of 0.01, 0.03, 0.05 and 0.07 at normoxia; toFi CO 2 of 0.19, 0.17, 0.15 and 0.13 at normocarbia; and to combinedFi CO 2 andFi O 2 of 0.01:0.19, 0.03:0.17, 0.05:0.15 and 0.07:0.13 were measured by volumetric plethysmography. The Burrowing Owl exhibited a significantly attenuated ventilatory response to these levels of hypercarbia, hypoxia and hypoxic hypercarbia compared to the non-burrow-dwelling Bobwhite. A reduced ventilatory response to the hypoxic and hypercarbic stimuli has been previously observed in fossorial mammals, and is reported here for the first time in a burrow-dwelling bird. Although serum is generally regarded to increase the permeability of liposomes containing entrapped substances, we found that a low concentration of serum (10%) significantly reduced the permeability of liposomes to the spin label tempocholine chloride and the polar drug methotrexate, although it increased the permeability of the lipid-soluble drug actinomycin D. Liposomes containing sphingomyelin and cholesterol were considerably less permeable than liposomes containing phosphatidylcholine and cholesterol. Although a higher concentration of serum (88%) increased the permeability of liposomes containing either lipid, the amount of tempocholine which had leaked from sphingomyelin-containing liposomes in 88% serum after 50 h at 37 degrees C was only 25%, three times less than that from phosphatidylcholine-containing liposomes. Thus the effect of serum on liposome permeability depends on the compound entrapped as well as the type of lipid used.  相似文献

—High circulating levels of phenylalanine caused depletions of threonine, valine, methionine, isoleucine, leucine, histidine, tryptophan, and tyrosine in immature and adult rat brains. The branched-chain amino acids were most affected. Their reductions ranged between 38–64 per cent of control values when phenylalanine was administered either parenterally or in the diet. The pattern of cerebral amino acid depletions found in phenylalanine-injected infant rats was similar to that of the adults. Phenylalanine loading caused depletions in serum amino acid levels in adult rats, but in infant rats the serum levels were either unchanged or somewhat elevated. Tyrosine, when administered to adult rats either parenterally or via the diet, caused cerebral depletions in essential amino acids, but the depletions were not as striking as with phenylalanine. In both the infant and adult rat, brain-blood ratios of most of the essential amino acids were significantly reduced by phenylalanine loading.  相似文献

Synthetic species of CBS containing palmitic, stearic, lignoceric, D-2-hydroxy palmitic, or D-2-hydroxy stearic acid were prepared and their phase behavior in the presence of a number of mono- and divalent cations was studied by differential scanning calorimetry and the use of fatty acid spin labels. The results showed that both the non-hydroxy fatty acid (NFA) and hydroxy fatty acid (HFA) forms of cerebroside sulfate (CBS) can occur in two different gel states, a metastable state and a lower entropy stable state. The phase behavior is more sensitive to the type and concentration of cation present than in the case with acidic phospholipids. The sensitivity of the transition temperature (Tm) to cation concentration reflects, in part, increased participation of the lipid in intermolecular hydrogen bonding interactions as the negative charge of the sulfate is shielded. The extra hydroxyl group on the HFA also contributes to the intermolecular hydrogen bonding network causing a significant increase in the Tm.The HFA has an even more significant effect in causing inhibition of formation of the stable state. Formation of the stable state is also inhibited by Li+ and divalent cations. A similar mechanism may be involved, i.e.; cross-linking of adjacent lipids or increased intermolecular interactions inhibit the molecular rearrangement necessary to form the stable state. This inhibition is counteracted by an increase in fatty acid chain length. The results suggest that the stable state may be interdigitated as a result of the unequal chain length between the sphingosine base and the fatty acid.  相似文献
