We have compared the influence of two aromatic cytokinin derivatives, N 6-benzyladenine (BA) and meta-topolin (mT), on the in vitro multiplication and senescence of wych elm (Ulmus glabra Huds.). After 3 months of cultivation, the micropropagation rate was higher (approx. six times more shoots developed) on Murashige and Skoog (MS) medium supplemented with mT than on MS supplemented with BA. Quantification of 50 endogenous cytokinins, using a recently developed UPLC-ESI(+)-MS/MS method, showed significant differences in the cytokinin metabolites (especially different glucosides) in explants cultivated on media supplemented with BA or mT. Analytic approximate solutions using Optimal Homotopy Perturbation Method (OHPM) are given for steady boundary layer flow over a nonlinearly stretching wall in presence of partial slip at the boundary. The governing equations are reduced to nonlinear ordinary differential equation by means of similarity transformations. Some examples are considered and the effects of different parameters are shown. OHPM is a very efficient procedure, ensuring a very rapid convergence of the solutions after only two iterations. 相似文献

Miscanthus × giganteus is a popular energy crop, which due to its hybrid origin is only vegetatively reproduced. Asexual embryogenesis in anther and microspore culture leading to double haploids production could allow to regain the ability for sexual reproduction and to increase the biodiversity of the species. Therefore, the goal of this paper was to investigate the requirements of androgenesis in Miscanthus. The standard protocols used for monocotyledonous plants were applied with many modifications regarding the developmental stage of the explants at the time of culture initiation, stress treatment applied to panicles and isolated anthers as well as various chemical and physical parameters of in vitro culture conditions. Our results indicated that the induction of androgenesis in M. × giganteus is possible. However, the very low efficiency of the process and the lack of regeneration ability of the androgenic structures presently prevent the use of this technique. 相似文献

In the shrewsSorex araneus Linnaeus, 1758 andS. minutus Linnaeus, 1766 a short period of fast post-natal development in the nest is followed by a longer summer period of developmental stasis and then by autumn regression. The aim of this paper has been to test whether this period of developmental stability also is accompanied by inhibited development of the eye lens — the most continuously growing organ of the mammalian body. If it is not, then can this be used as an age indicator in studies on shrews, and can it serve in distinguishing seasonal cohorts of young born in consecutive summer months? This aspect was examined on material comprising dead shrews collected from live traps and pitfalls set for rodent research in the marshy habitat of a river valley in Bia?owie?a Forest, Poland, in 1997–1999. In fact, the eye lens in shrews was shown to increase in size continuously; therefore it may be used as a reliable age indicator. Body mass of weaned young shrews also continued to increase slowly with age until autumn. On the basis of the distribution of individuals in successive lens-mass classes, three age groups (cohorts) of young could be distinguished each season in theS. araneus population, and two inS. minutus. In the former species, consistent patterns to the relative abundance of the three seasonal cohorts were observed. Gliomas are common brain tumours, but obtaining tissue for definitive diagnosis can be difficult. There is, therefore, interest in the use of non-invasive methods to diagnose and grade the disease. Although positron emission tomography (PET) with 18F-fluorethyltyrosine (18F-FET) can be used to differentiate between low-grade (LGG) and high-grade (HGG) gliomas, the optimal parameters to measure and their cut-points have yet to be established. We therefore assessed the value of single and dual time-point acquisition of 18F-FET PET parameters to differentiate between primary LGGs (n = 22) and HGGs (n = 24). PET examination was considered positive for glioma if the metabolic activity was 1.6-times higher than that of background (contralateral) brain, and maximum tissue-brain ratios (TBRmax) were calculated 10 and 60 min after isotope administration with their sums and differences calculated from individual time-point values. Using a threshold-based method, the overall sensitivity of PET was 97%. Several analysed parameters were significantly different between LGGs and HGGs. However, in a receiver operating characteristics analysis, TBR sum had the best diagnostic accuracy of 87% and sensitivity, specificity, and positive and negative predictive values of 100%, 72.7%, 80%, and 100%, respectively. 18F-FET PET is valuable for the non-invasive determination of glioma grade, especially when dual time-point metrics are used. TBR sum shows the greatest accuracy, sensitivity, and negative predictive value for tumour grade differentiation and is a simple method to implement. However, the cut-off may differ between institutions and calibration strategies would be useful. 相似文献
