The formation of mature secretory granules is essential for proper storage and regulated release of hormones and neuropeptides. In pancreatic β cells, cholesterol accumulation causes defects in insulin secretion and may participate in the pathogenesis of type 2 diabetes. Using a novel cholesterol analog, we show for the first time that insulin granules are the major sites of intracellular cholesterol accumulation in live β cells. This is distinct from other, non‐secretory cell types, in which cholesterol is concentrated in the recycling endosomes and the trans‐Golgi network. Excess cholesterol was delivered specifically to insulin granules, which caused granule enlargement and retention of syntaxin 6 and VAMP4 in granule membranes, with concurrent depletion of these proteins from the trans‐Golgi network. Clathrin also accumulated in the granules of cholesterol‐overloaded cells, consistent with a possible defect in the last stage of granule maturation, during which clathrin‐coated vesicles bud from the immature granules. Excess cholesterol also reduced the docking and fusion of insulin granules at the plasma membrane. Together, the data support a model in which cholesterol accumulation in insulin secretory granules impairs the ability of these vesicles to respond to stimuli, and thus reduces insulin secretion.  相似文献

Freshwater mussels of the order Unionida have been dramatically declining globally. Despite their ecological importance, conservation of these animals has been hindered by unresolved taxonomy and a lack of data on the distribution and status of populations, especially in southern Europe. Although the Italian Peninsula has been noted as a centre of endemism and one of the major refugia of the glacial ages for several taxa, few studies have been performed on the genetic diversity of Unionida. Most importantly, the taxonomic status of several freshwater mussel populations of the Italian Peninsula is still unresolved. Here we present the first comprehensive dataset for the Unionida of the region spanning Italy and the coastal Croatian region (west of the Dinaric Alps). In total, 191 specimens were collected (85 Anodonta, 64 Unio, 17 Microcondylaea bonellii and 25 Sinanodonta woodiana) from 34 sites across the Italian Peninsula and coastal Croatian river basins for molecular identification (COI, 16S and 28S). Genetic analyses were performed to understand major phylogenetic and phylogeographic patterns. Seven species were detected: three Anodonta species (A. anatina, A. cygnea and A. exulcerata), two Unio species (U. mancus and U. elongatulus), Microcondylaea bonellii, and the invasive Sinanodonta woodiana. The presence of three endemic species (A. exulcerata, U. elongatulus and M. bonellii) confirms the importance of the region as a centre of endemism for freshwater mussels. The Apennine Mountains act as an important biogeographic barrier.  相似文献

Concentrations of domoic acid (DA), the biotoxin responsible for amnesic shellfish poisoning (ASP), exceeding the regulatory limit of 20 μg g−1 have caused restricted harvesting and closures of wild king scallop fisheries. Toxin monitoring programmes have reported significant inter-animal variation in DA concentration between scallops from the same area. For the development of reliable sampling and management protocols an understanding of the magnitude and causes of inter-animal variation in toxin concentration are important. Ten samples were collected from an aquaculture site in Clew Bay, Co. Mayo off the west coast of Ireland between February 2003 and February 2004, each sample comprising 12 scallops of each of the following size groups: small (70–85 mm), medium (85–100 mm), large (100–115 mm) and very large (>115 mm). DA concentration in each hepatopancreas and in composite samples of both gonad and adductor muscle from each size group on each sampling occasion were measured. High inter-animal variability in DA concentration in hepatopancreas was recorded; CVs ranging from 12.5% to 82.5%. One negative correlation (R2 = 0.7079) between DA concentration in hepatopancreas and scallop shell length, three positive but weak correlations (R2 = 0.4536, 0.3459 and 0.4665) and six no correlations were exhibited. Negative correlations were attributed to faster DA uptake by smaller scallops, positive correlations to faster DA depuration by smaller scallops. If only scallops greater than or equal to 100 mm shell length, the minimum commercial size of this species were considered, no correlation occurred on any of the 10 sampling occasions.  相似文献

Domoic acid (DA), the toxin responsible for amnesic shellfish poisoning (ASP) can accumulate in king scallop Pecten maximus leading to extensive fishery closures. Approximately 59% of the total value of all fish and shellfish landed in the Isle of Man in 2004 comprised king scallop, hence the economy of the Manx marine sector is particularly susceptible to impacts from this biotoxin. Scallop from fishing grounds around the Isle of Man were sampled in October 2003, June 2004 and October 2004 to determine levels of inter-animal and spatial variability in DA concentration and factors that might influence toxin concentration such as scallop size and water depth. Mean DA concentrations in hepatopancreas ranged from 296.3 μg g−1 to below the detection limit, in gonad from 27.8 μg g−1 to below the limit of detection and in adductor muscle from 7.3 μg g−1 to below the limit of detection. High levels of inter-animal variability of DA concentration in hepatopancreas were recorded; CVs ranging from 16.1% to 70.0%. DA concentrations above 20 μg g−1 were recorded in gonads on all three sampling dates. Scallops from fishing grounds on the east of the Isle of Man were significantly less contaminated than those from the west and southwest. A significant positive correlation between DA concentration and shell length was recorded in some sites, but there was no relationship with water depth. The high inter-animal, spatial and seasonal variability in toxin concentration highlighted the importance of understanding field variability for the development of reliable sampling and management protocols.  相似文献

‘Umbonal sculptures’ of freshwater mussels (Unionida), which ornament the early ontogenetic shell, have long been used for species identification. Specificity of these sculptures to higher taxonomic levels and their value for phylogenetic reconstruction are still under considerable scientific debate. In particular, the distribution of beak sculpture morphotypes across the unionoid phylogeny and, consequently, evolution of this character remain poorly understood. Based on an examination of 187 taxa, covering five of the six extant unionoid families, this study presents a new model of character evolution of umbonal sculptures in the order. Ten morphotypes were recognized and conceptualized into the cladistic characters sculpture presence and category. Optimization of sculpture presence on two recent hypotheses of palaeoheterodont phylogenetic relationships using the program Mesquite indicates a sculptured common ancestor of the extant Unionida, with multiple losses of the umbonal ornament occurring subsequently within the clade. Reconstruction of changes in sculpture category is ambiguous and demonstrates the need for further research into the evolutionary relationships of freshwater mussels in general and of their early ontogenetic sculptures in particular. Ambiguity is reduced in analyses applying a model with unequal costs of transformation between character states, which was derived from observations on intermediate forms and polymorphisms. These analyses suggest ‘V‐shaped’ or ‘nodulous’ sculpture as the plesiomorphic sculptural category for Unionida. The relatively low levels of homoplasy inferred for V‐shaped, pseudo‐radial and double‐looped sculptures suggest that these types may comprise useful guides to relationships within Unionida. The high degree of homoplasy of W‐shaped, pseudo‐concentric, wrinkled and single‐looped sculptures, on the other hand, renders these sculpture types less fit for such purposes.  相似文献

The steroidogenic acute regulatory protein (StAR) is responsible for acute control of cholesterol transport across the mitochondrial membrane, however the mechanism of StAR-associated cholesterol transport is unknown and may involve the peripheral-type benzodiazepine receptor (PBR)/endozepine system. Several molecules of PBR may associate to form a channel through which cholesterol passes to the inner mitochondrial membrane, and endozepine is the natural ligand for PBR. Bioluminescence resonance energy transfer (BRET) was used to test StAR/PBR/endozepine interactions, PBR aggregation, and the effect of second messengers on interactions. There was no evidence of StAR/PBR, StAR/endozepine, or PBR/endozepine interactions. The StAR and PBR fusion proteins were trafficking to the mitochondria as expected, but the endozepine fusion protein was not localized to the mitochondria indicating that it was not biologically active. Data were obtained indicating that PBR forms aggregates in the mitochondrial membrane. Energy transfer between PBR fusion proteins was dose and time dependent, but there was no effect induced by PK11195 ligand binding or pharmacologic activation of PKA or PKC second messenger pathways. It appears that PBR aggregates in the mitochondrial membrane, however there was no evidence that PBR aggregation is regulated in the acute control of steroidogenesis, or that PBR and StAR interact.  相似文献

The eukaryotic nicotinamide riboside kinase (Nrk) pathway, which is induced in response to nerve damage and promotes replicative life span in yeast, converts nicotinamide riboside to nicotinamide adenine dinucleotide (NAD+) by phosphorylation and adenylylation. Crystal structures of human Nrk1 bound to nucleoside and nucleotide substrates and products revealed an enzyme structurally similar to Rossmann fold metabolite kinases and allowed the identification of active site residues, which were shown to be essential for human Nrk1 and Nrk2 activity in vivo. Although the structures account for the 500-fold discrimination between nicotinamide riboside and pyrimidine nucleosides, no enzyme feature was identified to recognize the distinctive carboxamide group of nicotinamide riboside. Indeed, nicotinic acid riboside is a specific substrate of human Nrk enzymes and is utilized in yeast in a novel biosynthetic pathway that depends on Nrk and NAD+ synthetase. Additionally, nicotinic acid riboside is utilized in vivo by Urh1, Pnp1, and Preiss-Handler salvage. Thus, crystal structures of Nrk1 led to the identification of new pathways to NAD+.  相似文献

The term freshwater bivalve is very inclusive and not very informative. There are representatives of at least 19 families that have at least one representative living in freshwater. This suggests at least 14 different invasions of freshwater. At least nine families have small to large radiations in the freshwater environment: Corbiculidae, Sphaeriidae, Dreissenidae, and the unioniforme families: Hyriidae, Margaritiferidae, Unionidae, Etheriidae, Iridinidae, and Mycetopodidae. The unioniforme families contain at least 180 genera and about 800 species. This order is characterized by the unique parasitic larval stage on the gills, fins or the body of a particular host fish. This order of freshwater bivalves is suffering a very high rate of extinction, with about 37 species considered presumed extinct in North America alone.
