Sea urchins are noted for the absence of neoplastic disease and represent a novel model to investigate cellular and systemic cancer protection mechanisms. Following intracoelomic injection of the DNA alkylating agent methyl methanesulfonate, DNA damage was detected in sea urchin cells and tissues (coelomocytes, muscle, oesophagus, ampullae and gonad) by the alkaline unwinding, fast micromethod. Gene expression analyses of the coelomocytes indicated upregulation of innate immune markers, including genes involved in NF-κB signalling. Results suggest that activation of the innate immune system following DNA damage may contribute to the naturally occurring resistance to neoplastic disease observed in sea urchins. 相似文献

80 mongrel rats were studied for peculiarities of the heart activity regulation in dehydration, conditions of the vegetative homeostasis being different. Data of the variation pulsometry were used. A sympathetic shift of the vegetative homeostasis was a common dehydration-caused response. The shift was significant in rats with an initial equilibrium of the vegetative homeostasis and prevailing parasympathetic effects. The survival rate of this group of rats was high. In the group of rats with the initial prevalence of a sympathetic tonus a short-term sympathetic shift was replaced by an increase of parasympathetic effects. The survival rate of this group was much lower. Therefore, rats with initial prevalence of the sympathetic compartment tonus of the vegetative nervous system are more labile to the effect of the dehydration stress. 相似文献

A model for eukaryotic DNA organization has been proposed in which DNA regulatory processes depend on multiple site-specific DNA-nuclear matrix interactions throughout a DNA domain. In this model gene regulation depends on combinations of a few control factors in a cell to activate cell type-specific genes. This model suggests simple molecular mechanisms for organismal development which can account for sequential activation of appropriate groups of genes throughout development and for specific constraints on developmental pathways. Additionally, these suggested developmental pathways are consistent with mechanisms of evolution in which gradualism and punctuated equilibrium are not exclusive of one another and are interrelated mechanisms of evolution that are both induced by specific chromosomal mutations. 相似文献

Mild, non-noxious, oscillating pinches to a rat's tail elicits hyperphagia. The present study examined whether tail-pinch (TP) would exert hyperalgesic and hyperactive effects in rats that also exhibit the overeating response. The first experiment assessed TP effects upon reactivity to electric shock as measured by flinch-jump thresholds. Significant decreases in jump thresholds were observed 0 and 15, but not 30, min following TP. This effect persisted regardless of whether food was present or absent during TP. The second experiment assessed TP effects upon reactivity to heat as measured by hot-plate latencies. In contrast to jump thresholds, the shortened hot-plate latencies observed following TP persisted into the recovery period. In examining TP effects upon activity levels (Experiment 3), it was found that animals display similar patterns of temporally-declining activity regardless of whether TP was administered or not. Finally, TP selectively decreased the analgesic responses to two different doses of morphine and two different cold-water swim temperatures (Experiment 4). The TP-induced reductions occurred when TP was administered either before or after the analgesic manipulation. These data are discussed in terms of the nociceptive selectivity of the TP effect, and its influences upon analgesic processes. 相似文献

The aim of the present experiments was to clarify the subunit stoichiometry of P2X2/3 and P2X2/6 receptors, where the same subunit (P2X2) forms a receptor with two different partners (P2X3 or P2X6). For this purpose, four non-functional Ala mutants of the P2X2, P2X3, and P2X6 subunits were generated by replacing single, homologous amino acids particularly important for agonist binding. Co-expression of these mutants in HEK293 cells to yield the P2X2 WT/P2X3 mutant or P2X2 mutant/P2X3 WT receptors resulted in a selective blockade of agonist responses in the former combination only. In contrast, of the P2X2 WT/P2X6 mutant and P2X2 mutant/P2X6 WT receptors, only the latter combination failed to respond to agonists. The effects of α,β-methylene-ATP and 2-methylthio-ATP were determined by measuring transmembrane currents by the patch clamp technique and intracellular Ca(2+) transients by the Ca(2+)-imaging method. Protein labeling, purification, and PAGE confirmed the assembly and surface trafficking of the investigated WT and WT/mutant combinations in Xenopus laevis oocytes. In conclusion, both electrophysiological and biochemical investigations uniformly indicate that one subunit of P2X2 and two subunits of P2X3 form P2X2/3 heteromeric receptors, whereas two subunits of P2X2 and one subunit of P2X6 constitute P2X2/6 receptors. Electronic stun devices (ESD) often used in law enforcement, military action or self defense can induce total body uncoordinated muscular activity, also known as electromuscular incapacitation (EMI). During EMI the subject is unable to perform purposeful or coordinated movements. The mechanism of EMI induction has not been reported, but has been generally thought to be direct muscle and nerve excitation from the fields generated by ESDs. To determine the neuromuscular mechanisms linking ESD to induction of EMI, we investigated EMI responses using an anesthetized pig model. We found that EMI responses to ESD application can best be simulated by simultaneous stimulation of motor and sensory peripheral nerves. We also found that application of local anesthetics limited the response of ESD to local muscle stimulation and abolished the total body EMI response. Stimulation of the pure sensory peripheral nerves or nerves that are primarily motor nerves induced muscle responses that are consistent with well defined spinal reflexes. These findings suggest that the mechanism of ESD‐induced EMI is mediated by excitation of multiple simultaneous spinal reflexes. Although direct motor‐neuron stimulation in the region of ESD contact may significantly add to motor reactions from ESD stimulation, multiple spinal reflexes appear to be a major, and probably the dominant mechanism in observed motor response.
