Three types of recombinant human immunodeficiency virus have been found to circulate in Russia. The first type of the virus belongs to genotype gagAenvB, the second type belongs to genotype gagDenvG and the third type, to genotype gagAenvE. The virus of genotype gagAenvB circUlates in the population of drug addicts simultaneously with its "parent" viruses of genotypes gagAenvA and gagBenvB. The recombinant variant gagDenvG has African origin. The recombinant variant gagAenvE has probably been imported to Russia from South-East Asia.  相似文献

Increased Level of β-Amyloid in the Brain of Bulbectomized Mice   总被引:1,自引:0,他引:1  
Six weeks after bilateral olfactory bulbectomy, a peptide with molecular weight of 4 kD was revealed in extracts of the neocortex and hippocampus from mice. Using monoclonal antibodies 4G8, this peptide was identified as beta-amyloid. Its level was significantly higher in the bulbectomized animals than in sham-operated mice. The bulbectomized mice displayed sharp impairment in spatial memory when tested in the Morris water maze. The results suggest that bulbectomy initiates in the brain a pathological process similar to human Alzheimer's disease in location, biochemistry, and behavioral manifestations.  相似文献

Catalytic properties of mutant 23 S ribosomes resistant to oxazolidinones   总被引:4,自引:0,他引:4  
Kinetic analysis of ribosomal peptidyltransferase activity in a methanolic puromycin reaction with wild type and drug-resistant 23 S RNA mutants was used to probe the structural basis of catalysis and mechanism of resistance to antibiotics. 23 S RNA mutants G2032A and G2447A are resistant to oxazolidinones both in vitro and in vivo with the latter displaying a 5-fold increase in the value of Km for initiator tRNA and a 100-fold decrease in Vmax in puromycin reaction. Comparison of the Ki values for oxazolidinones, chloramphenicol, and sparsomycin revealed partial cross-resistance between oxazolidinones and chloramphenicol; no cross-resistance was observed with sparsomycin, a known inhibitor of the peptidyltransferase A-site. Inhibition of the mutants using a truncated CCA-Phe-X-Biotin fragment as a P-site substrate is similar to that observed with the intact initiator tRNA, indicating that the inhibition is substrate-independent and that the peptidyltransferase itself is the oxazolidinone target. Mapping of all known mutations that confer resistance to these drugs onto the spatial structure of the 50 S ribosomal subunit allows for docking of an oxazolidinone into a proposed binding pocket. The model suggests that oxazolidinones bind between the P- and A-loops, partially overlapping with the peptidyltransferase P-site. Thus, kinetic, mutagenesis, and structural data suggest that oxazolidinones interfere with initiator fMet-tRNA binding to the P-site of the ribosomal peptidyltransferase center.  相似文献

The site-specific deletion in plasmid pBR322   总被引:3,自引:0,他引:3  
The formation of a deletion derivative of plasmid pBR322, designated pBR322 delta 1, was observed during cloning of various eukaryotic DNAs, when the BamHI site of the plasmid vector was used for construction of the recombinant molecules. The restriction analysis of six independently isolated pBR322 delta 1 plasmids allowed establishment of their complete identity. Similar deletion derivatives were also formed as a result of transformation of Escherichia coli cells by the linear form of vector pBR322 produced by BamHI cleavage, but not by SalI or HindIII. The endpoints of the deletion in one of the pBR322 delta 1 plasmids occurred at positions 375 and 16666 bp from the EcoRI site, as determined by sequence analysis. Formation of pBR322 delta 1 is most probably due to site-specific recombination between the sequence in the 1666-1670 bp region and the BamHI end of the linear pBR322 molecule. THe deletion was not controlled by the recA system of the host bacteria.  相似文献

Methods of cardiomyocyte nuclei isolation from the myocard homogeneous mixture, and of cardiomyocyte genome activity estimation were elaborated. In the experiments with hyperlipoproteidemic rats, cardiomyocyte genome activity was shown to reflect the primary adaptive changes in the myocard, and to serve a reliable index of their influence on the CNS regulatory centres exposed to electromagnetic field, which is used for hyperlipoproteidemia treatment. The cardiomyocyte genome activity was used to distinguish between three types of development of adaptive reactions in the myocard.  相似文献
