Inhaled nitric oxide (iNO) is used to treat a number of disease processes. Although in vitro data suggest that nitric oxide (NO) alters surfactant protein gene expression, the effects in vivo have not been studied. The objective of this study was to evaluate the effects of iNO on surfactant protein (SP)-A, -B, and -C gene expression in the intact lamb. Thirteen 4-wk-old lambs were mechanically ventilated with 21% oxygen and received iNO at 40 ppm (n = 7) or vehicle gas (n = 6) for 24 h. Peripheral lung biopsies were obtained at 0, 12, and 24 h and analyzed for surfactant mRNA, protein, and total DNA content. Inhaled NO increased SP-A and SP-B mRNA content by 80% from 0 to 12 h and by 78 and 71%, respectively, from 0 to 24 h. There was an increase in SP-A and SP-B protein content by 45% from 0 to 12 h, and a decrease by 70 and 65%, respectively, from 0 to 24 h. DNA content was unchanged. The mechanisms and physiological effects of these findings warrant further investigation.  相似文献

A double-blind randomized phase I trial was conducted in human immunodeficiency virus type 1 (HIV-1)-negative subjects receiving vaccines vectored by plasmid DNA and modified vaccinia virus Ankara (MVA) expressing HIV-1 p24/p17 gag linked to a string of CD8(+) T-cell epitopes. The trial had two groups. One group received either two doses of MVA.HIVA (2x MVA.HIVA) (n=8) or two doses of placebo (2x placebo) (n=4). The second group received 2x pTHr.HIVA followed by one dose of MVA.HIVA (n=8) or 3x placebo (n=4). In the pTHr.HIVA-MVA.HIVA group, HIV-1-specific T-cell responses peaked 1 week after MVA.HIVA vaccination in both ex vivo gamma interferon (IFN-gamma) ELISPOT (group mean, 210 spot-forming cells/10(6) cells) and proliferation (group mean stimulation index, 37), with assays detecting positive responses in four out of eight and five out of eight subjects, respectively. No HIV-1-specific T-cell responses were detected in either assay in the 2x MVA.HIVA group or subjects receiving placebo. Using a highly sensitive and reproducible cultured IFN-gamma ELISPOT assay, positive responses mainly mediated by CD4(+) T cells were detected in eight out of eight vaccinees in the pTHr.HIVA-MVA.HIVA group and four out of eight vaccinees in the 2x MVA.HIVA group. Importantly, no false-positive responses were detected in the eight subjects receiving placebo. Of the 12 responders, 11 developed responses to previously identified immunodominant CD4(+) T-cell epitopes, with 6 volunteers having responses to more than one epitope. Five out of 12 responders also developed CD8(+) T-cell responses to the epitope string. Induced T cells produced a variety of anti-viral cytokines, including tumor necrosis factor alpha and macrophage inflammatory protein 1 beta. These data demonstrate that prime-boost vaccination with recombinant DNA and MVA vectors can induce multifunctional HIV-1-specific T cells in the majority of vaccinees.  相似文献

Daily community rates of calcification, photosynthesis and respiration were measured on a coral reef located in the Northern Red Sea, Gulf of Eilat, Israel between March 2000 and March 2002. This reef is exposed to seasonally varying levels of inorganic nutrient loading due to mixing and stratification of the adjacent open sea water column. Net production measurements were positively and linearly correlated with open sea nutrient levels, and the community photosynthesis to respiration ratio varied between 0.9 and 1.7 accordingly. Community calcification varied between 30 ± 20 and 60 ± 20 mmol C m−2 day−1 during summer and winter, respectively. Under increased nutrient loading the relation between community calcification and aragonite saturation state is suppressed by 30% on average. Both of these findings demonstrate the deleterious effects of nutrient loading on coral reefs.  相似文献

Helminth infection may be protective against allergy and account for the low prevalence of allergy in developing countries. We studied prospectively the prevalence of allergy in Ethiopian immigrants with heavy helminth infection on arrival in Israel, and again after a year of adjustment to an urban industrialized setting, to explore the roles of helminth infection, changed environment and background immunity on the manifestations of allergy. 126 newly arrived Ethiopian immigrants were studied at baseline and 115 after a year of follow up in Israel. Allergic symptoms, Skin prick tests (SPT), Tuberculin (PPD) skin tests, stool and blood samples were obtained for determining parasites, blood IgE and eosinophil levels, respectively. Anti-helminthic therapy was offered to the entire infected individuals, but only 50/108 (46.3%) took the medication. At baseline, there was a significant negative association between helminth infection and allergy, 4/18 (22.2%) of uninfected participants were allergic compared to 7/108 (6.5%) of helminth-infected participants (p = 0.028), as well as between helminth infection and SPT reactivity, 12/18 (66.6%) of uninfected participants compared to 43/108 (39.8%) of helminth-infected participants (p = 0.033). After one year, a significant general increase in allergy and SPT was observed. While only 11/126 (8.7%) were allergic at baseline, 30/115 (26.1%) became allergic at follow-up (p<0.0001), and while 55/126 (43.7%) were SPT+ at baseline, 79/115 (68.7%) became SPT+ at follow-up (p<0.001). A twofold increase in allergen sensitization was also observed after one year in Israel, particularly for dust mites, grasses and olive tree (p<0.001). These results show that: a) Helminth infection is significantly associated with low allergy and low SPT reactivity; b) One year after immigration to Israel, allergy and SPT reactivity increased significantly in all immigrants; c) Higher increases in positive SPT and allergy were observed after a year in the group that remained infected with helminths, even though they had a lowered helminth load; d) The reasons for the increased allergy one year after immigration needs further investigation but probably reflects the combined influence of the decreased helminth load and novel environmental factors.  相似文献

To improve the effectiveness of the sterile insect technique against the Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), our objectives in this study were two‐fold. First, to evaluate the ability of sterile males of the Vienna‐8 strain to survive starvation, we compared them to wild males under laboratory conditions. The second objective was to determine the effect of protein‐rich nutrition on sterile male fly survival, under starvation conditions in the laboratory, under semi‐natural conditions in a field enclosure, and under natural conditions in the open field. Therefore, we released marked sterile flies of the two diet regimes, protein‐fed or protein‐deprived, and monitored their survival by recapturing them after 4, 6, and 7 days. In the laboratory, wild males endured starvation significantly better than sterile ones and protein addition to sterile fly diet resulted in even greater reduced capability to endure starvation. On the other hand, the addition of protein to sterile‐male diet did not affect their ability to survive in a field enclosure or in the open field. We conclude that under natural conditions, where food is available, sterile male fly survival is unaffected by protein‐rich pre‐release diet.  相似文献

Studies on feeding ecology of fishes are important for understanding ecosystem structure and function. This study tested the hypothesis of diet and niche breath variation in the marbled parrotfish (Leptoscarus vaigiensis) among coral reefs of different protection levels in Kenya. Fish samples were obtained from protected (Malindi and Watamu marine parks), moderately fished (Malindi and Watamu marine reserves) and highly fished (Vipingo and Kanamai) reefs. Total lengths of fish samples were measured and their stomach contents quantified using the point method. Seasonal dietary composition, niche breaths and feeding intensities were compared between the sites using multivariate statistics. Results showed the parrotfish is a predominantly reef macroalgal grazer. Fish from protected sites fed on diverse dietary items compared to those from reserves and highly fished sites. Fish niche breadths differed between sites and seasons. Higher niche breadths occurred in protected sites during the north‐east monsoon, while higher values occurred at fished sites during the south‐east monsoon season. This study, the first of its kind in Kenya and most of the western Indian Ocean, describes feeding in the marbled parrotfish and spatial variation in niche breadth as influenced by fishing pressure, environmental variability and biological interactions.  相似文献

On a global scale, invasive grasses threaten biodiversity and ecosystem function. Nevertheless, the importation of forage grasses is a significant economic force driven by globalization. Pastureland and rangeland are of critical economic and ecological importance, but novel grass species may lead to invasion. Recognizing that economically important species can also be ecologically damaging creates a contentious debate for land managers, policymakers, and ecologists alike. Many Afrotropical perennial grass species have been intentionally introduced pantropically given their high forage production and resistance to stress. However, these traits may also confer competitive ability, increasing the possibility of unintended escape and invasion. Further, these traits have posed challenges for traditional control methods using chemicals, prescribed fire, and mowing. The use of classic biological control may alleviate the ecological impact in invaded areas. In this literature synthesis we examine Guinea grass (Megathyrsus maximus); whose economic value in many countries is undeniable, yet its impact on native ecosystems is a mounting concern. First, we introduce Guinea grass taxonomy, general biology and ecology, and the geographic and genetic origins. Second, we review the economic value and the ecological impacts. Third, we review the control of Guinea grass in undesired areas using chemical and mechanical means. Finally, we review current efforts to use biological control.
