We investigated the gene and protein expressions of V-type ATPase protein subunit C1 (ATP6V1C1) in cases of oral squamous cell carcinoma (OSCC) and contralateral normal mucosa in smokers, nonsmokers and former smokers. Subjects were separated into five groups of 15: group 1, smokers with OSCC; group 2, normal contralateral mucosa of OSCC patients; group 3, chronic smokers; group 4, former smokers who had stopped smoking 1 year earlier; group 5, individuals who had never smoked. Exfoliative cytology specimens from oral mucosa of smokers, former smokers and nonsmokers showed normal gene and protein expression. We found significantly greater gene expression in the OSCC group than in the nonsmoker groups. No difference in gene expression was observed between normal contralateral mucosa and nonsmoker groups, smoker and nonsmoker groups or former smoker and nonsmoker groups. We observed intense immunostaining for ATP6V1C1 protein in all cases of OSCC and weak or no staining in smoker, former smoker and nonsmoker groups. Significantly greater expression of ATP6V1C1 protein was observed in the OSCC group compared to the other groups, which supports the role of ATP6V1C1 in effecting changes associated with oral cancer. Analysis of the mucosae of chronic smokers, former smokers and the normal contralateral mucosa of patients with OSCC showed unaltered ATP6V1C1 gene and protein expression. Early stages of carcinogenesis, represented by altered epithelium of chronic smokers, had neither gene nor protein alterations as seen in OSCC. Therefore, we infer that the changes in ATP6V1C1 occur during later stages of carcinogenesis. Our preliminary study provides a basis for future studies of using ATP6V1C1 levels for detecting early stage OSCC.  相似文献

We studied the rotifer community of a large reservoir which was completely emptied for repairs. The goal was to find out whether such a great disturbance would affect the rotifers. We compared two periods: (1) the time previous to emptying, and (2) the time of actual refilling plus the time with water level already considered as normal. Major modifications were found only during refilling, i.e. at the beginning of the second period. At the end of the second period the community already looked much as in the first period. We conclude that the rotifer community was not significantly affected by emptying of the reservoir.  相似文献

The number of fish habitat improvement schemes has greatly increased in response to the widespread degradation of aquatic ecosystems. However, many of these enhancement projects often fail to create expected habitat conditions, because they are rarely planned and executed with inputs from the species’ habitat requirements throughout their life histories, and it is frequently assumed that the implementation of a specific instream structure for habitat improvement will always benefit all species present. Using a 2D hydraulic model, the present study evaluates the potential habitat improvement for two critically endangered fish species—the Southwestern arched-mouth nase Iberochondrostoma almacai and the Arade chub Squalius aradensis—resulting from simulating the introduction of different instream structures (islands, lateral bays, and deflectors) in a modified Mediterranean river reach. The introduction of islands in the river channel was found to be the best improvement measure for YOY and juvenile nase and chub, as shown by increases in the mean annual Weighted Usable Area (WUA) of more than 100 and 50%, respectively, compared to the present conditions. On the other hand, the simulation with current deflectors proved to be the worst scenario, particularly for nase, with mean annual WUA decreasing by 3.6, 17.8 and 22.7% for YOY, juveniles and adults, respectively. The findings of this study point to the need to account for different species life-history stages when modelling the implementation of instream structures for habitat improvement and also provide a sound basis for future conservation-related studies conducted in Mediterranean rivers that harbour other threatened “sister” species.  相似文献

We studied recycling of phosphate by enzymatic hydrolysis in two temperate very eutrophic reservoirs. To assess the potential importance of phosphate regeneration by alkaline phosphatase, we determined the activity of this enzyme in lake water concomitantly with the determinations of the concentrations of phosphomonoesters, soluble reactive phosphate, total soluble phosphate and total phosphate. Contrary to our expectations for such productive waters where algal blooms are frequent, during the study period this process of phosphate regeneration was not significant, probably because the product of hydrolysis (contained in the soluble reactive phosphate fraction) was always abundant. We conclude that, in spite of what has been observed repeatedly in natural lakes with similar trophic characteristics, the readily available fraction of phosphate in these reservoirs is large and for that reason alkaline phosphatase production is low. Therefore hydrolysis by this enzyme is not significant for growth. What seems intriguing is the small amount of phosphomonoesters found in the water; with no phosphatase activity this phosphate fraction should always be high, unless hydrolysis takes place either during phosphomonoester release or later due to their instability.  相似文献

5-Aminolevulinic acid (ALA), a precursor of porphyrin synthesis, increased the production of various porphyrin compounds in Candida guilliermondii cells. Metalloporphyrins and protoporphyrin IX (PPIX) were predominantly accumulated, respectively, at ALA concentrations of 0.2-0.4 mM and at those higher than 1.5 mM. 2,2;-Dipyridyl which complexed with bivalent metals significantly increased the content of endogenous PPIX even at ALA concentrations lower than 0.5 mM. Under these conditions, the yeast sensitivity to photodynamic effect of visible light (400-600 nm) dramatically increased due to photosensitization by endogenous PPIX.  相似文献
