Maize, sorghum, sugarcane, switchgrass and miscanthus are the main crops suggested as potential sources of lignocellulosic biomass for the production of second‐generation ethanol. The attention these crops have received has been concentrated in the field of genomics, and very little research has been performed in the field of proteomics, particularly in the cell wall proteomic, despite the importance of these crops in biofuel production. New mass spectrometry‐based proteomic methods allow the identification and quantification of thousands of proteins in complex mixtures, as well as the detection of post‐translational changes in complex proteomes, providing important insight into the downstream consequences of gene expression. Together with other ‘omic’ approaches, proteomic might be decisive to bring new information in the study of cell wall formation. Here, we briefly highlight proteomic techniques and review the research that has been completed on the proteomes of these five crops.  相似文献

Background and aims

Gaseous losses of ammonia (NH3) have been observed in citrus orchards when urea is surface-applied to the soils, and this loss might significantly limit the effectiveness of the nitrogen (N) fertilizer. However, a portion of the volatilized NH3 might be absorbed by the plants through the leaves. To quantify the contribution of the leaf absorption of 15NH3, a study with sweet oranges was conducted in two field areas where trees were grown at standard (480 trees ha?1) and high densities (617 trees ha?1).

Methods

Plastic trays were filled with soil, covered with mown grass to simulate field management conditions, fertilized with 15N labeled urea (12 atom % excess) and placed under each of three trees in the orchards. This experimental procedure prevented the uptake of N from the labeled urea by the roots. Two weeks after 15N fertilization, the trays were removed from the field, and the soil was homogenized and sampled for chemical analyses. The citrus trees under which the trays were placed were destructively harvested, and the total N concentrations and 15N/14N ratios were determined.

Results

After urea application, the NH3 losses peaked within three days and subsequently decreased to negligible amounts after 10 days. The total NH3 losses accounted for 55–82 % of the applied N. Although the NH3 absorption by the citrus leaves was proportional to the tree density in the field, only 3–7 % of the 15NH3 volatilized from the soil was recovered by the citrus trees, and the NH3 absorption was also influenced by the proximity of citrus trees to the site of urea application and the leaf areas of the trees.

Conclusions

The citrus trees can absorb the NH3 volatilized from urea, even though, the amount recovered by the trees is small and does not represent a significant proportion of total gaseous N losses, what demonstrates the importance of enhanced N use efficiency practices in field to reduce losses of NH3 when urea is applied to soil surfaces.  相似文献

Abstract:  Substrate distribution was studied in three adult colonies of the leaf-cutting ant Atta bisphaerica Forel, 1908 using dye. Some supply holes were mapped using baits made from small plastic straws impregnated with citric pulp. Two holes, equidistant from one another, were then selected in each nest. The colonies were excavated completely 24 h after bait placement. During excavation, fungus chambers were checked for dye. We observed that the dyed baits were distributed in all sectors and at all depths regardless of where the baits had been placed. This supported the hypothesis that toxic baits placed in a single supply hole are uniformly distributed throughout the colony.  相似文献

Abstract:  This work was carried out to determine the internal and external structures of Atta bisphaerica (Forel) nests. Six nests were excavated and during excavation, all data referring to chambers and tunnels were recorded. Three nests had been internally cement-moulded, which enabled a better view of the chamber and tunnel structures. Atta bisphaerica nests presented a similar structural pattern, varying only in the number of chambers as a function of external mound area. Chambers were spherical with two communication tunnels. Internal tunnels had an elliptical section, sometimes circular, indirectly linked to chambers through ramifications and directly through short tunnels. Entrance holes were linked to the area of highest chamber concentration by tunnels from the elliptical section, which led to the nest in a radial manner. Knowledge of the colony's three-dimensional architecture permits successful application of chemical control processes, reduces the quantity of product applied, and consequently diminishes costs and environmental damage.  相似文献

The origins of clot rheological behavior associated with network morphology and factor XIIIa-induced cross-linking were studied in fibrin clots. Network morphology was manipulated by varying the concentrations of fibrinogen, thrombin, and calcium ion, and cross-linking was controlled by a synthetic, active-center inhibitor of FXIIIa. Quantitative measurements of network features (fiber lengths, fiber diameters, and fiber and branching densities) were made by analyzing computerized three-dimensional models constructed from stereo pairs of scanning electron micrographs. Large fiber diameters and lengths were established only when branching was minimal, and increases in fiber length were generally associated with increases in fiber diameter. Junctions at which three fibers joined were the dominant branchpoint type. Viscoelastic properties of the clots were measured with a rheometer and were correlated with structural features of the networks. At constant fibrinogen but varying thrombin and calcium concentrations, maximal rigidities were established in samples (both cross-linked and noncross-linked) which displayed a balance between large fiber sizes and great branching. Clot rigidity was also enhanced by increasing fiber and branchpoint densities at greater fibrinogen concentrations. Network morphology is only minimally altered by the FXIIIa-catalyzed cross-linking reaction, which seems to augment clot rigidity most likely by the stiffening of existing fibers.  相似文献

The photochemistry of the diphosphino Pt(II) hydrides [LPtH2] (L=(t-Bu)2P(CH2)2P(t-Bu)2 (7); L=(t-Bu)2P(CH2)3P(t-Bu)2 (8);L=(t-Bu)(Ph)P(CH2)2P(Ph)(t-Bu) (9)) is reported. The primary photoevent is the dissociation of H2 and formation of the 14-e [LPt] species. These coordinatively unsaturated intermediates provide a versatile entry point into the C---H bond activation of hydrocarbons. [LPt] reacts with benzene in an oxidative addition reaction to yield [LPt(H)(C6H5)] complexes. The importance of the metal centre and ancillary ligation in the C---H bond activation is discussed.  相似文献

Cortical circuits generate excitatory currents that must be cancelled by strong inhibition to assure stability. The resulting excitatory-inhibitory (E-I) balance can generate spontaneous irregular activity but, in standard balanced E-I models, this requires that an extremely strong feedforward bias current be included along with the recurrent excitation and inhibition. The absence of experimental evidence for such large bias currents inspired us to examine an alternative regime that exhibits asynchronous activity without requiring unrealistically large feedforward input. In these networks, irregular spontaneous activity is supported by a continually changing sparse set of neurons. To support this activity, synaptic strengths must be drawn from high-variance distributions. Unlike standard balanced networks, these sparse balance networks exhibit robust nonlinear responses to uniform inputs and non-Gaussian input statistics. Interestingly, the speed, not the size, of synaptic fluctuations dictates the degree of sparsity in the model. In addition to simulations, we provide a mean-field analysis to illustrate the properties of these networks.  相似文献

Galectins, beta-galactoside-binding lectins, are extensively distributed in the animal kingdom and share some basic molecular properties. Galectin-3, a member of this family, is generally associated with differentiation, morphogenesis, and metastasis. In this study, galectin-3 was isolated from ovine placental cotyledons round the middle of the gestation period by lactose extraction followed by affinity chromatography on lactosyl-agarose, and separated from galectin-1 by size exclusion chromatography on a Superose 12 column. Under native conditions this lectin behaved as a monomer with an apparent molecular weight of approximately 29,000 and an isoelectric point of 9.0. The partial amino acid sequence of the peptides obtained by tryptic digestion of this protein followed by HPLC separation showed striking homology with other members of the galectin-3 subfamily. Furthermore, ovine placental galectin-3 exhibited specific mitogenic activity toward rat spleen mononuclear cells. Besides, this protein strongly reacted with a rabbit antiserum raised against a chicken galectin. Results obtained by Western blot analysis showed that its expression was greatly decreased in term placenta with respect to the middle of the gestation period, suggesting a regulated expression throughout development.   相似文献

The aim of this study was to investigate the impact of moderate aerobic training on functional, anthropometric, biochemical, and health-related quality of life (HRQOL) parameters on women with metabolic syndrome (MS). Fifteen untrained women with MS performed moderate aerobic training for 15 weeks, without modifications of dietary behaviours. Functional, anthropometric, biochemical, control diet record and HRQOL parameters were assessed before and after the training. Despite body weight maintenance, the patients presented decreases in waist circumference (P = 0.001), number of MS components (P = 0.014), total cholesterol (P = 0.049), HDL cholesterol (P = 0.004), LDL cholesterol (P = 0.027), myeloperoxidase activity (P = 0.002) and thiobarbituric acid-reactive substances levels (P = 0.006). There were no differences in total energy, carbohydrate, protein and lipid intake pre- and post-training. Furthermore, improvements in the HRQOL subscales of physical functioning (P = 0.03), role-physical (P = 0.039), bodily pain (P = 0.048), general health (P = 0.046) and social functioning scoring (P = 0.011) were reported. Despite the absence of weight loss, aerobic training induced beneficial effects on functional, anthropometric, biochemical and HRQOL parameters in women with MS.  相似文献
