The function of the N-terminal amino acids of Saccharomyces cerevisiae hexokinase II was studied in vivo using strains producing a form of hexokinase II lacking its first 15 amino acids (short form). This short form of hexokinase II was produced from a fusion between the promoter region of the PGK1 gene and the HXK2 coding sequence except the first 15 codons. As expected, the in vitro analysis of the short form protein by gel filtration chromatography indicates that the short protein does not form dimers under conditions where the wild-type protein dimerizes. Kinetic studies show that the enzymatic activities are very similar to wild-type behavior. The physiological experiments performed on the strains containing the fusion allele demonstrate that the short form of the enzyme is similar to the wild-type both in terms of phosphorylation of hexoses and glucose repression. We conclude that the N-terminal amino acids of hexokinase II are not required in vivo either for phosphorylation of hexoses or for glucose repression. 相似文献

Tomato root growth and distribution were related to inorganic nitrogen (N) availability and turnover to determine 1) if roots were located in soil zones where N supply was highest, and 2) whether roots effectively depleted soil N so that losses of inorganic N were minimized. Tomatoes were direct-seeded in an unfertilized field in Central California. A trench profile/monolith sampling method was used. Concentrations of nitrate (NO3
-) exceeded those of ammonium (NH4
+) several fold, and differences were greater at the soil surface (0–15 cm) than at lower depths (45–60 cm or 90–120 cm). Ammonium and NO3
- levels peaked in April before planting, as did mineralizable N and nitrification potential. Soon afterwards, NO3
- concentrations decreased, especially in the lower part of the profile, most likely as a result of leaching after application of irrigation water. Nitrogen pool sizes and rates of microbial processes declined gradually through the summer.Tomato plants utilized only a small percentage of the inorganic N available in the large volume of soil explored by their deep root systems; maximum daily uptake was approximately 3% of the soil pool. Root distribution, except for the zone around the taproot, was uniformly sparse (ca. 0.15 mg dry wt g-1 soil or 0.5 cm g-1 soil) throughout the soil profile regardless of depth, distance from the plant stem, or distance from the irrigation furrow. It bore no relation to N availability. Poor root development, especially in the N-rich top layer of soil, could explain low fertilizer N use by tomatoes. 相似文献

The orientational order profile along the lipid acyl chain has been characterized under several different conditions of polar headgroup composition, temperature, and cholesterol content. Despite the different nature of these factors, the variation of the order is governed by two common trends. First, the relative change of order induced by the variation of these factors is always more pronounced towards the end of the chain than for the methylene groups near the interface. Second, there is, to a first approximation, a distinct correlation between the magnitude of the order parameters and the shape of the order profile. For example when the chain is highly ordered, the relative width of the order distribution is narrow indicating that the plateau region is longer. These conclusions suggest that the orientational order profile depends on only a small number of parameters and demonstrate clearly that the correlation length for changes in orientational order is much greater than one C-C bond length. Our results also show that the reduced temperature is not related in simple terms to orientational order and probably has little theoretical significance. The orientational order profiles of POPC and POPE bilayers are significantly different even when expressed in terms of reduced temperature. The behavior of POPC/cholesterol systems also indicates that the orientational order of the lipid chain and the gel-to-liquid crystalline phase transition temperature are not related in a straightforward manner.Abbreviations POPC

The influence of the long-acting somatostatin analogue, SMS 201-995, on FMLP-induced neutrophil elastase release in vitro has been investigated. Doses from 150 ng/ml upwards inhibited elastase release, with 100% inhibition by 2500 ng/ml. Inhibition was demonstrated both by an assay measuring elastase immunometrically and by an assay based on its enzyme activity. The demonstration that SMS 201-995 inhibits protease release from polymorphonuclear leukocytes may have implications for the long-term clinical use of this somatostatin analogue. 相似文献

In order to investigate the role of protein phosphorylation in the early differentiative events of mouse preimplantation development, timed groups of embryos of various stages were incubated in medium containing [32P]orthophosphate and harvested immediately after labelling or following a chase period. The phosphoproteins obtained were separated by electrophoresis in one and two dimensions. While some of the phosphoproteins found were common to all the stages examined, the detection of many depended on both the combination of pulse-labelling and chase periods used and on the developmental stage examined. Some phosphoproteins were only found in compacted 8-cell embryos, a correlation which suggests a possible link with the post-translational mechanisms which underlie compaction. 相似文献
