In a context of growing resistance to classical antifungal therapy, the design of new drugs targeting alternative pathways is highly expected. Benzofuro[3,2-d]pyrimidine derivatives, derived from (?)-cercosporamide, were synthesized and evaluated as potential Candida albicans PKC inhibitors in the aim of restoring susceptibility to azole treatment. Co-administration assay of benzofuropyrimidinedione 23 and fluconazole highlighted a synergistic effect on inhibition of cell growth of a Candida albicans resistant strain.  相似文献

Malaria is a hazardous disease caused by Plasmodium parasites and often results in lethal complications, including malaria-associated acute respiratory distress syndrome (MA-ARDS). Parasite sequestration in the microvasculature is often observed, but its role in malaria pathogenesis and complications is still incompletely understood. We used skeleton binding protein-1 (SBP-1) KO parasites to study the role of sequestration in experimental MA-ARDS. The sequestration-deficiency of these SBP-1 KO parasites was confirmed with bioluminescence imaging and by measuring parasite accumulation in the lungs with RT-qPCR. The SBP-1 KO parasites induced similar lung pathology in the early stage of experimental MA-ARDS compared to wildtype (WT) parasites. Strikingly, the lung pathology resolved subsequently in more than 60% of the SBP-1 KO infected mice, resulting in prolonged survival despite the continuous presence of the parasite. This spontaneous disease resolution was associated with decreased inflammatory cytokine expression measured by RT-qPCR and lower expression of cytotoxic markers in pathogenic CD8+ T cells in the lungs of SBP-1 KO infected mice. These data suggest that SBP-1-mediated parasite sequestration and subsequent high parasite load are not essential for the development of experimental MA-ARDS but inhibit the resolution of the disease.  相似文献

Establishment and maintenance of transgenic mouse strains require being able to distinguish homozygous from heterozygous animals. To date, the developed real-time quantitative PCR techniques are often complicated, time-consuming and expensive. Here, we propose a very easy and rapid method with a simple data analysis to determine zygosity in transgenic mice. We show that the real-time quantitative PCR using SYBR Green fluorescent dye can be applied to discriminate two-fold differences in copy numbers of the transgene. Our procedure has to fit only three simple requirements: (1) to design primers capable of detecting one Ct difference for two-fold differences in DNA amounts (2) to measure genomic DNA concentrations accurately and (3) to have a reference animal of known zygosity in each run. Then, if the Ct values for the control gene are similar in all samples, we are able to compare directly the Ct values for the transgene in every sample, and so, to deduce the zygosity status of each mouse relative to the reference animal. This method is really simple and reliable, and it may be valuable as a rapid screening tool for zygosity status in transgenic animals.  相似文献

Characteristics of the reproductive cycle of Basilichthys microlepidotus (Jenyns), an atherinid fish living in a South American Mediterranean arid zone, are analysed. Specimens 9-170 mm in total length were captured from August 1982 to April 1984 in the Rio Choapa, in northern Chile (31%%45' S, 71°15' W). Analysis of gonadosomatic index fluctuations, and determination of the proportion of mature males and females captured during the study period, allowed the estimation of the duration of the reproductive cycle.
Females 70–170 mm in total length contained 2000–9000 intraovarian oocytes, of which approximately 80% were vitellogenic (100–1300 μm in diameter) and exhibited a polymodal distribution. A further 4 to 15% of the total oocytes, measured 1700–2000 μm in diameter and appeared ready to be spawned. In 30% of the examined ovaries some of the largest oocytes exhibited signs of degeneration, or atresia. Juveniles approximately 30 mm in total length were captured over most of the study period. Results strongly suggest that B. microlepidotus has multiple spawnings.  相似文献

Invasive fungal infection is a serious complication following allogeneic hematopoietic stem cell transplantation. Pulmonary infection due to Hormographiella aspergillata is an uncommon condition associated with a high mortality rate. The susceptibility of H. aspergillata to available antifungal agents is not well established. We report for the first time a case of H. aspergillata lung infection that responded poorly to conventional treatment with liposomal amphotericin B (LAmB; 3 mg kg?1 of body weight per day) with renal damage at higher posology (5 mg kg?1 of body weight per day), but improved rapidly after addition of nebulized LAmB to intravenous LAmB (3 mg kg?1 of body weight per day). Successful treatment of our patient using nebulized LAmB would be worth evaluating in cases refractory to standard treatment or when the reference treatment may not be extended due to interaction or side effects.  相似文献

Tight junctions (TJs) and adherens junctions (AJs) are dynamic structures linked to the actin cytoskeleton, which control the paracellular permeability of epithelial and endothelial barriers. TJs and AJs are strictly regulated in a spatio-temporal manner by a complex signaling network, including Rho/Ras-GTPases, which have a pivotal role. Rho preferentially regulates TJs by controlling the contraction of apical acto-myosin filaments, whereas Rac/Cdc42 mainly coordinate the assembly-disassembly of AJ components. However, a subtle balance of Rho/Ras-GTPase activity and interplay between these molecules is required to maintain an optimal organization and function of TJs and AJs. Conversely, integrity of intercellular junctions generates signals through Rho-GTPases, which are involved in the regulation of multiple cellular processes. Rho/Ras-GTPases and the control of intercellular junctions are the target of various bacterial toxins responsible for severe diseases in man and animals, and are part of their mechanism of action. This review focuses on the regulation of TJs and AJs by Rho/Ras-GTPases through molecular approaches and bacterial toxins.  相似文献

We explored the patterns in bacterioplankton community metabolism (BCM) and four components of community structure [composition (BCC), metabolic capacities (MC), physiological structure (PS) and single-cell characteristics (SCC)], between lakes, rivers and marshes within a watershed in Québec, to assess the connections that exist between them and with the main resources (organic matter, nutrients). Habitat types were well segregated by both resources and BCM and their corresponding dissimilarity matrices were significantly correlated, suggesting that BCM tracks resource conditions in a consistent manner across ecosystem types. MC also segregated the various habitats and was correlated to BCM but less so to resources, whereas BCC at times resulted in a clear separation of habitats, but was rarely correlated to resources and never to BCM, suggesting a higher degree of ecosystem specificity at this particular level. Finally, there was no clear separation of habitats in terms of PS and SCC, and none covaried with resources or BCM. The habitat patterns based on these different components of structure were rarely correlated to each other, indicating weak deterministic connections between them. MC appears to mediate the link between resources and BCM more directly and consistently across systems; BCC appears to be more influenced by ecosystem-specific factors that weaken its overall connection to both resources and BCM, whereas PS and SCC show no discernible patterns. Our results thus suggest that the bottom-up regulation of BCM by resources is mediated by complex shifts within components of community structure that can be directional, ecosystem-specific or apparently random, which combined nevertheless result in a systematic overall response to resources in terms of C metabolism.  相似文献

The goal of this study was to understand the mechanisms of greater weight loss by gastric bypass (GBP) compared to gastric banding (GB) surgery. Obese weight‐ and age‐matched subjects were studied before (T0), after a 12 kg weight loss (T1) by GBP (n = 11) or GB (n = 9), and at 1 year after surgery (T2). peptide YY3–36 (PYY3–36), ghrelin, glucagon‐like peptide‐1 (GLP‐1), leptin, and amylin were measured after an oral glucose challenge. At T1, glucose‐stimulated GLP‐1 and PYY levels increased significantly after GBP but not GB. Ghrelin levels did not change significantly after either surgery. In spite of equivalent weight loss, leptin and amylin decreased after GBP, but not after GB. At T2, weight loss was greater after GBP than GB (P = 0.003). GLP‐1, PYY, and amylin levels did not significantly change from T1 to T2; leptin levels continued to decrease after GBP, but not after GB at T2. Surprisingly, ghrelin area under the curve (AUC) increased 1 year after GBP (P = 0.03). These data show that, at equivalent weight loss, favorable GLP‐1 and PYY changes occur after GBP, but not GB, and could explain the difference in weight loss at 1 year. Mechanisms other than weight loss may explain changes of leptin and amylin after GBP.  相似文献
