Split‐root experiments were conducted to test the hypothesis that adjustments in lateral root initiation, as might occur in response to localized soil conditions, are determined by the sugar content of the root and do not depend on changes in the import of phloem‐translocated phytohormones. Wheat ( Triticum aesticum L. cv. Alexandria) seedlings were grown in hydroponics with their seminal roots divided between two compartments within the culture vessel. Two seminal roots of treated plants were supplied with standard nutrient solution supplemented with 50 m M glucose, whilst the remaining three roots received nutrient solution without glucose. Control plants had their roots divided in the same ratio, but both 'halves' received nutrient solution without glucose. Feeding glucose to one 'half' of the root system increased the frequency (number per unit length) of lateral root primordia in the fed axes. The increase was first observed 15 h after the start of treatment and was located within the apical 30 mm of root. At this time there was no significant treatment effect on the frequency of primordia in non‐fed axes. The enhanced initiation of lateral roots in glucose‐fed root tips was associated with an increase in their concentration of glucose and sucrose plus low molecular mass fructans. In contrast, there was a reduction in partitioning of 14C‐photosynthate to these root tips compared to the non‐fed roots of treated plants and controls. The results indicate that lateral root initiation can be stimulated by sugars in the absence of an increase in phloem translocation. It is proposed that proliferation of lateral roots in response to localized soil conditions, such as nutrient patches, may be signalled by an increase in sugar content of the tissue, rather than an altered flux of phytohormones or other material co‐transported with sucrose in the phloem.  相似文献

Our aim was to develop a biodegradable fibrous dressing to act as a tissue guide for in situ wound repair while releasing Ibuprofen to reduce inflammation in wounds and reduce pain for patients on dressing changes. Dissolving the acid form of Ibuprofen (from 1% to 10% by weight) in the same solvent as 75% polylactide, 25% polyglycolide (PLGA) polymers gave uniformly loaded electrospun fibers which gave rapid release of drug within the first 8 h and then slower release over several days. Scaffolds with 10% Ibuprofen degraded within 6 days. The Ibuprofen released from these scaffolds significantly reduced the response of fibroblasts to major pro‐inflammatory stimulators. Fibroblast attachment and proliferation on scaffolds was unaffected by the addition of 1–5% Ibuprofen. Scaffolds loaded with 10% Ibuprofen initially showed reduced cell attachment but this was restored by soaking scaffolds in media for 24 h. In summary, addition of Ibuprofen to electrospun biodegradable scaffolds can give acute protection of adjacent cells to inflammation while the scaffolds provide an open 3D fibrous network to which cells can attach and migrate. Glycosaminoglycan synthesis in endotoxin-induced lung injury   总被引:2,自引:0,他引:2  
Endotoxin-induced lung injury has previously been shown to produce lesions that resemble emphysema morphologically and biochemically as demonstrated by the reduction in the content of lung elastin. The purpose of this study was to define the changes in one other connective tissue component, glycosaminoglycans, during the acute phase of the lung injury. Intravenous administration of a single dose of endotoxin in rats resulted in an increase in the total synthesis of glycosaminoglycans by the pulmonary parenchyma. There was a significant increase in the proportion of dermatan sulfate synthesized during the first 48 hr and a concomitant decrease in heparin/heparan sulfate synthesis. At 48 hr the increased synthesis of dermatan sulfate had reached 7.3 times control values and began to decline, whereas the synthesis of chondroitin-4-sulfate rose from 4.1 to 10.7 times control values between 48 and 72 hr. Analysis of the rates of synthesis revealed that the total amount of heparin/heparan sulfate remained constant while the synthesis of chondroitin-6-sulfate increased proportionally to the overall synthesis of glycosaminoglycans. These findings indicate that dramatic changes in glycosaminoglycan synthesis are an integral part of endotoxin lung injury.  相似文献

The Myc family of oncoproteins are thought to regulate proliferation and differentiation in a wide variety of cell types. Recent studies show that Myc proteins form sequence-specific DNA-binding complexes with Max, a new member of the helix-loop-helix leucine zipper protein class. In sediments, the bioavailability and toxicity of Ni are strongly influenced by its sorption to manganese (Mn) oxides, which largely originate from the redox metabolism of microbes. However, microbes are concurrently susceptible to the toxic effects of Ni, which establishes complex interactions between toxicity and redox processes. This study measured the effect of Ni on growth, pellicle biofilm formation and oxidation of the Mn-oxidizing bacteria Pseudomonas putida GB-1. In liquid media, Ni exposure decreased the intrinsic growth rate but allowed growth to the stationary phase in all intermediate treatments. Manganese oxidation was 67% less than control for bacteria exposed to 5 μM Ni and completely ceased in all treatments above 50 μM. Pellicle biofilm development decreased exponentially with Ni concentration (maximum 92% reduction) and was replaced by planktonic growth in higher Ni treatments. In solid media assays, growth was unaffected by Ni exposure, but Mn oxidation completely ceased in treatments above 10 μM of Ni. Our results show that sublethal Ni concentrations substantially alter Mn oxidation rates and pellicle biofilm development in P. putida GB-1, which has implications for toxic metal bioavailability to the entire benthic community and the environmental consequences of metal contamination.  相似文献
