Mammographic density (MD), associated with higher water and lower fat content in the breast, is strongly related to breast cancer risk. Optical attenuation spectroscopy (OS) is a non-imaging method of evaluating breast tissue composition by red and near-infrared light transmitted through the breast that, unlike mammography, does not involve radiation. OS provides information on wavelength dependent light scattering of tissue and on absorption by water, lipid, oxy-, deoxy-hemoglobin. We propose that OS could be an alternative marker of breast cancer risk and that OS breast tissue measures will be associated with MD. In the present analysis, we developed an algorithm to estimate breast tissue composition and light scattering parameters using a spectrally constrained global fitting procedure employing a diffuse light transport model. OS measurements were obtained from 202 pre- and post-menopausal women with normal mammograms. Percent density (PD) and dense area (DA) were measured using Cumulus. The association between OS tissue composition and PD and DA was analyzed using linear regression adjusted for body mass index. Among pre-menopausal women, lipid content was significantly inversely associated with square root transformed PD (β = -0.05, p = 0.0002) and DA (β = -0.05, p = 0.019); water content was significantly positively associated with PD (β = 0.06, p = 0.008). Tissue oxygen saturation was marginally inversely associated with PD (β = -0.03, p = 0.057) but significantly inversely associated with DA (β = -0.10, p = 0.002). Among post-menopausal women lipid and water content were significantly associated (negatively and positively, respectively) with PD (βlipid = -0.08, βwater = 0.14, both p<0.0001) and DA (βlipid = -0.10, p<0.0001; βwater = 0.11, p = 0.001). The association between OS breast content and PD and DA is consistent with more proliferation in dense tissue of younger women, greater lipid content in low density tissue and higher water content in high density tissue. OS may be useful for assessing physiologic tissue differences related to breast cancer risk, particularly when mammography is not feasible or easily accessible.  相似文献

Summary Infestations of the phloem feeding coccid Icerya seychellarum on the natural vegetation of Aldabra atoll are attended at night by large numbers of the large ant Componotus maculatus and, during the day, by small numbers of other ant species.Large ant species preferentially tended larger coccids. The efficiency of C. maculatus worker spacing among the patchily distributed Icerya resource on a plant, was dependent upon the overall ant/Icerya biomass ratio. At high ratios (i.e. low resource availability per ant) they were more evenly dispersed.Size of honeydew loads carried by C. maculatus workers declined as the night progressed, although ant numbers remained constant. From this, and the observations of more even dispersion at higher ant/Icerya biomass ratios, it is concluded that the ants are probably stimulating the coccids to produce more honeydew.The results of an ant exlusion experiment were inconclusive, but suggested that I. seychellarum can exist at reasonably high densities in the absence of ants and that numbers of Chilocorus nigritus, a diaspid scale predator, are reduced by ants.The implication of these results for the biological control of I. seychellarum on Aldabra are discussed, together with the processes involved in honeydew collection in a patchily distributed Icerya population.  相似文献

1,2-Diacylglycerol (DAG) was measured in neutral lipid extracts from isolated hepatocytes using high pressure liquid chromatography followed by refractive index detection. Maximally effective doses of epinephrine, angiotensin II, and vasopressin increased DAG by approximately 65, 80, and 180-250%, respectively, with maximal increases being observed at 8-10 min. Depletion of cellular Ca2+ resulted in a 50% decrease in DAG accumulation elicited by vasopressin. Other agents which increased DAG levels were the tumor promoter 4 beta-phorbol 12 beta-myristate 13 alpha-acetate (120% increase at 10(-6) M), the Ca2+ ionophore A23187 (385% increase at 10(-5) M), and ATP (180% increase at 1 mM). The concentration dependence of DAG accumulation in response to epinephrine, angiotensin II, and vasopressin was similar to that found for myoinositol triphosphate accumulation (Charest, R., Prpic, V., Exton, J. H., and Blackmore, P.F. (1985) Biochem. J. 227, 79-90), which was approximately 5-10 times less sensitive to hormone than was phosphorylase activation. Fatty acid analysis revealed that hormonally induced DAG was partially derived from sources other than inositol phospholipids. It is proposed from these studies that Ca2+-mobilizing hormones elicit a prolonged increase in the levels of hepatocyte DAG, which may activate protein kinase C.  相似文献

Current work in behavioral ecology contrasts traditional hypotheses of context‐specific adaptation with the suggestion that behaviors are often coupled such that animals exhibit context‐general behavioral syndromes. Wasteful killing, the partial consumption and/or abandonment of prey, is a perplexing phenomenon because the costs of this behavior seem high (e.g. energetic loss, risk of injury). Wasteful killing may be the product of a context‐specific, adaptive foraging strategy restricted spatially and/or temporally to conditions of prey abundance. Alternatively, wasteful killing may represent a context‐general syndrome of high aggression that results in the capture of prey that are not consumed. We investigated the conditions in which a web‐building spider the North American black widow (Latrodectus hesperus) participates in wasteful killing. We employed a powerful repeated‐measures design that reduces error variance by controlling for individual differences. Across the adult life cycle of female spiders collected from urban habitats throughout Phoenix, AZ, we repeatedly measured each spider’s foraging voracity and wastefulness after varied periods of food restriction (2, 7, and 14 d). While food restriction decreased body condition, condition alone was a poor predictor of voracity and wasteful killing. Our results indicate that previous food restriction i) shortens latency to attack, ii) heightens the number of prey killed per trial, and iii) reduces wastefulness. Latency to attack and wasteful killing were neither repeatable within individuals nor correlated with each other and therefore appear unlikely to be components of a behavioral syndrome. We discuss other possible benefits for wasteful killing in the contexts of predation risk and mate attraction.  相似文献

The generation of new neurons within the dentate gyrus of the mature hippocampus is critical for spatial learning, object recognition and memory, whereas new neurons born in the subventricular zone (SVZ) contribute to olfactory function. Adult neurogenesis is a multistep process that begins with the activation and proliferation of a pool of stem/precursor cells. Although the presence of self-renewing and multipotent neural precursors is well established in the SVZ, it is only recently that the existence of such a precursor population has been demonstrated in the hippocampus, the region of the brain involved in learning and memory. Determining how this normally latent pool can be activated therefore offers considerable potential for the development of targeted neurogenic-based therapeutics to ameliorate the cognitive decline associated with hippocampal dysfunction in several neurodegenerative diseases. In this review, we summarize the effects of neural activity, various molecular factors and pharmaceutical agents, as well as voluntary exercise, in activating endogenous neural precursors in the two neurogenic niches of the adult brain, and highlight the role of activation-driven enhancement of neurogenesis for the treatment of psychiatric illness and aging dementia.  相似文献

Knowing the age of lineages is key to understanding their biogeographic history. We aimed to provide the best estimate of the age of Cichorieae and its subtribes based on available fossil evidence and DNA sequences and to interpret their biogeography in the light of Earth history. With more than 1,550 species, the chicory tribe (Cichorieae, Asteraceae) is distributed predominantly in the northern Hemisphere, with centres of distribution in the Mediterranean region, central Asia, and SW North America. Recently, a new phylogenetic hypothesis of Cichorieae based on ITS sequences has been established, shedding new light on phylogenetic relationships within the tribe, which had not been detected so far. Cichorieae possess echinolophate pollen grains, on the surface of which cavities (lacunae) are separated by ridges. These lacunae and ridges show patterns characteristic of certain groups within Cichorieae. Among the fossil record of echinolophate pollen, the Cichorium intybus-type is the most frequent and also the oldest type (22 to 28.4 million years old). By using an uncorrelated relaxed molecular clock approach, the Cichorieae phylogenetic tree was calibrated with this fossil find. According to the analysis, the tribe originated no later than Oligocene. The species-rich core group originated no later than Late Oligocene or Early Miocene and its subtribes diversified no later than Middle/Late Miocene or Early Pliocene—an eventful period of changing geological setting and climate in the Mediterranean region and Eurasia. The first dispersal from Eurasia to North America, which resulted in the radiation of genera and species in North America (subtribe Microseridinae), also occurred no later than Middle or Late Miocene, suggesting the Bering land bridge as the route of dispersal.  相似文献

Accumulation of soluble recombinant hemoglobin (rHb1.1) in Escherichia coli requires proper protein folding, prosthetic group (heme) addition, and subunit assembly. This served as a new model system for the study of the effects of temperature, protein synthesis rates, and protein accumulation rates on protein solubility in E. coli. Fermentation expression of rHb1.1 at 30 degrees C from cultures containing a medium or high globin gene dosage (pBR-based or pUC-based plasmids with rHb1.1 genes under the control of the tac promoter) was compared. A medium gene dosage resulted in rHb1.1 accumulating to approximately 7% of the soluble cell protein, of which 78% was soluble. A high globin gene dosage resulted in a > or = 3-fold increase in total globin to 23 to 24% of the soluble cell protein, but 70% was insoluble. Accumulation of insoluble rHb1.1 began immediately upon induction. The proportion of rHb1.1 from the high globin gene dosage that accumulated as insoluble globin was affected by reducing (i) the inducer concentration and (ii) the temperature. Reducing the inducer concentration reduced globin synthesis up to eightfold but increased the proportion of soluble rHb1.1 to 93%. In contrast, total globin protein synthesis was barely affected by reducing the temperature from 30 to 26 degrees C, while soluble globin accumulation increased > 2-fold to approximately 15% of the soluble cell protein. The contrast between the effects of reducing rates of protein synthesis and accumulation and those of reducing temperature suggests that lower temperature stabilizes one or more folding intermediates. We propose a simplified physical model which integrates protein synthesis, folding, and heme association. This model shows that temperature-dependent apoglobin stability is the most critical factor in soluble rHb1.1 accumulation.  相似文献
