Slow‐colonizing forest understorey plants are probably not able to rapidly adjust their distribution range following large‐scale climate change. Therefore, the acclimation potential to climate change within their actual occupied habitats will likely be key for their short‐ and long‐term persistence. We combined transplant experiments along a latitudinal gradient with open‐top chambers to assess the effects of temperature on phenology, growth and reproductive performance of multiple populations of slow‐colonizing understorey plants, using the spring flowering geophytic forb Anemone nemorosa and the early summer flowering grass Milium effusum as study species. In both species, emergence time and start of flowering clearly advanced with increasing temperatures. Vegetative growth (plant height, aboveground biomass) and reproductive success (seed mass, seed germination and germinable seed output) of A. nemorosa benefited from higher temperatures. Climate warming may thus increase future competitive ability and colonization rates of this species. Apart from the effects on phenology, growth and reproductive performance of M. effusum generally decreased when transplanted southwards (e.g., plant size and number of individuals decreased towards the south) and was probably more limited by light availability in the south. Specific leaf area of both species increased when transplanted southwards, but decreased with open‐top chamber installation in A. nemorosa. In general, individuals of both species transplanted at the home site performed best, suggesting local adaptation. We conclude that contrasting understorey plants may display divergent plasticity in response to changing temperatures which may alter future understorey community dynamics.  相似文献

Even though the liver synthesizes most of circulating IGF-1, it lacks its receptor under physiological conditions. However, according to previous studies, a damaged liver expresses the receptor. For this reason, herein, we examine hepatic histology and expression of genes encoding proteins of the cytoskeleton, extracellular matrix, and cell-cell molecules and inflammation-related proteins. A partial IGF-1 deficiency murine model was used to investigate IGF-1’s effects on liver by comparing wild-type controls, heterozygous igf1+/?, and heterozygous mice treated with IGF-1 for 10 days. Histology, microarray for mRNA gene expression, RT-qPCR, and lipid peroxidation were assessed. Microarray analyses revealed significant underexpression of igf1 in heterozygous mice compared to control mice, restoring normal liver expression after treatment, which then normalized its circulating levels. IGF-1 receptor mRNA was overexpressed in Hz mice liver, while treated mice displayed a similar expression to that of the controls. Heterozygous mice showed overexpression of several genes encoding proteins related to inflammatory and acute-phase proteins and underexpression or overexpression of genes which coded for extracellular matrix, cytoskeleton, and cell junction components. Histology revealed an altered hepatic architecture. In addition, liver oxidative damage was found increased in the heterozygous group. The mere IGF-1 partial deficiency is associated with relevant alterations of the hepatic architecture and expression of genes involved in cytoskeleton, hepatocyte polarity, cell junctions, and extracellular matrix proteins. Moreover, it induces hepatic expression of the IGF-1 receptor and elevated acute-phase and inflammation mediators, which all resulted in liver oxidative damage.  相似文献

The protein kinase R (PKR) functions in the antiviral response by controlling protein translation and inflammatory cell signaling pathways. We generated a transgenic, knock-in mouse in which the endogenous PKR is expressed with a point mutation that ablates its kinase activity. This novel animal allows us to probe the kinase-dependent and -independent functions of PKR. We used this animal together with a previously generated transgenic mouse that is ablated for PKR expression to determine the role of PKR in regulating the activity of the cryopyrin inflammasome. Our data demonstrate that, in contradiction to earlier reports, PKR represses cryopyrin inflammasome activity. We demonstrate that this control is mediated through the established function of PKR to inhibit protein translation of constituents of the inflammasome to prevent initial priming during innate immune signaling. These findings identify an important role for PKR to dampen inflammation during the innate immune response and caution against the previously proposed therapeutic strategy to inhibit PKR to treat inflammation.  相似文献

Urinary tract infections are commonly caused by α-hemolysin (HlyA)-producing Escherichia coli. In erythrocytes, the cytotoxic effect of HlyA is strongly amplified by P2X receptors, which are activated by extracellular ATP released from the cytosol of the erythrocytes. In renal epithelia, HlyA causes reversible [Ca2+]i oscillations, which trigger interleukin-6 (IL-6) and IL-8 release. We speculate that this effect is caused by HlyA-induced ATP release from the epithelial cells and successive P2 receptor activation. Here, we demonstrate that HlyA-induced [Ca2+]i oscillations in renal epithelia were completely prevented by scavenging extracellular ATP. In accordance, HlyA was unable to inflict any [Ca2+]i oscillations in 132-1N1 cells, which lack P2R completely. After transfecting these cells with the hP2Y2 receptor, HlyA readily triggered [Ca2+]i oscillations, which were abolished by P2 receptor antagonists. Moreover, HlyA-induced [Ca2+]i oscillations were markedly reduced in medullary thick ascending limbs isolated from P2Y2 receptor-deficient mice compared with wild type. Interestingly, the following HlyA-induced IL-6 release was absent in P2Y2 receptor-deficient mice. This suggests that HlyA induces ATP release from renal epithelia, which via P2Y2 receptors is the main mediator of HlyA-induced [Ca2+]i oscillations and IL-6 release. This supports the notion that ATP signaling occurs early during bacterial infection and is a key player in the further inflammatory response.  相似文献

A drill core through the Lower Ordovician Tøyen Shale Formation at Lerhamn, NW Scania, southern Sweden, provides important new information for the precise biostratigraphic resolution of the Floian to lower Darriwilian time interval in southern Scandinavia. The Hunnegraptus copiosus, Tetragraptus phyllograptoides, Cymatograptus protobalticus (new), Baltograptus vacillans (new), Baltograptus sp. cf. Baltograptus deflexus (new), Baltograptus minutus (new), Isograptus victoriae, Undulograptus austrodentatus (Arienigraptus zhejiangensis and Undulograptus sinicus Subzones) and the (?)Corymbograptus retroflexus (new) biozones are differentiated in the core and their correlation in Scania is discussed. The Lerhamn drill core provides the most detailed graptolite record of the Floian Stage in Scandinavia. The interval is dominated by a number of species of the genus Baltograptus, endemic to the Atlantic Faunal realm and highly useful for regional biostratigraphic correlation. The biostratigraphic framework is based on endemic and pandemic faunal elements. The mixture of both elements in the drill core allows a more precise inter‐continental correlation of Lower to Middle Ordovician graptolite faunas and may – in the future – provide information as to the climatic history of regions dominated by Baltograptus faunas in the Floian. Translocation of vesicles within the cytoplasm is essential to normal cell function. The vesicles are typically transported along the microtubules to their destination. The aim of this study was to characterize the vesicular movement in resting and stimulated renal epithelial cells. MDCK cells loaded with either quinacrine or acridine orange, dyes taken up by acidic vesicles, were observed at 37°C in semiopen perfusion chambers. Time-lapse series were analyzed by Imaris software. Our data revealed vigorous movement of stained vesicles in resting MDCK cells. These movements seem to require intact microtubules because nocodazole leads to a considerable reduction of the vesicular movements. Interestingly, we found that extracellular ATP caused the vesicular movement to cease. This observation was obvious in time lapse. Similarly, other stimuli known to increase the intracellular Ca2+ concentration ([Ca2+]i) in MDCK cells (increment in the fluid flow rate or arginine vasopressin) also reduced the vesicular movement. These findings were quantified by analysis of single vesicular movement patterns. In this way, ATP was found to reduce the lateral displacement of the total population of vesicles by 40%. Because all these perturbations increase [Ca2+]i, we speculated that this increase in [Ca2+]i was responsible for the vesicle arrest. Therefore, we tested the effect of the Ca2+ ionophore, ionomycin (1 μM), which in the presence of extracellular Ca2+ resulted in a considerable and sustained reduction of vesicular movement amounting to a 58% decrease in average lateral vesicular displacement. Our data suggest that vesicles transported on microtubules are paused when subjected to high intracellular Ca2+ concentrations. This may provide an additional explanation for the cytotoxic effect of high [Ca2+]i.  相似文献

Abstract: The biostratigraphically important Middle Ordovician (Darriwilian) biserial graptolite Fucoides dentatus Brongniart, 1828 is redescribed and illustrated from its type material and from additional specimens collected at the type locality – Lévis, Quebec, Canada. It is referred to Levisograptus gen. nov., which includes also the austrodentatus group of early axonophoran graptolites. The species has previously been confused with a younger, mid‐Darriwilian species, now referred to as Eoglyptograptus gerhardi sp. nov., and recognized as the type species of the genus Eoglyptograptus Mitchell. Both species can be differentiated easily by their respective proximal development types and show nonoverlapping biostratigraphical ranges. Levisograptus dentatus (Brongniart) is an important biostratigraphical index species in the early Darriwilian. Eoglyptograptus species are found in the higher Darriwilian and are biogeographically restricted to the Atlantic Faunal Realm.  相似文献

Abstract: Several new taxa of snakeflies (Raphidioptera) are described from the Lower Cretaceous deposits of the Wealden, UK (Barremian), Montsec, Spain (Barremian) and the Crato Formation, Brazil (Aptian). Mesoraphidia ednae sp. nov. and M. hilli sp. nov. are described from the Wealden; Nanoraphidia lithographica sp. nov. and Iberoraphidia dividua gen. et sp. nov. are described from Montsec, and Baissoptera lisae sp. nov. is described from the Crato Formation. Abstract:  Early Middle Ordovician graptolites from the Albjära and Lovisefred drill cores (Scania, southern Sweden) can be differentiated into the Isograptus victoriae , Isograptus sp. nov. 2 and Maeandrograptus schmalenseei , Arienigraptus dumosus type/ Pseudisograptus manubriatus biozones of Castlemainian and Yapeenian age and the Arienigraptus zhejiangensis and Undulograptus sinicus biozones of early Darriwilian age, replacing the old, long-ranging Didymograptus hirundo Biozone. Faunas from the drill cores include a number of species not previously recognized in Scandinavia.  相似文献
