Aims: In the present communication, attempts have been made to regulate the tacrolimus production by supplementing commercial source of carbons and amino acids timely.
Methods and Results: Tacrolimus production was regulated by supplying vegetable oils and amino acids, individually and in combination. Tacrolimus quantification was done by HPLC. Streptomyces spp. MA6858 B3178 was found to produce 115·3 mg l−1 of tacrolimus. The rotation speed of shake flask, pH of the broth and supply of air were maintained at 7·1, 230 rev min−1 and 2·0 vvm air respectively.
Conclusions: The effect of carbons on tacrolimus production was noticed to be of diphasic manner. During the first 24 h of culture, monosaccharide is used for the growth of microbe. However, after the lapse of 36 h, addition of soya oil and l -lysine in combination enhanced the tacrolimus production to 115·3 mg l−1 . Besides this, pH of broth was also noticed as a critical factor in monitoring tacrolimus biosynthesis.
Significance and Impact of the Study: The newly isolated mutant Streptomyces spp. MA6858 B3178 having high potential for tacrolimus production as compared to existing data can be well used for the commercialization of tacrolimus. 相似文献

Abstract Sustainable application of fly ash and its management in agriculture is a major challenge nowadays. A pot culture experiment was conducted to find out the most suitable level of fly ash application for soil amendments that can improve the plant growth and productivity of Brahmi (Bacopa monnieri L.). After growing seedlings of B. monnieri under different levels of fly ash for 90?days, a significant increase in plant biomass, essential oil content and tolerance index (more than 100%) was observed under 25% of fly ash amended soil in comparison to garden soil and higher fly ash treatments. Leaf chlorophyll content and photosynthetic parameters were remained unchanged under 25% of fly ash as compared to seedlings grown on garden soil. However, these parameters were significantly declined under higher concentrations of fly ash treatments. Higher levels of fly ash caused oxidative damage and the induction of some antioxidative enzymes activities in B. monnieri indicates its capability to endure oxidative stress tolerance. Overall, our study showed that 25% of fly ash can be used as soil amendment for cultivation of B. monnieri L. leading to enhance plant biomass and essential oil production. 相似文献

Trichloroacetic acid (TCA) has been tested for mutagenicity in a mouse in vivo system. Three different cytogenetic assays--bone marrow chromosomal aberrations, micronucleus and sperm-head abnormalities--have been carried out. Swiss mice have been treated with the chemical, administered via different routes (i.p. and p.o.), and in three acute and/or fractionated doses (5 consecutive daily) equivalent to the highest acute dose, and their cells sampled at different intervals. A variety of anomalies, occurring in higher percentages compared to controls, was observed in all cases. Comparison between single and fractionated dosing revealed the single dosing to be more effective cytogenetically. The results were route and time-dependent but not dose-responsive (exclusive of gaps). The relative sensitivity of the assays has been found to be: chromosome aberration greater than sperm-head abnormality greater than micronucleus. The results revealed the genotoxic property of TCA in the present test system. 相似文献

Cytogenetic analyses of bone marrow and gonadal cells in a male mouse, which appeared to be normal, revealed mosaicism in both tissues. Three chromosome complements, 39,X, 40,XY, and 41,XYY, were found in both bone marrow and spermatogonia, while only the last two complements were found in spermatocytes. In this mouse, unlike in the human, the XYY cells showed a proliferative advantage over the XY cells. In XYY cells at diakinesis/metaphase I the gonosomes showed all possible types of association, and a pairing advantage of the X chromosome was clearly demonstrated. The fertility of the mouse was not determined. However, since the epididymal sperm count was reduced by only 55% and the incidence of sperm head abnormality was near normal, it is not evident that the mouse was sterile. 相似文献
