A method was established for staining and counting of actively respiring bacteria in natural stone by using the tetrazolium salt 5-cyano-2,3-ditolyltetrazolium chloride (CTC) in combination with confocal laser scanning microscopy (CLSM). Applying 5 mM CTC for 2 h to pure cultures of representative stone-inhabiting microorganisms showed that chemoorganotrophic bacteria and fungi-in contrast to lithoautotrophic nitrifying bacteria-were able to reduce CTC to CTF, the red fluorescing formazan crystals of CTC. Optimal staining conditions for microorganisms in stone material were found to be 15 mM CTC applied for 24 h. The cells could be visualized on transparent and nontransparent mineral materials by means of CLSM. A semi-automated method was used to count the cells within the pore system of the stone. The percentage of CTC-stained bacteria was dependent on temperature and humidity of the material. At 28 degrees C and high humidity (maximum water holding capacity) in the laboratory, about 58% of the total bacterial microflora was active. On natural stone exposed for 9 years at an urban exposure site in Germany, 52-56% of the bacterial microflora was active at the east, west, and north side of the specimen, while only 18% cells were active at the south side. This is consistent with microclimatic differences on the south side which was more exposed to sunshine thus causing UV and water stress as well as higher temperatures on a microscale level. In combination with CLSM, staining by CTC can be used as a fast method for monitoring the metabolic activity of chemoorganotrophic bacteria in monuments, buildings of historic interest or any art objects of natural stone. Due to the small size of samples required, the damage to these objects and buildings can be minimized.  相似文献

Porcine endogenous retroviruses (PERVs) pose a potential stumbling block for therapeutic xenotransplantation, with the greatest threat coming from viruses generated by recombination between members of the PERV subgroup A (PERV-A) and PERV-C families (PERV-A/C recombinants). PERV-A and PERV-B have been shown to infect human cells in culture, albeit with low titers. PERV-C has a more restricted host range and cannot infect human cells. A recombinant PERV-A/C virus (PERV-A14/220) contains the PERV-A sequence between the end of pol and the middle of the SU region in env. The remaining sequence is derived from PERV-C. PERV-A14/220 is approximately 500-fold more infectious than PERV-A. To determine the molecular basis for the increased infectivity of PERV-A14/220, we have made a series of vector constructs. The primary determinant for the enhanced replicative potential of the recombinant virus appeared to be the env gene. Using a series of chimeric env genes, we could identify two determinants of high infectivity; one was an isoleucine to valine substitution at position 140 between variable regions A and B, and the other lies within the proline rich region. Taken together, these results show that the novel juxtaposition of env gene sequences enhanced the infectivity of PERV-A14/220 for human cells, perhaps by stabilization of the envelope glycoprotein or increased receptor binding.  相似文献

Fecal bacteria were studied in healthy elderly volunteers (age, 63 to 90 years; n = 35) living in the local community, elderly hospitalized patients (age, 66 to 103; n = 38), and elderly hospitalized patients receiving antibiotic treatment (age, 65 to 100; n = 21). Group- and species-specific primer sets targeting 16S rRNA genes were used to quantitate intestinal bacteria by using DNA extracted from feces and real-time PCR. The principal difference between healthy elderly volunteers and both patient cohorts was a marked reduction in the Bacteroides-Prevotella group following hospitalization. Reductions in bifidobacteria, Desulfovibrio spp., Clostridium clostridiiforme, and Faecalibacterium prausnitzii were also found in the hospitalized patients. However, total 16S rRNA gene copy numbers (per gram of wet weight of feces) were generally lower in the stool samples of the two groups of hospitalized patients compared to the number in the stool samples of elderly volunteers living in the community, so the relative abundance (percentage of the group- and species-specific rRNA gene copies in relation to total bacterial rRNA gene copies) of bifidobacteria, Desulfovibrio spp., C. clostridiiforme, and F. prausnitzii did not change. Antibiotic treatment resulted in further reductions in the numbers of bacteria and their prevalence and, in some patients, complete elimination of certain bacterial communities. Conversely, the numbers of enterobacteria increased in the hospitalized patients who did not receive antibiotics, and due to profound changes in fecal microbiotas during antibiotic treatment, the opportunistic species Enterococcus faecalis proliferated.  相似文献

In the vascular plant Arabidopsis thaliana, synthesis of cysteine and its precursors O-acetylserine and sulfide is distributed between the cytosol, chloroplasts, and mitochondria. This compartmentation contributes to regulation of cysteine synthesis. In contrast to Arabidopsis, cysteine synthesis is exclusively restricted to chloroplasts in the unicellular green alga Chlamydomonas reinhardtii. Thus, the question arises, whether specification of compartmentation was driven by multicellularity and specified organs and tissues. The moss Physcomitrella patens colonizes land but is still characterized by a simple morphology compared to vascular plants. It was therefore used as model organism to study evolution of compartmented cysteine synthesis. The presence of O-acetylserine(thiol)lyase (OAS-TL) proteins, which catalyze the final step of cysteine synthesis, in different compartments was applied as criterion. Purification and characterization of native OAS-TL proteins demonstrated the presence of five OAS-TL protein species encoded by two genes in Physcomitrella. At least one of the gene products is dual targeted to plastids and cytosol, as shown by combination of GFP fusion localization studies, purification of chloroplasts, and identification of N termini from native proteins. The bulk of OAS-TL protein is targeted to plastids, whereas there is no evidence for a mitochondrial OAS-TL isoform and only a minor part of OAS-TL protein is localized in the cytosol. This demonstrates that subcellular diversification of cysteine synthesis is already initialized in Physcomitrella but appears to gain relevance later during evolution of vascular plants.  相似文献

The precise mechanisms regulating hepatitis C virus (HCV) entry into hepatic cells remain unknown. However, several cell surface proteins have been identified as entry factors for this virus. Of these molecules, claudin-1, a tight junction (TJ) component, is considered a coreceptor required for HCV entry. Recently, we have demonstrated that HCV envelope glycoproteins (HCVgp) promote structural and functional TJ alterations. Additionally, we have shown that the intracellular interaction between viral E2 glycoprotein and occludin, another TJ-associated protein, could be the cause of the mislocalization of TJ proteins. Herein we demonstrated, by using cell culture-derived HCV particles (HCVcc), that interference of occludin expression markedly reduced HCV infection. Furthermore, our results with HCV pseudotyped particles indicated that occludin, but not other TJ-associated proteins, such as junctional adhesion molecule A or zonula occludens protein 1, was required for HCV entry. Using HCVcc, we demonstrated that occludin did not play an essential role in the initial attachment of HCV to target cells. Surface protein labeling experiments showed that both expression levels and cell surface localization of HCV (co)receptors CD81, scavenger receptor class B type I, and claudin-1 were not affected upon occludin knockdown. In addition, immunofluorescence confocal analysis showed that occludin interference did not affect subcellular distribution of the HCV (co)receptors analyzed. However, HCVgp fusion-associated events were altered after occludin silencing. In summary, we propose that occludin plays an essential role in HCV infection and probably affects late entry events. This observation may provide new insights into HCV infection and related pathogenesis.Hepatitis C virus (HCV) is a small enveloped positive-strand RNA virus that belongs to the Flaviviridae family (20). More than 80% of acute infections become chronic, which eventually progress to cirrhosis and hepatocellular carcinoma (28). HCV infects mainly hepatocytes, but the precise mechanisms of infection are largely unknown (11). The HCV particle consists of a nucleocapsid surrounded by a lipid bilayer in which the two envelope glycoproteins (HCVgp), E1 and E2, are anchored as a heterodimer and play a major role in HCV entry (20). The development of an infectious cell culture model based on the production of infective HCV particles (cell culture-derived HCV particles [HCVcc]) (34) and the generation of HCV pseudotyped retroviral particles (HCVpp) (4) have provided powerful tools to study the HCV cycle. HCV entry is a complex multistep process that requires the presence of several factors. There are multiple pieces of evidence for the involvement of host cell proteins in HCV entry, including glycosaminoglycans, the low-density lipoprotein receptor, scavenger receptor class B type I (SR-BI), and the tetraspanin CD81 (11). Recently, claudin-1, a tight junction (TJ) component, has been identified as a coreceptor required for a late step in HCV entry (13).TJs are major components of cell-cell adhesion complexes and are composed of integral membrane proteins, including occludin and claudins, which associate with actin cytoskeleton-interacting proteins, such as zonula occludens protein 1 (ZO-1) (2). These structures maintain cell polarity, separating apical from basolateral membrane domains, and form a paracellular barrier that allows the selective passage of certain solutes (2). In hepatocytes, TJs seal the bile canaliculi and form the intercellular barrier between bile and blood (12). Recently, we have shown that TJ-associated proteins occludin and claudin-1 disappeared from their normal localization in both HCV-infected and genomic HCV replicon-containing Huh7 cells. Furthermore, TJ function was also altered in these cells (5). In this matter, we have reported an intracellular interaction between E2 and occludin (5). Moreover, it has been reported that claudin-1 and several TJ-associated proteins, such as coxsackievirus and adenovirus receptor (35) and junctional adhesion molecule (JAM) (3), act as virus (co)receptors. Since coxsackievirus entry across epithelial TJs requires occludin (10), we have explored the role of occludin in HCV infection.  相似文献

Repeated interactions within individual human and animal dyads can lead to the establishment of human–animal relationships (HARs), which may vary in quality from good to bad, defined in terms of the positivity (e.g., friendly contact, play) or negativity (e.g., aggression) of the interactions on which they are based. Particularly good HARs can be regarded as Human– Animal Bonds (HABs) if they are reciprocal and promote wellbeing in both parties. Although there is extensive evidence of the effects of HARs in agricultural animals and HABs in companion animals, there has been less investigation of these relationships in zoos, even though the development of HARs/HABs between zoo animals and their keepers could have important consequences for the welfare of both. Here we apply a modified version of the Lexington Attachment to Pets Scale (LAPS) in a zoo setting to quantify the strength of attachment of a sample of 22 keepers to the animals in their care at the zoo (ZA), in comparison with their attachment to their companion animals at home (PA). Results showed that mean PA scores (47.54 ± 3.6) were significantly higher than mean ZA scores (32.89 ± 2.6; t = –5.16, df = 13, p < 0.001), indicating stronger attachment to the companion animals. PA scores were lower in keepers who thought it inappropriate to have a bond with a zoo animal, compared with those who deemed it appropriate. Thus, HABs do appear to occur in the zoo context, though they are weaker than those developed in the home. This work also shows that a modified LAPS questionnaire is a suitable instrument for further investigation of HABs in zoos.  相似文献

Oestrous and dioestrous rats were observed during the initial 2 min of open-field exposure, and after a loud bell had sounded. Although defecation and ambulation showed characteristic difference between the two phases (less defecation and more ambulation at oestrus), other measures associated with emotional behaviour (latency to move from the wall, flight, and freezing to the bell) showed no differences. These findings supported Drewett's suggestion that open-field defecation and ambulation change with oestrous phase because of general metabolic changes and not as a result of 'emotionality' changes.  相似文献

Hepatitis C virus (HCV) is an oncogenic virus associated with the onset of hepatocellular carcinoma (HCC). The present study investigated the possible link between HCV infection and Netrin-1, a ligand for dependence receptors that sustains tumorigenesis, in particular in inflammation-associated tumors. We show that Netrin-1 expression is significantly elevated in HCV+ liver biopsies compared to hepatitis B virus (HBV+) and uninfected samples. Furthermore, Netrin-1 was upregulated in all histological stages of HCV+ hepatic lesions, from minimal liver fibrosis to cirrhosis and HCC, compared to histologically matched HCV- tissues. Both cirrhosis and HCV contributed to the induction of Netrin-1 expression, whereas anti-HCV treatment resulted in a reduction of Netrin-1 expression. In vitro, HCV increased the level and translation of Netrin-1 in a NS5A-La-related protein 1 (LARP1)-dependent fashion. Knockdown and forced expression experiments identified the receptor uncoordinated receptor-5 (UNC5A) as an antagonist of the Netrin-1 signal, though it did not affect the death of HCV-infected cells. Netrin-1 enhanced infectivity of HCV particles and promoted viral entry by increasing the activation and decreasing the recycling of the epidermal growth factor receptor (EGFR), a protein that is dysregulated in HCC. Netrin-1 and HCV are, therefore, reciprocal inducers in vitro and in patients, as seen from the increase in viral morphogenesis and viral entry, both phenomena converging toward an increase in the level of infectivity of HCV virions. This functional association involving a cancer-related virus and Netrin-1 argues for evaluating the implication of UNC5 receptor ligands in other oncogenic microbial species.  相似文献
