Endopeptidase Activity of Phage λ-Endolysin

JACOB and Fuerst^1,2 demonstrated the presence of a bacteriolytic enzyme (λ-endolysin) in the induced cultures of lysogenic Escherichia coli K12 (λ). The enzyme was later identified as the product of gene R; of phage λ³ which is involved in bacterial lysis at the end of a latent period. The enzyme is apt to form spheroplast-like structures in E. coli² and one would therefore expect its substrate to be murein.     

Development and maturation of surghum seeds on detached panicles grown in vitro

Summary A procedure for culturing detached panicles of sorghum, Sorghum bicolor (L.) Moench, was developed to achieve flowering, fertilization, and subsequent seed development and maturation in vitro. Sixteen sorghum genotypes (five high and eleven low in tannin) were tested for their ability to develop normally in culture. Panicles collected one to two days before the initiation of anthesis were cultured in flasks containing liquid medium. Contamination and medium darkening were the major obstacles encountered. Up to 55% of the panicles cultured reached physiological maturity in vitro. The frequency of seed set ranged from 30 to 97% depending upon genotype and medium. Seed and glume color were normal. Seed produced in vitro resembled those grown in vivo and germinated well, but were smaller than normal (100 kernel weight reached 50 to 70% of the control). Grain polyphenols were synthesized in the cultured panicles. Seed of high tannin genotypes produced in vitro were lower in total phenols and tannins and higher in flavan-4-ols and the 3-deoxyanthocyanidin pigments than control seed. This technique can be used for harvesting late-maturing stocks and for various sorghum studies.     

Hydrolysis of phosphatidylcholine couples Ras to activation of Raf protein kinase during mitogenic signal transduction.

We have investigated the relationship between hydrolysis of phosphatidylcholine (PC) and activation of the Raf-1 protein kinase in Ras-mediated transduction of mitogenic signals. As previously reported, cotransfection of a PC-specific phospholipase C (PC-PLC) expression plasmid bypassed the block to cell proliferation resulting from expression of the dominant inhibitory mutant Ras N-17. In contrast, PC-PLC failed to bypass the inhibitory effect of dominant negative Raf mutants, suggesting that PC-PLC functions downstream of Ras but upstream of Raf. Consistent with this hypothesis, treatment of quiescent cells with exogenous PC-PLC induced Raf activation, even when normal Ras function was blocked by Ras N-17 expression. Further, activation of Raf in response to mitogenic growth factors was blocked by inhibition of endogenous PC-PLC. Taken together, these results indicate that hydrolysis of PC mediates Raf activation in response to mitogenic growth factors.     

慢性心功能不全大鼠心脏和血淋巴细胞β-肾上腺素受体的改变

在主动脉与肾动脉缩窄造成的慢性心功能不全大鼠,血浆儿茶酚胺浓度增高;心脏β-肾上腺素受体(β-AR)数量增加,其中β_1-AR及其mRNA增加,而β_2-AR及其mRNA不变;左心房异丙基肾上腺素(ISO)浓度-收缩效应曲线右移;而心肌ISO浓度-cAMP蓄积曲线无显著改变;血淋巴细胞β-AR数量显著减少.结果提示心功能不全时心脏β_1-AR数量增多,但其介导的正性变力效应反而降低,在cAMP生成以后的信号转导过程或心肌收缩成分功能存在障碍,而血淋巴细胞β-AR的改变与心脏β-AR的功能改变平行.     

Mechanism of Action of Lycoricidinol, a Plant Growth Inhibitor Isolated from Lycoris radiata Herb.

We studied the action mechanism of lycoricidinol, a plant growthinhibitor isolated from Lycoris radiata Herb. Lycoricidinolinhibited protein synthesis in mung bean hypocotyls, but notRNA synthesis. Protein synthesis in Escherichia coli was notaffected by the inhibitor. Results of in vitro translation experimentswith the wheat germ system and the E. coli system indicatedthat lycoricidinol inhibited only eukaryotic but not prokaryotictranslation. Use of specific inhibitors of initiation and polypeptidechain elongation of polypeptide synthesis revealed that chainelongation was inhibited by lycoricidinol. ¹Permanent address: Department of Biology, Yonsei University,Seoul 120, Korea. Protection against acute paraquat toxicity by ambroxol

An expression vector for G-CSF, pASLB3-3, was constructed and introduced into Namalwa KJM-1 cells (Hosoi et al., 1988), and cells resistant to 100 nM of methotrexate (MTX) were obtained. Among them, the highest producer, clone SC57, was selected and the productivity of this clone was further characterized. The maximal production of G-CSF was at the most 1.8 g/ml/day using a 25 cm² tissue culture flask, even though the cell number was above 7×10⁵ cells/ml. The limiting factors at high density were analyzed as the deficiency of nutrients, such as glucose, cysteine and serine, and pH control. The depression of specific G-CSF productivity per cell under the batch culture conditions was overcome by using a perfusion culture system, Biofermenter^TM (Sato, 1983) with modifications of nutrients supplementation by a dialysis membrane and/or dissolved oxygen (DO) supplementation by microsilicone fibers. ITPSGF medium was modified to elevate concentrations of amino acids and glucose by 2.0- and 2.5-times, respectively. Under the control of pH at 7.4 and DO at 3 ppm, the specific G-CSF productivity was not depressed even at high cell density (above 1×10⁷ cells/ml), and the amount of G-CSF reached 41 g/ml.
