The moth Spodoptera littoralis, is a major pest of agriculture whose olfactory system is tuned to odorants emitted by host plants and conspecifics. As in other insects, the paired mushroom bodies are thought to play pivotal roles in behaviors that are elicited by contextual and multisensory signals, amongst which those of specific odors dominate. Compared with species that have elaborate behavioral repertoires, such as the honey bee Apis mellifera or the cockroach Periplaneta americana, the mushroom bodies of S. littoralis were originally viewed as having a simple cellular organization. This has been since challenged by observations of putative transmitters and neuromodulators. As revealed by immunocytology, the spodopteran mushroom bodies, like those of other taxa, are subdivided longitudinally into discrete neuropil domains. Such divisions are further supported by the present study, which also demonstrates discrete affinities to different mushroom body neuropils by antibodies raised against two putative transmitters, glutamate and gamma-aminobutyric acid, and against three putative neuromodulatory substances: serotonin, A-type allatostatin, and tachykinin-related peptides. The results suggest that in addition to longitudinal divisions of the lobes, circuits in the calyces and lobes are likely to be independently modulated.  相似文献

Female mosquitoes depend on blood to complete their reproductive cycle and rely mainly on chemosensory systems to obtain blood meals. An immunocytochemical analysis reveals a number of serotonin-immunoreactive neurons that innervate the chemosensory systems, suggesting a potential role of serotonin in modulating chemosensory processes. In the primary olfactory system, we identify a single ipsilateral centrifugal neuron with arborizations in higher brain centers; the varicosities of this neuron display volumetric changes in response to both blood feeding and during a circadian rhythm. Six to eight pairs of serotonin-immunoreactive neurons are identified in the primary gustatory neuropil, including the subesophageal ganglion and tritocerebrum. The peripheral chemosensory organs, i.e. the antenna, the maxillary palp and the labium, are described as having extensive serotonergic neurohemal plexi. In addition, we describe the presence of serotonin-immunoreactive fibers in the mechanosensory Johnston's organ. Taking these results together, we discuss the potential role of serotonin as a neuromodulator in the chemosensory system of disease vector mosquitoes.  相似文献

Hot-cold hemolysis is the phenomenon whereby red blood cells, preincubated at 37 degrees C in the presence of certain agents, undergo rapid hemolysis when transferred to 4 degrees C. The mechanism of this phenomenon is not understood. PlcHR 2, a phospholipase C/sphingomyelinase from Pseudomonas aeruginosa, that is the prototype of a new phosphatase superfamily, induces hot-cold hemolysis. We found that the sphingomyelinase, but not the phospholipase C activity, is essential for hot-cold hemolysis because the phenomenon occurs not only in human erythrocytes that contain both phosphatidylcholine (PC) and sphingomyelin (SM) but also in goat erythrocytes, which lack PC. However, in horse erythrocytes, with a large proportion of PC and almost no SM, hot-cold hemolysis induced by PlcHR 2 is not observed. Fluorescence microscopy observations confirm the formation of ceramide-enriched domains as a result of PlcHR 2 activity. After cooling down to 4 degrees C, the erythrocyte ghost membranes arising from hemolysis contain large, ceramide-rich domains. We suggest that formation of these rigid domains in the originally flexible cell makes it fragile, thus highly susceptible to hemolysis. We also interpret the slow hemolysis observed at 37 degrees C as a phenomenon of gradual release of aqueous contents, induced by the sphingomyelinase activity, as described by Ruiz-Arguello et al. [(1996) J. Biol. Chem. 271, 26616]. These hypotheses are supported by the fact that ceramidase, which is known to facilitate slow hemolysis at 37 degrees C, actually hinders hot-cold hemolysis. Differential scanning calorimetry of erytrocyte membranes treated with PlcHR 2 demonstrates the presence of ceramide-rich domains that are rigid at 4 degrees C but fluid at 37 degrees C. Ceramidase treatment causes the disapperance of the calorimetric signal assigned to ceramide-rich domains. Finally, in liposomes composed of SM, PC, and cholesterol, which exhibit slow release of aqueous contents at 37 degrees C, addition of 10 mol % ceramide and transfer to 4 degrees C cause a large increase in the rate of solute efflux.  相似文献

Data replication is a key issue in a Data Grid and can be managed in different ways and at different levels of granularity: for example, at the file level or object level. In the High Energy Physics community, Data Grids are being developed to support the distributed analysis of experimental data. We have produced a prototype data replication tool, the Grid Data Mirroring Package (GDMP) that is in production use in one physics experiment, with middleware provided by the Globus Toolkit used for authentication, data movement, and other purposes. We present here a new, enhanced GDMP architecture and prototype implementation that uses Globus Data Grid tools for efficient file replication. We also explain how this architecture can address object replication issues in an object-oriented database management system. File transfer over wide-area networks requires specific performance tuning in order to gain optimal data transfer rates. We present performance results obtained with GridFTP, an enhanced version of FTP, and discuss tuning parameters.  相似文献

This study examines the use levonorgestrel (Norplant®‐2) as a means of contracepting white‐faced sakis (Pithecia pithecia). A 70‐mg silastic tubule of levonorgestrel was implanted subdermally between the scapulae in four multiparous and two nulliparous females. Implants remained in the females housed with males of proven fertility for 81 to 734 days. All females experienced low, acyclic levels of estrone conjugates (E1C, < 500 ng/mg Cr) and pregnanediol‐3‐glucuronide (PdG, < 1.5 μg/mg Cr) after insertion of the implant. In five females having nine contraceptive bouts, ovulation was completely blocked for the entire duration of Norplant insertion (range: 81–734 days) as evidenced by a lack of cyclic changes in excretory hormone levels. However, one female was observed to have cyclic ovarian hormonal levels 5.8 months after Norplant insertion. Thus, levonorgestrel can be an effective method for blocking cyclic ovarian hormonal changes for 1 year (n = 2) and possibly as long as 2 years (n = 3). Mutations in the amyloid precursor protein (APP) are responsible for the formation of amyloid‐β peptides. These peptides play a role in Alzheimer's and other dementia‐related diseases. The cargo binding domain of the kinesin‐1 light chain motor protein (KLC1) may be responsible for transporting APP either directly or via interaction with C‐jun N‐terminal kinase‐interacting protein 1 (JIP1). However, to date there has been no direct experimental or computational assessment of such binding at the atomistic level. We used molecular dynamics and free energy estimations to gauge the affinity for the binary complexes of KLC1, APP, and JIP1. We find that all binary complexes (KLC1:APP, KLC1:JIP1, and APP:JIP1) contain conformations with favorable binding free energies. For KLC1:APP the inclusion of approximate entropies reduces the favorability. This is likely due to the flexibility of the 42‐residue APP protein. Euplotes is diversified into dozens of widely distributed species that produce structurally homologous families of water‐borne protein pheromones governing self‐/nonself‐recognition phenomena. Structures of pheromones and pheromone coding genes have so far been studied from species lying in different positions of the Euplotes phylogenetic tree. We have now cloned the coding genes and determined the NMR molecular structure of four pheromones isolated from Euplotes petzi, a polar species which is phylogenetically distant from previously studied species and forms the deepest branching clade in the tree. The E. petzi pheromone genes have significantly shorter sequences than in other congeners, lack introns, and encode products of only 32 amino acids. Likewise, the three‐dimensional structure of the E. petzi pheromones is markedly simpler than the three‐helix up‐down‐up architecture previously determined in another polar species, Euplotes nobilii, and in a temperate‐water species, Euplotes raikovi. Although sharing the same up‐down‐up architecture, it includes only two short α‐helices that find their topological counterparts with the second and third helices of the E. raikovi and E. nobilii pheromones. The overall picture that emerges is that the evolution of Euplotes pheromones involves progressive increases in the gene sequence length and in the complexity of the three‐dimensional molecular structure.  相似文献

Male and female C57BL/6J mice, initially 19 days old, consumed a complete purified diet either ad libitum (age-matched control) or in restricted daily quantities (energy deficiency), or they consumed a purified isocaloric low-protein diet ad libitum (protein and energy deficit). In a 14-day experimental period, malnourished animals lost approximately 1.5% of their initial body weight daily. Zero-time controls, 19 days old, were also included in the study. Serum levels of Th2-type (IgG1 and IgE) and Th1-type (IgG2a and IgG3) immunoglobulins were quantified by enzyme-linked immunosorbent assay, and total IgG concentration was also assessed. Both malnourished groups exhibited high serum concentrations of IgG1 and IgE relative to the age-matched control group, whereas levels of the Th1-type immunoglobulins were unaffected. Total IgG concentration in the malnourished groups reflected the usual finding in humans (i.e., no effect or elevated). The results are consistent with the proposition of Th2-polarized immune competence in acute weanling deficiencies of energy, protein, or both.  相似文献
