Northern corn leaf blight, caused by the fungal pathogen Exserohilum turcicum, is a major disease of maize. The first major locus conferring resistance to E. turcicum race 0, Ht1, was identified over 50 years ago, but the underlying gene has remained unknown. We employed a map-based cloning strategy to identify the Ht1 causal gene, which was found to be a coiled-coil nucleotide-binding, leucine-rich repeat (NLR) gene, which we named PH4GP-Ht1. Transgenic testing confirmed that introducing the native PH4GP-Ht1 sequence to a susceptible maize variety resulted in resistance to E. turcicum race 0. A survey of the maize nested association mapping genomes revealed that susceptible Ht1 alleles had very low to no expression of the gene. Overexpression of the susceptible B73 allele, however, did not result in resistant plants, indicating that sequence variations may underlie the difference between resistant and susceptible phenotypes. Modelling of the PH4GP-Ht1 protein indicated that it has structural homology to the Arabidopsis NLR resistance gene ZAR1, and probably forms a similar homopentamer structure following activation. RNA sequencing data from an infection time course revealed that 1 week after inoculation there was a threefold reduction in fungal biomass in the PH4GP-Ht1 transgenic plants compared to wild-type plants. Furthermore, PH4GP-Ht1 transgenics had significantly more inoculation-responsive differentially expressed genes than wild-type plants, with enrichment seen in genes associated with both defence and photosynthesis. These results demonstrate that the NLR PH4GP-Ht1 is the causal gene underlying Ht1, which represents a different mode of action compared to the previously reported wall-associated kinase northern corn leaf blight resistance gene Htn1/Ht2/Ht3.  相似文献

Zimbabwe provides a significant context to examine the interplay of the new development rhetoric, the actions of powerful conservation organizations, donor policies, a relatively strong and stable government, and the empowerment of local communities. This interplay exists in diverse rural areas where the Communal Areas Management Program for Indigenous Resources (CAMPFIRE) is in various stages of experimentation and implementation. CAMPFIRE has been described as a philosophy of sustainable rural development that enables rural communities to manage, and benefit directly form indigenous wildlife. It is the best known of African efforts to permit African communities to re- assert their management of selected natural resources. The program has the official support of the Zimbabwean government. Nonetheless, there are many potential areas of serious conflict. Three case studies are utilized to explore how the challenges of repossession of critical environmental resources by marginalized communities in the changing context of state and NGO relationships where international tourism is a revenue generator for both the private sector and government.  相似文献

The ternary phase diagram of cholesteryl myristate--dimyristoyl lecithin--water has been determined by polarizing light microscopy, scanning calorimetry, and x-ray diffraction. Hydrated dimyristoyl lecithin forms a lamellar liquid--crystalline phase (L alpha) at temperatures greater than 23 degrees C into which limited amounts of cholesteryl myristate (less than 5 wt. %) can be incorporated. The amount of cholesterol ester incorporated is dependent upon the degree of hydration of the L alpha phase. Below 23 degrees C dimyristoyl lecithin forms ordered hydrocarbon chain structures (L beta' and P beta') which do not incorporate cholesterol ester. Comparison with other phospholipid--cholesterol ester--water phase diagrams suggests the following general principles: i) the incorporation of cholesterol ester occurs only into liquid crystalling phospholipid bilayers, ii) the extent of incorporation is temperature-dependent, with increasing amounts of cholesterol ester being incorporated at higher temperatures, and iii) unsaturated cholesterol esters induce increased disordering of the phospholipid bilayers.  相似文献

In the Whitehall study of 18 403 male civil servants aged 40-64 years the 10 year mortality rates from coronary heart disease and stroke showed a non-linear relation to two hour blood glucose values, with a significantly increased risk for glucose intolerant subjects with concentrations above the 95th centile point (5.4-11.0 mmol/l; 96-199 mg/100 ml) and for diabetics (blood glucose greater than or equal to 11.1 mmol/l; greater than or equal to 200 mg/100 ml). Multiple logistic analysis showed that between one half and three quarters of the relative risks for deaths from coronary heart disease and stroke were "unexplained" by between group differences in risk factors such as age, blood pressure, obesity, smoking, cholesterol concentration, and electrocardiographic abnormalities. Within the glucose intolerant and diabetic groups the risk factors most strongly related to subsequent death from coronary heart disease were age and blood pressure, with less consistent relations for smoking, cholesterol concentration, and obesity. This study confirms the importance of hypertension as a cardiovascular risk factor in groups with glucose intolerance and diabetes, and this may have important preventive implications.  相似文献

Several agents that induce differentiation have previously been shown to induce the terminal differentiation of leukemic cells and enhance the radiosensitivity of certain solid tumor cell lines in vitro using millimolar concentrations. We now report that aclacinomycin A (ACM), a potent inducer of leukemic cell differentiation in vitro, can significantly enhance the radiosensitivity of a human colon tumor cell line (Clone A) at a concentration of 10 nM. Based on colony-forming efficiency, the maximum increase in radiosensitivity was found using 15 nM ACM for 3 days with a dose enhancement factor of 1.4 at a surviving fraction of 0.10. This treatment increased cell doubling time, but had no effect on cell-cycle phase distribution. To gain insight into the mechanisms responsible for this radiosensitization, gamma-ray-induced DNA single- and double-strand breaks were examined. Aclacinomycin A had no effect on the induction of DNA single-strand breaks but significantly enhanced the formation of gamma-ray-induced DNA double-strand breaks. The rate or extent of repair of the induced double-strand breaks was not influenced by ACM treatment. These data suggest that ACM, at achievable plasma concentrations, can enhance the radiosensitivity of a human tumor cell line by increasing the initial level of radiation-induced DNA double-strand breaks.  相似文献

The tethered and free flight of Manduca sexta were studied during period 1,2, and 0 times normal gravity (g) produced in an aeroplane by flying through parabolic trajectories. Moths in tethered flight did not change their aerodynamic output in response to increases or decreases in gravity. Some moths in free flight at 0 g maintained a position in the box by flying against a surface, or into the angle between two surfaces. In the absence of gravity as an orienting stimulus, the positive dorsophotic response to light was dominant. As the period of 0 g continued, moths were increasingly likely to periodically reduce the amplitude of their wingbeat and/or stop flying, for the equivalent of a few wingbeats. Only at 0 g, moths very occasionally spread their wings and floated freely for a few seconds. At 0 g moths retained control of rolling and yawing movements but stability in pitch was greatly reduced or absent.  相似文献

Utilizing X-ray diffraction and differential scanning calorimetry (DSC), we have studied (1) the structure and thermotropic properties of hydrated N-palmitoylsphingomyelin, (2) the interaction of N-palmitoylsphingomyelin with dimyristoyllecithin, and (3) the interaction of cholesterol with N-palmitoylsphingomyelin and dimyristoyllecithin, both individually and in a 50:50 (mol/mol) mixture. N-Palmitoylsphingomyelin forms bilayers which undergo a thermotropic order--disorder (gel--liquid crystalline) transition at 40.5 degrees C (delta H = 5.8 kcal/mol). The bilayer repeat distance is 66.8 A at 10 degrees C and 61.6 A at 50 degrees C. N-Palmitoylsphingomyelin exhibits miscibility with dimyristoylecithin in both the gel and liquid-crystalline phases, and no lateral phase separation occurs. Scanning calorimetry indicates that interaction with cholesterol is similar for both N-palmitoylsphingomyelin and dimyristoyllecithin and that in a 50:50 (mol/mol) mixture cholesterol shows no preferential affinity for either phospholipid.  相似文献
