Mice with mosaic mutation could be one of the models of human Menkes disease, which is associated with abnormal cooper metabolism. The aim of the present study was to localize androgen receptors (ARs) in the testes by means of immunohistochemistry. AR expression was observed in the nuclei of all somatic cells such as Leydig cells, Sertoli cells, and peritubular cells in sections from testes of control and mosaic mutant males. In the latter, very strong immunoreactivity for AR as well as higher levels of steroid hormones in homogenates were noticed in comparison to control mice. No positive immunoreaction for ARs was seen in control sections incubated without the primary antibody.  相似文献

During first stages of oogenesis in an ichneumonid wasp Coleocentrotus soldanskii. in each 32 cell cluster, as many as 8 cystocytes enter meiosis. Only one differentiates as an oocyte, the rest are gradually transdetermined to become nurse cells. Significantly, the nurse cells that have passed the first stages of meiosis retain some features characteristic of the oocyte. Namely the nuclei of these cells, as the oocyte nucleus, produce accessory nuclei (AN). Gradual transdetermination of pro-oocytes into nurse cells and the occurrence of ANs in transdetermined cells suggest that a diffusing substance, forming a concentration gradient, is responsible for the determination of the oocyte within the cystocyte groups in hymenopterans.  相似文献

Anaerobic synthesis of catalase T (a typical oxygen-inducible haem containing enzyme) in the cz1 mutant of yeast was demonstrated. The synthesis of catalase T in anaerobically grown mutant cells is stimulated by haemin under carbon-derepressed conditions of growth (galactose as carbon source) but not in glucose repressed cultures. Haem is practically undetectable in the anaerobically grown glucose repressed wild type strain and its level in derepressed cells amounts to 3% of the fully derepressed aerobically grown cells. In the cz1 mutant cultures grown in anoxia both on galactose and glucose the haem level usually exceeds 10% of that in the aerobic control.  相似文献

The content of haem and its precursors was determined in yeast cells grown under various conditions. The cells grown aerobically on 2% galactose contain about three times more haem (about 300 nmoles/g dry wt.) than the cells grown on 10% glucose. A trace amount of haem was found in anoxia irrespective of the carbon source used. The "efficiency" of the first enzyme of the haem biosynthetic pathway--delta-aminolevulinic acid (ALA) synthetase--expressed as the sum of all intermediates of the pathway in the cells grown on galactose, is similar in anaerobic and aerobic cells. The "efficiency" of the second enzyme--ALA dehydratase--is lower about three times both in anoxia and under conditions of glucose repression. In anoxia, not haem but delta-aminolevulinic acid is the main biosynthetic product. The role of glucose repression and of the feedback mechanisms in regulation of haem synthesis in yeast is discussed. A method for haem determination in the intact yeast cells, based on the formation of pyridine haemochrome, is described.  相似文献

Steroidal aryliminium salts were prepared from D-seco-pregnene aldehyde 2b, and their BF3.OEt2-catalyzed reactions were studied. The nature of the substituent R1 in the anilines 3-6 essentially influenced the chemoselectivity. Using unsubstituted 3, 4-methoxy- (4) or 4-bromoaniline (5), different tetrahydroquinoline derivatives 7a-13a via intramolecular hetero Diels-Alder reaction were formed. In the case of 4-nitroaniline (6) the N-arylamino-D-homopregnane (14a) were also obtained. We assume, that an intramolecular Prins reaction led to this type of fluoro-D-homosteroid. The main products represent a new class of tetrahydroquinolino-androstenes.  相似文献

NO has been implicated in the mechanism of ischaemic preconditioning. To verify this hypothesis further we have attempted to reproduce effects of ischaemic preconditioning by nitric oxide donors administration prior to the ischaemia. The effect of glyceryl trinitrate (GTN) and 3-morpholino-sydnonimine-hydrochloride (SIN- 1), NO donors, on reperfusion induced ventricular tachycardia (VT) and ventricular fibrillation (VF) in Langendorff perfused rat hearts subjected to 10 min regional ischaemia followed by 10 min reperfusion were examined. Results: GTN, 500 M and SIN-1, 10 M, administered for 5 min and washed for another 5 min prior to ischaemia (to mimic ischaemic preconditioning), almost completely abolished reperfusion induced VF. GTN and SIN-1, administered at the time of reperfusion, increased the incidence of sustained VF and the duration of VT and VF. When given 5 min before the ischaemia and throughout the ischaemia and the reperfusion, SIN-1 abolished VF. Adenosine, 10 M, applied according to the above three protocols, did not affect reperfusion arrhythmias, although adenosine induced changes in coronary flow and post-ischaemic reflow were similar to those produced by the NO donors. In conclusions: (1) NO is able to mimic the effect of ischaemic preconditioning on reperfusion arrhythmias in rat heart, supporting the view that NO may be one of the endogenous substances triggering ischaemic preconditioning; (2) In crystalloid-perfused heart, NO may be deleterious when its administration is restricted to the reperfusion period.  相似文献

INTRODUCTION: Evidence collected over the years has demonstrated that cryptorchidism is associated with a defect in spermatogenesis and, as a consequence, with either reduced fertility or infertility. However, the effect of cryptorchidism on Leydig cell function is less clear. The aim of our study therefore was to investigate the regulation of steroid hormone biosynthesis and, additionally, intercellular communication in the cryptorchid equine testes. MATERIAL AND METHODS: Testes of mature bilaterally cryptorchid horse and healthy stallions were used for this study. The expression of luteinising hormone receptor (LHR), 3beta-hydroxysteroid dehydrogenase (3beta-HSD), aromatase and connexin43 (Cx43) was detected by means of immunohistochemistry. Testosterone and oestradiol levels were measured in testicular homogenates using appropriate radioimmunoassays. RESULTS: In the testes of both normal and cryptorchid stallions, immunostaining for LHR, 3beta-HSD and aromatase was confined to the Leydig cells. In the cryptorchid horse, the intensity of the staining for LHR and 3beta-HSD was weaker, whereas the staining for aromatase was clearly stronger than that of the normal stallion. Radioimmunological analysis revealed disturbance of the androgen-oestrogen balance in the cryptorchid testes. Additionally, in both the seminiferous tubules and interstitial tissue of the cryptorchid a clear reduction of the Cx43 signal was observed. CONCLUSIONS: Decreased expression of LHR and 3beta-HSD and increased expression of aromatase in the cryptorchid testes suggest that hormonal imbalance was caused both by reduced testosterone synthesis and by increased androgen aromatisation. Impaired expression of Cx43 in the seminiferous tubules as well as in the interstitial tissue of the cryptorchid horse indicates that cryptorchidism affects intercellular communication in the testes.  相似文献

L'étude des pelotes de pollen et de nectar récoltés par les abeilles (Apis mellifera Var. mellifera), porte sur deux ruches d'un rucher situé dans une plantation d'Actinidia deliciosa Chev., dans la Province de Pontevedra (N. O. de l'Espagne). Les pelotes de pollen ont été recueillies dans des trappes à pollen pendant la période de floraison d'Actinidia deliciosa Chev. Des prélèvements de pollen ont également été effectués dans les jabots et les intestins des abeilles capturées a l'entrée des ruches. L'étude de tous ces échantillons a permis d'établir divers spectres palynologiques. Simultanément, une analyse de la concentration du pollen de cette espèce libéré dans l'atmosphère a été entreprise.

Les taxons les plus fréquemment récoltés par les abeilles sont: Taraxacum officinale Weber, Cistus psilosepalus Sweet, et des Crucifères avec un type pollinique affine de celui de Sinapis sp. Le pollen d'Actinidia deliciosa Chev. n'apparaît en quantité élevée dans l'atmosphère que pendant la période de pollinisation. Eucalyptus globulus Labill., Castanea sativa Miller et Taraxacum officinale Weber sont les principales sources nectarifères recherchées par les abeilles.

In order to carry out a study of pollen and nectar collected by bees (Apis mellifera Var. mellifera) we have choosen two hives from a apiary situated in a culture of Actinidia deliciosa Chev. in the province of Pontevedra (NW Spain). The pollen loads were collected during the flowering period, the intestine content and the honey sac of the bees were analysed from a palynological point of view. At the same time, a comparative analysis of the pollen content of this specimen in the atmosphere was made. The most important taxa in the pollen collected by bees are: Taraxacum officinale Weber, Cistus psilosepalus Sweet, and Sinapis sp. During the pollinization period the pollen of Actinidia deliciosa Chev. appears in high quantity in the air. The analysis of honey sac and intestine show that Eucalyptus globulus Labill., Castanea sativa Miller and Taraxacum officinale Weber are the principle nectar sources.  相似文献
