A neutrophil-specific peptide, cinnamoyl-F(D)LF(D)LFK (cFLFLFK), was conjugated consecutively with a polyethylene glycol moiety (3.4 K) and 2,2′,2″,2-(1,4,7,10-tetraazacyclododecane-1,4,7,10-tetrayl)tetraacetic acid (DOTA) to form cFLFLFK-PEG-DOTA. After 64Cu labeling, Positron Emission Tomography (PET) imaging was successfully able to detect mouse lung inflammation.  相似文献

Stereochemistry limits but also defines proteins, as conformational constructs stereospecific for poly-L structure. Employed as a variable in sequence, stereochemistry could make proteins customizable in the letters of L and D amino acid alphabet. In proof of concept, we previously demonstrated stereochemical reengineering of canonical beta-hairpins as bracelet and boat shaped molecules. Illustrating the prospect for functional design, a 20-residue four-stranded mini-beta protein is now customized stereochemically as a canoe shaped molecule. A conformational construct of four side by side hydrogen-bonded strands in alternately (l)beta, (d)beta conformation, joined via Type-II/II' beta-turns, is planned to be preponderantly apolar in beta-sheet favoring residues, interspersing two ion pairs, and suitably L and D in sequence. Synthesis followed by MD, NMR, CD, and MALDI-MS studies established the molecule as a canoe shaped fold in water, demonstrable in affinity of alkali and alkaline-earth metal ions as expected given its catgrip like elements. Another success in accomplishing a synthetic miniprotein complex in stereochemistry and stereospecific in conformation, exceptionally small yet functional in metal ion affinity, affirms the value in combined L and D alphabet in programming molecular shapes and functions stereochemically.  相似文献

The study described patterns of leaf dry mass change, leaf mass per area (LMA), relative growth rate and leaf life span (LL) for 14 evergreen and 7 deciduous species of a tropical forest of Southern Assam, India. Leaf expansion in both the groups was, in general, completed before June (i.e. well before the onset of monsoon rains). Although leaf dry mass during leaf initiation phase was significantly higher (P < 0.01) in evergreen species than in deciduous species, at the time of full leaf expansion, average leaf dry mass relative to the peak leaf dry mass, realised by the evergreen species was lower (66 %) than for deciduous species (76 %). Leaf dry mass increase in both groups continued after leaf full expansion. Evergreen species had a longer leaf dry mass steady phase than deciduous species (2–6 vs 2–3 months). Average LMA of mature leaves for evergreen species (77.43 g m?2) was significantly greater than that of deciduous species (48.43 g m?2). LL ranged from 165 days in Gmelina arborea (deciduous) to 509 days in Dipterocarpus turbinatus (evergreen). LMA was correlated positively with LL, indicating that evergreen species with higher leaf construction cost retain leaves for longer period to pay back. The average leaf dry mass loss before leaf shedding was greater (P < 0.01) for deciduous species (30.29 %) than for evergreen species (18.31 %). Although the cost of leaf construction in deciduous species was lower than for evergreen species, they replace leaves at a faster rate. Deciduous species perhaps compensate the cost involved in faster leaf replacement through higher reabsorption of dry mass during senescence, which they remobilise to initiate growth in the following spring when soil resources remain limiting.  相似文献

Analysis of wildfire extinguishment can help to identify the relative contribution of weather and management to the prevention of fire spread. Here we examine the role of weather, previous fire scars and other fuel interruptions at stopping the spread of nine large (mean 90 000 ha) late dry season fires in Arnhem Land, in the tropical savannas of northern Australia. Daily spread was mapped using Moderate‐resolution Imaging Spectroradiometer (MODIS) satellite imagery with a resolution of 250 m. We sampled points along the boundary of the fires and 1 km inside the boundary and compared conditions between the two sets. Using a combination of binomial regression and regression tree analysis, we found that recent burn scars (from the same year) were very effective at stopping fires. Where there was any recent burning within 500 m of a point, there was a 92% likelihood that it was a boundary. Interruptions such as roads, rivers and topography had small but significant effects. Vegetation type and vegetation greenness also had minor effects. Weather had a small effect via wind speed. This minor role of weather was reinforced by the fact that on most days the fires were both spreading and stopping at different parts of their perimeter. In these savannas, the weather in the late dry season is relatively invariant and is probably always conducive to some degree of fire spread. Here, interruptions to the fuel are critical to stopping fires. Nevertheless, for approximately half of boundary cases, the cause of stopping was not clear. This is probably due to the coarse scale of the analysis that does not reflect fine patterns of fuel arrangements.  相似文献

The frontal cortex might play an important role in the fear network, and white matter (WM) integrity could be related to the pathophysiology of panic disorder (PD). A few studies have investigated alterations of WM integrity in PD. The aim of this study was to determine frontal WM integrity differences between patients with PD without comorbid conditions and healthy control (HC) subjects by using diffusion tensor imaging. Thirty-six patients with PD who had used medication within 1 week and 27 age- and sex-matched HC subjects participated in this study. Structural brain magnetic resonance imaging was performed on all participants. Panic Disorder Severity Scale and Beck Anxiety Inventory (BAI) scores were assessed. Tract-based spatial statistics (TBSS) was used for image analysis. TBSS analysis showed decreased fractional anisotropy (FA) in frontal WM and WM around the frontal lobe, including the corpus callosum of both hemispheres, in patients with PD compared to HC subjects. Moreover, voxel-wise correlation analysis revealed that the BAI scores for patients with PD were positively correlated with their FA values for regions showing group differences in the FA of frontal WM of both hemispheres. Altered integrity in frontal WM of patients with PD without comorbid conditions might represent the structural pathophysiology in these patients, and these changes could be related to clinical symptoms of PD.  相似文献

Mechanisms that control nuclear membrane remodeling are essential to maintain the integrity of the nucleus but remain to be fully defined. Here, we identify a phosphatidic acid (PA)–binding capacity in the nuclear envelope (NE)–specific ESCRT, Chm7, in budding yeast. Chm7’s interaction with PA-rich membranes is mediated through a conserved hydrophobic stretch of amino acids, which confers recruitment to the NE in a manner that is independent of but required for Chm7’s interaction with the LAP2-emerin-MAN1 (LEM) domain protein Heh1 (LEM2). Consistent with the functional importance of PA binding, mutation of this region abrogates recruitment of Chm7 to membranes and abolishes Chm7 function in the context of NE herniations that form during defective nuclear pore complex (NPC) biogenesis. In fact, we show that a PA sensor specifically accumulates within these NE herniations. We suggest that local control of PA metabolism is important for ensuring productive NE remodeling and that its dysregulation may contribute to pathologies associated with defective NPC assembly.  相似文献

This article describes a large-scale model of turtle visual cortex that simulates the propagating waves of activity seen in real turtle cortex. The cortex model contains 744 multicompartment models of pyramidal cells, stellate cells, and horizontal cells. Input is provided by an array of 201 geniculate neurons modeled as single compartments with spike-generating mechanisms and axons modeled as delay lines. Diffuse retinal flashes or presentation of spots of light to the retina are simulated by activating groups of geniculate neurons. The model is limited in that it does not have a retina to provide realistic input to the geniculate, and the cortex and does not incorporate all of the biophysical details of real cortical neurons. However, the model does reproduce the fundamental features of planar propagating waves. Activation of geniculate neurons produces a wave of activity that originates at the rostrolateral pole of the cortex at the point where a high density of geniculate afferents enter the cortex. Waves propagate across the cortex with velocities of 4 m/ms to 70 m/ms and occasionally reflect from the caudolateral border of the cortex.  相似文献

Proline tRNA 3′-maturation in Escherichia coli occurs through a one-step RNase E endonucleolytic cleavage immediately after the CCA determinant. This processing pathway is distinct from the 3′-end maturation of the other tRNAs by avoiding the widespread use of 3′ → 5′ exonucleolytic processing, 3′-polyadenylation and subsequent degradation. Here, we show that the cytosine (C) at the mature 5′-terminus of the proK and proL tRNAs is required for both the RNase E cleavage immediately after the CCA determinant and their functionality. Thus, changing the C nucleotide at the mature 5′-terminus of the proL and proK tRNAs to the more common G nucleotide led to RNase E cleavages 1–4 nucleotides downstream of the CCA determinant. Furthermore, the 5′-modified mutant tRNAs required RNase T and RNase PH for their 3′-maturation and became substrates for polyadenylation and degradation. Strikingly, the aminoacylation of the 5′-modified proline tRNAs was blocked due to the change in the recognition element for prolyl-tRNA-synthetase. An analogous modification of the pheV 5′-mature terminus from G to C nucleotide did not support cell viability. This result provides additional support for the importance of first nucleotide of the mature tRNAs in their processing and functionality.  相似文献
