Even with the completion of a draft version of the human genome sequence only a fraction of the genes identified from this sequence have known functions. Chromosomal engineering in mouse cells, in concert with gene replacement assays to prove the functional significance of a given genomic region or gene, represents a rapid and productive means for understanding the role of a given set of genes. Both techniques rely heavily on detailed maps of chromosomal regions, initially to understand the scope of the regions being modified and finally to provide the cloned resources necessary to allow both finished sequencing and large insert complementation. This report describes the creation of a BAC clone contig on mouse chromosome 11 in a region showing conservation of synteny with sequences on human chromosome 17. We have created a detailed map of an approximately 3-cM region containing at least 33 genes through the use of multiple BAC mapping strategies, including chromosome walking and multiplex oligonucleotide hybridization and gap filling. The region described is one of the targets of a large effort to create a series of mice with regional deletions on mouse chromosome 11 (33-80 cM) that can subsequently be subjected to further mutagenesis. 相似文献

Changes in the cover and density of shrubs and trees were assessed from aerial photographs (1940, 1974 and 1998) as well as from fixed-point photographs taken in 1984 and 1996 in the Kruger National Park, South Africa. Woody cover (trees and shrubs combined) increased by 12% on granite substrates but decreased by 64% on basalt substrates over the past 58 years. Both these figures are expressed in terms of the initial values, respectively. The density of the large tree component of woody vegetation decreased on both substrates. Woody vegetation cover declined as fire return periods became shorter, but the relationship was weak. The increases in woody plant density and cover on granite are thought to be the result of decreased competition from grasses, which in turn is a result of overgrazing by wild herbivores whose numbers have been kept high through the provision of surface water. These effects were not seen on the relatively nutrient-rich basalts, where grasses can recover rapidly even after heavy grazing. The decline in overall woody cover on basalts is interpreted as a result of regular, short-interval prescribed burning over the past 40 years, while the universal decline in large trees seems to result from an interaction between regular, frequent fires and utilization by elephants. Background
Anemia affects over 500 million women, and in pregnancy is associated with impaired maternal and infant outcomes. Intermittent antenatal iron supplementation is an attractive alternative to daily dosing; however, the impact of this strategy on infant outcomes remains unclear. We compared the effect of intermittent antenatal iron supplementation with daily iron supplementation on maternal and infant outcomes in rural Viet Nam.Methods and Findings
This cluster randomised trial was conducted in Ha Nam province, Viet Nam. 1,258 pregnant women (<16 wk gestation) in 104 communes were assigned to daily iron–folic acid (IFA), twice weekly IFA, or twice weekly multiple micronutrient (MMN) supplementation. Primary outcome was birth weight. Mean birth weight was 3,148 g (standard deviation 416). There was no difference in the birth weights of infants of women receiving twice weekly IFA compared to daily IFA (mean difference [MD] 28 g; 95% CI −22 to 78), or twice weekly MMN compared to daily IFA (MD −36.8 g; 95% CI −82 to 8.2). At 32 wk gestation, maternal ferritin was lower in women receiving twice weekly IFA compared to daily IFA (geometric mean ratio 0.73; 95% CI 0.67 to 0.80), and in women receiving twice weekly MMN compared to daily IFA (geometric mean ratio 0.62; 95% CI 0.57 to 0.68), but there was no difference in hemoglobin levels. Infants of mothers who received twice weekly IFA had higher cognitive scores at 6 mo of age compared to those who received daily IFA (MD 1.89; 95% CI 0.23 to 3.56).Conclusions
Twice weekly antenatal IFA or MMN did not produce a clinically important difference in birth weight, when compared to daily IFA supplementation. The significant improvement in infant cognitive outcomes at 6 mo of age following twice weekly antenatal IFA requires further exploration, and provides additional support for the use of intermittent, rather than daily, antenatal IFA in populations with low rates of iron deficiency.Trial registration

Background
There are no data on the metabolic consequences of post-term birth (≥42 weeks gestation). We hypothesized that post-term birth would adversely affect insulin sensitivity, as well as other metabolic parameters and body composition in childhood.Methods
77 healthy pre-pubertal children, born appropriate-for-gestational-age were studied in Auckland, New Zealand: 36 born post-term (18 boys) and 41 (27 boys) born at term (38–40 weeks gestation). Primary outcome was insulin sensitivity measured using intravenous glucose tolerance tests and Bergman’s minimal model. Other assessments included fasting hormone concentrations and lipid profiles, body composition from whole-body dual-energy X-ray absorptiometry, 24-hour ambulatory blood pressure monitoring, and inflammatory markers.Results
Insulin sensitivity was 34% lower in post-term than in term children (7.7 vs. 11.6 x10-4·min-1·(mU/l); p<0.0001). There was a compensatory increase in acute insulin response among post-term children (418 vs 304 mU/l; p=0.037), who also displayed lower glucose effectiveness than those born at term (2.25 vs 3.11 x10-2·min-1; p=0.047). Post-term children not only had more body fat (p=0.014) and less fat-free mass (p=0.014), but also had increased central adiposity with more truncal fat (p=0.017) and greater android to gynoid fat ratio (p=0.007) compared to term controls. Further, post-term children displayed other markers of the metabolic syndrome: lower normal nocturnal systolic blood pressure dipping (p=0.027), lower adiponectin concentrations (p=0.005), as well as higher leptin (p=0.008) and uric acid (p=0.033) concentrations. Post-term boys (but not girls) also displayed a less favourable lipid profile, with higher total cholesterol (p=0.018) and LDL-C (p=0.006) concentrations, and total cholesterol to HDL-C ratio (p=0.048).Conclusions
Post-term children have reduced insulin sensitivity and display a number of early markers of the metabolic syndrome. These findings could have important implications for the management of prolonged pregnancies. Background
Goa is a coastal state in India and salt making is being practiced for many years. This investigation aimed in determining the culturable haloarchaeal diversity during two different phases of salt production in a natural solar saltern of Ribandar, Goa. Water and sediment samples were collected from the saltern during pre-salt harvesting phase and salt harvesting phase. Salinity and pH of the sampling site was determined. Isolates were obtained by plating of the samples on complex and synthetic haloarchaeal media. Morphology of the isolates was determined using Gram staining and electron microscopy. Response of cells to distilled water was studied spectrophotometrically at 600nm. Molecular identification of the isolates was performed by sequencing the 16S rRNA.Results
Salinity of salt pans varied from 3-4% (non-salt production phase) to 30% (salt production phase) and pH varied from 7.0-8.0. Seven haloarchaeal strains were isolated from water and sediment samples during non-salt production phase and seventeen haloarchaeal strains were isolated during the salt production phase. All the strains stained uniformly Gram negative. The orange-red acetone extract of the pigments showed similar spectrophotometric profile with absorption maxima at 393, 474, 501 and 535 nm. All isolates obtained from the salt dilute phase were grouped within the genus Halococcus. This was validated using both total lipid profiling and 16S rRNA data sequencing. The isolates obtained from pre-salt harvesting phase were resistant to lysis. 16S rRNA data showed that organisms belonging to Halorubrum, Haloarcula, Haloferax and Halococcus genera were obtained during the salt concentrated phase. The isolates obtained from salt harvesting phase showed varied lysis on suspension in distilled water and /or 3.5% NaCl.Conclusion
Salterns in Goa are transiently operated during post monsoon season from January to May. During the pre-salt harvesting phase, all the isolates obtained belonged to Halococcus sp. During the salt harvesting phase, isolates belonging to Halorubrum, Haloarcula, Haloferax and Halococcus genera were obtained. SUBSIDY AND STRESS RESPONSES OF STREAM PERIPHYTON TO GRADIENTS IN WATER VELOCITY AS A FUNCTION OF COMMUNITY GROWTH FORM 总被引:1,自引:0,他引:1
Previous studies have shown major differences in the way biomass of stream periphyton is controlled by spatial variations in velocity. We hypothesize that these differences may be the result of different growth forms within the community. Some dense and coherent growth forms (e.g. mucilaginous diatom/cyanobacterial mats) may be resistant to diffusion and also resistant to dislodgment by shear stress. Higher velocities applied to such communities could therefore be expected to enhance biomass accrual by increasing rates of mass transfer, but without greatly increasing losses through sloughing. Conversely, other growth forms (e.g. long filamentous green algae) have an open matrix, and high rates of diffusion into the mats can potentially occur even at low velocity. However, as velocities increase, high skin friction and form drag should lead to higher rates of sloughing. The overall result of these processes should be that maximum biomass occurs at low velocities. This “subsidy-stress” hypothesis was tested twice with each of three different periphytal growth forms: a coherent, mucilaginous, diatom community; a moderately coherent, stalked/ short, filamentous diatom community; and an open-weave, long, filamentous green algal community. A monotonic increase in chl a biomass occurred as a function of near-bed velocities for the first of the two mucilaginous diatom communities investigated. No biomass-velocity relationship was found, however, with the second mucilaginous community, probably because the waters were highly enriched and mass transfer driven by molecular diffusion was probably high throughout the velocity gradient. Biomass was moderate at low velocities, peaked at near-bed velocities from 0.18 to 0.2 m·s?1 (~0.40–0.45 m·s?1 mean column velocity), and then decreased at higher velocities in both of the stalked/ short filament communities of diatoms analyzed. With the long filamentous green algal communities, a monotonic reduction in biomass occurred as a function of increases in velocity. Proliferations greater than 100 mg·m?2 chl a occurred at low near-bed velocities (i.e. <0.2 m·s?1), after which biomass declined nearly exponentially as a function of increasing velocity to less than 10 mg·m?2 chl a at velocities greater than 0.4 m·s?1. These biomass-velocity trends support our hypothesis that community growth form determines periphytal responses to spatial variations in velocity within stream reaches. 相似文献
