A new pond complex, designed to enhance aquatic biodiversity, was monitored over a 7-year period. The Pinkhill Meadow site, located in grassland adjacent to the R. Thames, proved unusually rich in terms of its macrophyte, aquatic macroinvertebrate and wetland bird assemblages. In total, the 3.2 ha mosaic of ca. 40 permanent, semi-permanent and seasonal ponds and pools was colonized by approximately 20% of all UK wetland plant and macroinvertebrate species over the 7-year survey period. This included eight invertebrate species that are Nationally Scarce in the UK. The site supported three breeding species of wading bird and was used by an additional 54 species of waders, waterfowl and other wetland birds. The results from four monitoring ponds investigated in more detail showed that these ponds significantly supported more plant and macroinvertebrate species than both minimally impaired UK reference ponds, and other new ponds for which compatible data were available. Comparisons of the physico-chemical, hydrological and land-use characteristics of the Pinkhill pools with those of other new ponds showed that the site was unusual in having a high proportion of wetlands in the near surrounds. It also had significantly lower water conductivity than other ponds and a higher proportion of (non-woodland) semi-natural land in its surroundings. Given that ponds are known to contribute significantly to UK biodiversity at a landscape level, and that several thousand new ponds are created each year in the UK alone, the findings suggest that well designed and located pond complexes could be used to significantly enhance freshwater biodiversity within catchments. Background:

Anemia is an important public health and clinical problem. Observational studies have linked iron deficiency and anemia in children with many poor outcomes, including impaired cognitive development; however, iron supplementation, a widely used preventive and therapeutic strategy, is associated with adverse effects. Primary-school–aged children are at a critical stage in intellectual development, and optimization of their cognitive performance could have long-lasting individual and population benefits. In this study, we summarize the evidence for the benefits and safety of daily iron supplementation in primary-school–aged children.

Methods:

We searched electronic databases (including MEDLINE and Embase) and other sources (July 2013) for randomized and quasi-randomized controlled trials involving daily iron supplementation in children aged 5–12 years. We combined the data using random effects meta-analysis.

Results:

We identified 16 501 studies; of these, we evaluated 76 full-text papers and included 32 studies including 7089 children. Of the included studies, 31 were conducted in low- or middle-income settings. Iron supplementation improved global cognitive scores (standardized mean difference 0.50, 95% confidence interval [CI] 0.11 to 0.90, p = 0.01), intelligence quotient among anemic children (mean difference 4.55, 95% CI 0.16 to 8.94, p = 0.04) and measures of attention and concentration. Iron supplementation also improved age-adjusted height among all children and age-adjusted weight among anemic children. Iron supplementation reduced the risk of anemia by 50% and the risk of iron deficiency by 79%. Adherence in the trial settings was generally high. Safety data were limited.

Interpretation:

Our analysis suggests that iron supplementation safely improves hematologic and nonhematologic outcomes among primary-school–aged children in low- or middle-income settings and is well-tolerated.An estimated 25% of school-aged children worldwide are anemic.1 Iron deficiency is thought to account for about half of the global cases of anemia2 and is associated with inadequate dietary iron and, in developing settings, hookworm and schistosomiasis.3 In developed settings, anemia is prevalent among disadvantaged populations, including newly arrived refugees, indigenous people4 and some ethnic groups (e.g., Hispanic people in the United States).5,6 About 3% of primary-school–aged children in Canada are anemic.7 Programs to address anemia are constrained by concerns that iron supplements cause adverse effects, including an increased risk of infections such as malaria in endemic areas.8In observational studies, iron deficiency has been associated with impaired cognitive and physical development. It has been estimated that each 10 g/L decrement in hemoglobin reduces future intelligence quotient (IQ) by 1.73 points.9 However, observational data are susceptible to confounding,10 and a causal relation between iron deficiency and cognitive impairment has not been confirmed.11 Randomized controlled trials should overcome confounding, but results of trials examining this question have not agreed.Optimizing cognitive and physical development in primary-school–aged children could have life-long benefits.12 However, anemia-control recommendations must balance safety and efficacy. We performed a systematic review of the effects of daily iron supplementation, a commonly used strategy to combat anemia,2 in primary-school–aged children. In spite of their potential use as indicators of both present and past environmental conditions, little is known about the diatom communities in the many small water bodies at high altitudes in New Zealand. We sampled benthic diatoms at 20 sites in a typical subalpine mire pool/tarn complex near Arthur’s Pass in South Island, New Zealand in the austral spring 2001. The aims were to characterise the diatom communities, including identification of a possible endemic component, and to investigate relationships with environmental variables. The community at genus level was consistent with the peat-bog diatom flora reported from elsewhere except for the common occurrence of the Tasmania/New Zealand endemic genus Eunophora. At the species level, 27 of the 52 most common taxa appear to correspond to known species from the Northern Hemisphere and are therefore presumed to be cosmopolitan in their distribution. Just two taxa are known from the Southern Hemisphere only, however identification of the remaining common species proved problematic. Analysis using the BIO-ENV procedure of the PRIMER computer program confirmed an expected strong association between diatom community composition and pH, with water conductivity and gilvin also important. Weighted averaging regression and cross-validation using C2 software enabled selection of four diatom species as potentially sensitive indicators of certain pH levels. Neither species of Eunophora showed a strong preference for pH or for any of the other environmental variables measured, indicating that other factors are determining their distributions. The strength of the species–environment relationships found in this small survey suggests good potential for monitoring current conditions and for palaeoecological applications. Extension of the dataset with information from other alpine/subalpine areas is desirable, as is the compilation of a regional diatom identification guide for these habitats.  相似文献

Recessive resistance to Bacillus thuringiensis (Bt) cotton, Gossypium hirsutum L., in laboratory-selected strains of pink bollworm, Pectinophora gossypiella (Saunders), is associated with three resistance alleles (r1, r2, and r3) of a cadherin gene. Previous experiments based on measurement of fitness components in Bt-resistant and Bt-susceptible strains revealed that fitness costs and incomplete resistance are associated with resistance. Here, we used two hybrid strains of pink bollworm, each containing a mixture of susceptible and resistant individuals, and polymerase chain reaction (PCR) amplifications to test the association between cadherin genotype and fitness components for individuals sharing a common genetic background. All survivors on Bt cotton had two r alleles, confirming that recessive cadherin alleles are tightly linked with resistance to Bt cotton. On non-Bt cotton, significantly greater developmental time for rr than ss larvae indicated a recessive fitness cost, but costs did not affect survival or pupal weight. Incomplete resistance was manifested as longer developmental time, lower survival, and smaller pupal weight in rr individuals developing on Bt cotton compared with non-Bt cotton. As in previous experiments, no significant variation in performance on Bt cotton was detected among rr genotypes. However, a meta-analysis of data from seven experiments revealed that survival on Bt cotton relative to non-Bt cotton was lower in r2r3 and higher in r1r2 compared with the other rr genotypes. Assessment of fitness components associated with cadherin genotypes in hybrid strains of pink bollworm confirms that recessive resistance to Bt cotton is associated with recessive fitness costs and incomplete resistance.  相似文献

Proteins are one of the major metabolites in biomass from microalgae that constitute the diet of marine organisms grown in aquaculture, and are essential for their growth. The quantity of this component is influenced by nutrients, temperature and light intensity, among others. We examined the growth, biomass production and protein of Chaetoceros muelleri with two sources of nitrogen (nitrate and urea) at three concentrations, using the medium f/2 (0.88 mol/L) (nitrates) as control. The treatments were the medium 2f (3.53 mol/L) and 4f (7.05 mol/L) with NO3-, and the medium f/2 (0.88 mol/L), 2f (3.53mol/L) and 4f (7.05 mol/L) with urea. In general, the productive parameters were greater using urea than nitrate in the media. Higher cell concentrations (2.83 x 106 cell/mL), average and cumulative growth rates (1.50 div/day and 6.01 divisions), dry weight (0.0044 g/L), and proportion of proteins (23.74%) were found when urea was used as the N source. However, most of the bands on the electrophoretic profile were present in the mediums with NO3- (~6.5 to 90 kDa).  相似文献

Two strains of pink bollworm, Pectinophora gossypiella (Saunders), each derived in 1997 from a different field population, were selected for resistance to Bacillus thuringiensis (Bt) toxin Cry1Ac in the laboratory. One strain (MOV97-R) originated from Mohave Valley in western Arizona; the other strain (SAF97-R) was from Safford in eastern Arizona. Relative to a susceptible laboratory strain, Cry1Ac resistance ratios were 1700 for MOV97-R and 520 for SAF97-R. For the two resistant strains, larval survival did not differ between non-Bt cotton and transgenic cotton producing CrylAc. In contrast, larval survival on Bt cotton was 0% for the two unselected parent strains from which the resistant strains were derived. Previously identified resistance (r) alleles of a cadherin gene (BtR) occurred in both resistant strains: r1 and r3 in MOV97-R, and r1 and r2 in SAF97-R. The frequency of individuals carrying two r alleles (rr) was 1.0 in the two resistant strains and 0.02 in each of the two unselected parent strains. Furthermore, in two hybrid strains with a mixture of susceptible (s) and r alleles at the BtR locus, all survivors on Bt cotton had two r alleles. The results show that resistance to Cry1Ac-producing Bt cotton is associated with recessive r alleles at the BtR locus in the strains of pink bollworm tested here. In conjunction with previous results from two other Bt-resistant strains of pink bollworm (APHIS-98R and AZP-R), results reported here identify the cadherin locus as the leading candidate for molecular monitoring of pink bollworm resistance to Bt cotton.  相似文献

Growth under conditions of oxygen restriction results in a generalized decrease in the definition of the mitochondrial membranes, a decrease in the mitochondrial cytochromes, and a decrease in citric acid cycle enzymes of the obligate aerobic yeast Candida parapsilosis. Addition of unsaturated fatty acids and ergosterol to cultures exposed to limited oxygen results in improved definition of the mitochondrial membranes and an increase in the total mitochondrial cytochrome content of the cells. Euflavine completely inhibits mitochondrial protein synthesis in vitro. Its in vivo effect is to cause the formation of giant mitochondrial profiles with apparently intact outer membranes and modified internal membranes; the cristae (in-folds) appear only as apparently disorganized remnants while the remainder of the inner membrane seems intact. Cytochromes a, a3, b, and c1 are not synthesized by the cells in the presence of euflavine. Ethidium appears to have effects identical to those of euflavine, whereas chloramphenicol, lincomycin, and erythromycin have similar effects in principle but they are less marked. The effects of all the inhibitors are freely reversible after removal of the drugs. The results are discussed in terms of a functionally three-membrane model of the mitochondrion. In addition, the phylogenetic implications of the observed differences between this organism and the facultative anaerobic yeasts are considered.  相似文献

Background

Deficiency of 3-hydroxy-isobutyryl-CoA hydrolase (HIBCH) caused by HIBCH mutations is a rare cerebral organic aciduria caused by disturbance of valine catabolism. Multiple mitochondrial respiratory chain (RC) enzyme deficiencies can arise from a number of mechanisms, including defective maintenance or expression of mitochondrial DNA. Impaired biosynthesis of iron-sulphur clusters and lipoic acid can lead to pyruvate dehydrogenase complex (PDHc) deficiency in addition to multiple RC deficiencies, known as the multiple mitochondrial dysfunctions syndrome.

Methods

Two brothers born to distantly related Pakistani parents presenting in early infancy with a progressive neurodegenerative disorder, associated with basal ganglia changes on brain magnetic resonance imaging, were investigated for suspected Leigh-like mitochondrial disease. The index case had deficiencies of multiple RC enzymes and PDHc in skeletal muscle and fibroblasts respectively, but these were normal in his younger brother. The observation of persistently elevated hydroxy-C4-carnitine levels in the younger brother led to suspicion of HIBCH deficiency, which was investigated by biochemical assay in cultured skin fibroblasts and molecular genetic analysis.

Results

Specific spectrophotometric enzyme assay revealed HIBCH activity to be below detectable limits in cultured skin fibroblasts from both brothers. Direct Sanger sequence analysis demonstrated a novel homozygous pathogenic missense mutation c.950G <A; p.Gly317Glu in the HIBCH gene, which segregated with infantile-onset neurodegeneration within the family.

Conclusions

HIBCH deficiency, a disorder of valine catabolism, is a novel cause of the multiple mitochondrial dysfunctions syndrome, and should be considered in the differential diagnosis of patients presenting with multiple RC deficiencies and/or pyruvate dehydrogenase deficiency.

In this ‘perspectives’ article, we share experiences gained from the century-old Kruger National Park (KNP) in South Africa to illustrate the dynamic complexity of biophysical and socio-political systems, the interactions that occur between them, and the consequences for ecosystem-scale functions and resources and for their management. As in KNP, the social-ecological milieu surrounding many national parks and protected areas is changing rapidly. There will be significant managerial adjustments as human populations grow and the needs for resources accelerate. The changes, driven largely by global-scale environmental shifts as well as by new knowledge, are intimately intertwined with evolving societal perceptions, values, and expectations. Many KNP resource-related issues of the past century originated more internally and were largely environmental, whereas the emerging issues are more external and largely social. Here, we illustrate how interrelated scientific and managerial advances in integrating biophysical and social systems are acting to conserve and rehabilitate resources within KNP, and to aid in their conservation. Where appropriate, we relate these advances to similar examples in the region or other protected areas in the world. Strategies to address emerging issues are identified and discussed—and their combined effects on resource conservation and management are evaluated. In our experience the approach to conservation within KNP has been successful, despite well-intended but damaging management actions in the past. We believe that the perceived success stems from a willingness to continually incorporate new knowledge into management, to foster close working and personal associations among scientists, managers, and rangers, to acquire an intimate knowledge and understanding of the social-ecological system by the administrators as well as by the staff, and to be actively ‘forward’ thinking in an increasingly complex and uncertain world. We accept that many decisions taken today will be challenged by future managers and scientists, and we expect that some will be found wanting as emerging knowledge and continued learning shape future decisions. Further, evolving political, social, and environmental contexts may mean that protected areas will need to be managed in different ways. Therefore, we emphasize the importance of minimizing the permanency and impact of decisions so that today’s actions do not compromise future decisions when meaningful changes need to be made.
