Switchgrass (Panicum virgatum L.) is a warm perennial grass with valuable characteristics as a biofuel crop. To avoid competition with food crops, biofuel crops will be likely relegated to less productive soils such as marginal lands. Consequently, the salinity and water scarcity problems that commonly affect marginal lands compromise biofuel crop germination, emergence, and seedling establishment. The aims of this study were to study the germination and seedling growth of switchgrass under salinity and water stress and to describe the morpho-anatomical responses of the roots and leaves in the seedlings to these stress conditions. The effect of salt and water stress was assessed using sodium chloride (NaCl) and polyethylene glycol (PEG) 8000 at the same water potentials of ??0.8, ??1.0, and ??1.2 MPa. Seeds were moist prechilled for 7 days at 5 °C and germinated at 30/15 °C (8 h light/16 h dark). NaCl treatments (??0.8 and ??1.0 MPa) delayed germination rates but did not reduce the final germination percentage, whereas at a lower potential (??1.2 MPa), the final germination percentage was diminished. The effects of PEG (??1.0 and ??1.2 MPa) on the germination rate and final percentage were more detrimental than those induced by isosmotic concentrations of NaCl. PEG and NaCl reduced significantly the vigor index of ??0.8 to ??1.2 MPa. The morpho-anatomical changes such as the reduction in the root cross-sectional area and the thickening of the endodermis walls for both stress conditions and aerenchyma formation in the cortex under salinity could significantly contribute in the survival and tolerance during the early seedling stages. 相似文献

Purinergic receptors are widespread in the human organism and are involved in several physiological functions like neurotransmission, nociception, platelet aggregation, etc. In the immune system, they may regulate the expression and release of pro-inflammatory factors as well as the activation and death of several cell types. It is already described the participation of some purinergic receptors in the inflammation and pathological processes, such as a few neglected tropical diseases (NTDs) which affect more than 1 billion people in the world. Although the high social influence those diseases represent endemic countries, most of them do not have an efficient, safe or affordable drug treatment. In that way, this review aims to discuss the current literature involving purinergic receptor and immune response to NTDs pathogens, which may contribute in the search for new therapeutic possibilities. 相似文献

Studies in knockout mice support the involvement of alcohol dehydrogenases ADH1 and ADH4 in retinoid metabolism, although kinetics with retinoids are not known for the mouse enzymes. Moreover, a role of alcohol dehydrogenase (ADH) in the eye retinoid interconversions cannot be ascertained due to the lack of information on the kinetics with 11-cis-retinoids. We report here the kinetics of human ADH1B1, ADH1B2, ADH4, and mouse ADH1 and ADH4 with all-trans-, 7-cis-, 9-cis-, 11-cis- and 13-cis-isomers of retinol and retinal. These retinoids are substrates for all enzymes tested, except the 13-cis isomers which are not used by ADH1. In general, human and mouse ADH4 exhibit similar activity, higher than that of ADH1, while mouse ADH1 is more efficient than the homologous human enzymes. All tested ADHs use 11-cis-retinoids efficiently. ADH4 shows much higher k(cat)/K(m) values for 11-cis-retinol oxidation than for 11-cis-retinal reduction, a unique property among mammalian ADHs for any alcohol/aldehyde substrate pair. Docking simulations and the kinetic properties of the human ADH4 M141L mutant demonstrated that residue 141, in the middle region of the active site, is essential for such ADH4 specificity. The distinct kinetics of ADH4 with 11-cis-retinol, its wide specificity with retinol isomers and its immunolocalization in several retinal cell layers, including pigment epithelium, support a role of this enzyme in the various retinol oxidations that occur in the retina. Cytosolic ADH4 activity may complement the isomer-specific microsomal enzymes involved in photopigment regeneration and retinoic acid synthesis. 相似文献

We have identified a novel mammalian gene, termed nicolin 1 gene (NICN1), that is present in human, dog and mouse, whereas it is absent from the available genome sequences of nonmammalian organisms. The NICN1 gene consists of six exons and spans about 6 kb of genomic DNA. It encodes a 213 amino acid protein that does not belong to any known protein family. Experiments using green fluorescent protein (GFP)-tagged nicolin 1 fusion proteins indicate that nicolin 1 is a nuclear protein. Northern analysis and semiquantitative RT-PCR demonstrated that the 2.5 kb NICN1 mRNA is expressed in a tissue-specific manner. The highest NICN1 expression levels are found in brain, testis, liver, and kidney. On the other hand the NICN1 expression is weak in spleen, leukocytes, small intestine and colon. The sodium salt of n-butyric acid was found to inhibit the growth of asynchronous cultures of rabbit articular chondrocytes. This inhibitory effect was dose-dependent between 1 mM and 5 mM, reversible, and accompanied by volume enhancement and modification of cellular morphology. Flow-cytometric analysis showed that drug exposure led to a slowing-down of the cell-cycle progression; after 1 day's exposure, cells accumulated in G1, and after 2 or 3 days' treatment, in G2, without a blockage in M; the increase of cells in G2 was in fact due to an enhancement of binculeated cells. The treated cells had an increased RNA content. Articular chondrocytes seem to be target cells for sodium butyrate and therefore it represents a valuable biological tool for studying the mechanisms of their growth regulation. 相似文献

The European grapevine moth, Lobesia botrana (Denis & Schiffermüller) (Lepidoptera: Tortricidae), is a major grapevine pest in Europe. The larva is polyphagous and able to develop on more than 25 plant species, several of them being more suitable than Vitaceae for the fitness of L. botrana. Larvae normally eat the pulp of the berry, but may also consume the seeds according to the development stage of the berry and the larval density per bunch. Understanding the effect on individual fitness of such feeding behaviour is important to assess how suitable the different berry tissues are for this insect. We offered to the larvae either entire berries, seeds, or pulp with skin of the variety Vitis vinifera cv. ‘Cabernet Sauvignon’ as larval food in order to assess several life history traits from egg hatching to adult death. Two control groups were raised on semisynthetic diets, offering a low (no plant material) or a higher (with corn flour) nutritive value medium. The larvae performed differently when fed on diets containing different parts of berries. The larvae fed on a diet containing grape berry seeds had a prolonged development time and showed higher mortality. The females emerging from these larvae had a lower fecundity and mating success than the females emerging from larvae fed on diets containing other parts of the berries. However, their longevity was greater in comparison to the other groups. We conclude that seeds of Cabernet Sauvignon are not toxic to larvae but affect the reproductive life history traits in L. botrana. 相似文献
