During a 10 day-incubation on agar surfaces at 30 degrees C, cells of the gram-negative soil bacterium Pseudomonas rhodos pass through three phases distinguishable by physiological and morphological criteria. When viewed by electron microscopy, typically "rolled" mesosomes could frequently be observed in young cells. In aged cells instead, loosely rolled or stretched-out, flattened tubules could be discerned, presumed to be degenerate mesosomes. Tubular flattened structures have been isolated from these cells by lysozyme treatment or sonication and were concentrated by differential centrifugation. Electron micrographs of these preparations showed long, straight tubules which sometimes appeared sealed at one end. Their width was 34 +/- 5 nm. They contained a lining of material, which could be digested by trypsin leaving behind an electron-transparent matrix. In rare cases, isolated tubules showed a periodic fine structure composed of ellipsoidal subunits. Optical diffraction analysis yielded a lattice consisting of subunits arranged in helices of pitch-angle 27 degrees; the unit cell dimensions were shown to be 112 X 56 A. Owing to their sensitivity to trypsin, components of the regular lattice are supposed to consist of protein. It is postulated that these protein components are layered onto a tubular membrane. These tubules are clearly distinguishable by their shape and fine structure from the periodic structure of a P. rhodos cell wall layer, which exhibits a tetragonal pattern, and also from polyheads and polysheaths of defective bacteriophages. The aim of this study was to test the method Kunos and his colleagues (1999) developed on the first rib to assess adult age-at-death. The method was applied on a sample of known age and sex, selected from a Thai collection. The procedure being subjective, we chose to estimate an age category rather than a precise age.

The results show that only 55% of the individuals are correctly classified and that the number of subjects over 60 years old is under-estimated. Three explanations are given: the modifications of the first rib related with ageing are more variable than previously stated, the technique is too subjective and difficult to apply and the morphological changes of the first rib are different between North-American and Thai people.  相似文献

In the context of ultrasound dynamic elastography imaging and characterization of venous thrombosis, we propose a method to induce mechanical resonance of confined soft heterogeneities embedded in homogenous media. Resonances are produced by the interaction of horizontally polarized shear (SH) waves with the mechanical heterogeneity. Due to such resonance phenomenon, which amplifies displacements up to 10 times compared to non-resonant condition, displacement images of the underlying structures are greatly contrasted allowing direct segmentation of the heterogeneity and a more precise measurement of displacements since the signal-to-noise ratio is enhanced. Coupled to an analytical model of wave scattering, the feasibility of shear wave induced resonance (SWIR) elastography to characterize the viscoelasticity of a mimicked venous thrombosis is demonstrated (with a maximum variability of 3% and 11% for elasticity and viscosity, respectively). More generally, the proposed method has the potential to characterize the viscoelastic properties of a variety of soft biological and industrial materials.  相似文献

The ability of some ant species (including Camponotus spp.) to forage on vertebrate urine to extract urea may extend their niche in competitive and strongly nitrogen‐limited environments. We examined the preference of Camponotus terebrans, a sand‐dwelling ant widespread in southern Australia, for baits including urine, and the duration of their foraging on those baits. We baited ants with liquid stains of urine (human and kangaroo), urea in water (2.5%. 3.5%, 7.0%, 10.0%) and sucrose in water (20% and 40%) poured directly on the ground, as well as hard baits in plots drawn on sandy soil (Kangaroo Island, South Australia). We counted individuals of this mostly nocturnal species to determine their attraction to different baits for one month. We checked plant growth on the plots after nine and 13 months. Ants collected insects and meat; they foraged for at least 29 days on stains. Ants were most numerous on 10% urea, followed by 7% urea, 3.5% urea, urine (which contains ~2.5% urea) and 2.5% urea, 40% sucrose and 20% sucrose; sucrose was less attractive to them than equimolar urea bait. Ants were attracted to human, kangaroo, and unidentified urines, and they collected bird guano. Baits and ant foraging did not affect plant recruitment in plots. We observed incidentally Camponotus consobrinus foraging on urine, which may be a common resource for this genus at the site. The remarkable ability of C. terebrans to extract nitrogen from dry sand over weeks explains partly its success on sandy soils. Foraging on urine may be an important strategy to address nitrogen limitation on sandy soils and exploit commensally niches in which hosts are kangaroos, wallabies and other vertebrates. The understanding of plant–vertebrate interactions must factor in the role of ants as commensal organisms. Such ants could also reduce greenhouse gas emissions from urine.  相似文献
