A series of acylated chitin derivatives was prepared by reacting chitin in a solution of trifluoroacetic anhydride and each of the cyclopropyl, cyclobutyl, cyclopentyl, and cyclohexyl carboxylic acids. The degree of O-acyl substitution was in a range of 1.1-1.4 depending upon the nature of the cyclic acid added, as determined by FT-IR analysis. The solubility of the products in the organic solvents of DMF and THF increased with an increase in the cyclic chain length of the carboxylic acid. Thermal gravimetric analysis indicated that the products were stable up to 220 °C for chitin cyclopropanoate and cyclobutanoate, and 250 °C for chitin cyclopentanoate and cyclohexanoate. The surface morphology of the products by scanning electron microscopic analysis revealed porous and globular surface for chitin cyclobutanoate, cyclopentanoate, and cyclohexanoate, contrast to the dense and smooth organization for the cyclopropanoate.  相似文献

The human mitochondrial chaperonin is a macromolecular machine that catalyzes the proper folding of mitochondrial proteins and is of vital importance to all cells. This chaperonin is composed of 2 distinct proteins, Hsp60 and Hsp10, that assemble into large oligomeric complexes that mediate the folding of non-native polypeptides in an ATP dependent manner. Here, we report the bacterial expression and purification of fully assembled human Hsp60 and Hsp10 recombinant proteins and that Hsp60 forms a stable tetradecameric double-ring conformation in the absence of co-chaperonin and nucleotide. Evidence of the stable double-ring conformation is illustrated by the 15 Å resolution electron microscopy reconstruction presented here. Furthermore, our biochemical analyses reveal that the presence of a non-native substrate initiates ATP-hydrolysis within the Hsp60/10 chaperonin to commence protein folding. Collectively, these data provide insight into the architecture of the intermediates used by the human mitochondrial chaperonin along its protein folding pathway and lay a foundation for subsequent high resolution structural investigations into the conformational changes of the mitochondrial chaperonin.  相似文献

Soil samples isolated from dye-contaminated sites were exploited for isolation of dye decolorizing microorganisms. A novel bacterial mixture, RkNb1, was selected based on its efficiency, showing maximum and faster decolorization of textile dyes. Seven bacterial strains were isolated and identified from the bacterial mixture as Ochrobactrum intermedium (HM480365), Ochrobactrum intermedium strain M16-10-4 (HM030758), Enterococcus faecalis (HM480367), Arthrobacter crystallopoietes (HM480368), Kocuria flavus (HM480369), Bacillus beijingensis (HM480370), and Citrobacter freundii (HM480371) by 16S rRNA gene sequence analysis. This bacterial mixture showed 98.17% decolorization of Reactive Violet 5 (400 mg L?1) within 8 h. The culture exhibited good decolorization ability at pH 8 and at a temperature of 37°C. Malt extract and peptone was found to enhance the decolorization rate of Reactive Violet 5. Plackett-Burman experimental design was used for elucidation of medium components affecting Reactive Violet 5 decolorization. Dye degradation products obtained during the course of decolorization were analyzed by high-performance thin-layer chromatography (HPTLC), Fourier transform infrared (FTIR), and nuclear magnetic resonance (NMR). The potential of this bacterial mixture to decolorize Reactive Violet 5 dye from manufacturing industry effluent is to be carried out using appropriate bioreactors.  相似文献

Hemangioma is the most common soft-tissue tumor of infancy. Despite the frequency of these vascular tumors, the origin of hemangioma-endothelial cells is unknown. Circulating endothelial progenitor cells (EPCs) have recently been identified as vascular stem cells with the capacity to contribute to postnatal vascular development. We have attempted to determine whether circulating EPCs are increased in hemangioma patients and thereby provide insight into the role of EPCs in hemangioma growth. METHODS AND RESULTS: Peripheral blood mononuclear cells (PBMCs) were isolated from hemangioma patients undergoing surgical resection (N = 5) and from age-matched controls (N = 5) undergoing strabismus correction surgery. PBMCs were stained with fluorescent-labeled antibodies for AC133, CD34, and VEGFR2/KDR. Fluorescent-labeled isotype antibodies served as negative controls. Histologic sections of surgical specimens were stained with the specific hemangioma markers Glut1, CD32, and merosin, to confirm the diagnosis of common hemangioma of infancy. EPCs harvested from healthy adult volunteers were stained with Glut1, CD32, and merosin, to assess whether cultured EPCs express known hemangioma markers. Hemangioma patients had a 15-fold increase in the number of circulating CD34 AC133 dual-staining cells relative to controls (0.78+/-0.14% vs.0.052+/-0.017%, respectively). Similarly, the number of PBMCs that stained positively for both CD34 and KDR was also increased in hemangioma patients (0.49+/-0.074% vs. 0.19+/-0.041% in controls). Cultured EPCs stained positively for the known hemangioma markers Glut1, CD32, merosin. CONCLUSIONS: This is the first study to suggest a role for EPCs in the pathogenesis of hemangioma. Our results imply that increased levels of circulating EPCs may contribute to the formation of this vascular tumor.  相似文献

A series of potent inhibitors of prolylcarboxypeptidase (PrCP) was developed by modifying a lead structure that was discovered by high-throughput screening. The tert-butyl pyrrolidine was replaced by an aminocyclopentane to reduce the metabolic liabilities of the original lead. The compounds demonstrated sub-nanomolar in vitro IC(50) values, minimal activity shifts in pure plasma and improved pharmacokinetics. Complete ex vivo plasma target engagement was achieved with low brain exposure at the 20 h time point following p.o. dosing in a mouse. The results indicate that the aminocyclopentanes are useful tools for studying the therapeutic potential of peripheral (non-CNS) PrCP inhibition.  相似文献

Three tropical range grasses (Cenchrus ciliaris, Dichanthium annulatum, and Panicum antidotale) and two range legumes [Macroptilium atropurpureum (siratro) and Stylosanthes hamata (stylo)] were grown under four irradiances, i.e. 100 (I 100, control), 75 (I 75), 50 (I 50), and 25 (I 25) % of full sunlight. Accumulation of chlorophyll (Chl) b increased but that of Chl a decreased under low irradiances. The greater accumulation of Chl (a+b) in grasses (particularly in D. annulatum and P. antidotale) under shade predicted their shade adaptability. Among legumes Stylosanthes was more adaptive to the shade than Macroptilium due to its higher accumulation of Chl (a+b). Significant difference in the accumulation of carotenoids under I 25 over I 100 was observed in all the species, which shows the increase in quality of the fodder under limited irradiance. There was a significant decrease in soluble protein content in C. ciliaris under I 75, however, no significant difference in protein content was observed under I 50 and I 25, which was also reflected in the SDS pattern with the reduction in content of polypeptides at I 75 and following increase at I 50 and I 25. This was possibly due to reduction of light-induced protein at I 75 and then expression of the stress-induced protein at further reduction of irradiance. Peroxidase activity in C. ciliaris increased with the decrease in irradiance and its isozyme pattern showed differences among all treatments, which indicated the role of different peroxidase isoforms at different irradiances.  相似文献

Objectives

To trace the critical practicing, clinical and epidemiological risk factors in bacterial load and points of intervention in spread of community-acquired methicillin resistant Staphylococcus aureus (CA-MRSA) in healthy community.

Study Design

2872 individuals with no prominent clinical features were enrolled and administered a pre-tested questionnaire prepared on the basis of outcome of a prior pilot study in same region. Swab samples from skin, throat and nasal nares were tested for MRSA and molecular identification was done to track the strains moving from hospital to community.

Methods

Swab samples from skin, throat and nasal nares were tested for MRSA culture followed by molecular characterization of isolates and antimicrobial resistance pattern. Bacterial load was estimated to better understand the burden in different categories. Statistical analysis was done using SPSS 16.0 version.

Results

History of prior infection (OR 3.9, 95% CI 1.363 – 5.793), habit of self remedy (OR 3.2, 95% CI 0.991 – 1.473) and incomplete treatment (OR 0.26, 95% CI 0.08 – 0.80) (P < 0.05 for each) were the predominant factors that contributed to spread of CA-MRSA. Increased drug resistance in CA-MRSA was observed for 4 different clones: SCCmec + IVa/PVL +, SCCmec + IVa/PVL ? and SCCmec + IVc/PVL +, SCCmec + IVc/PVL ?. Bacterial load was found significantly high in below poverty line dwellers and drug abusers (P < 0.05).

Conclusion

We identified habit of self remedy, drug abusing and incomplete treatment as practicing risk factors where interventions can be made to manage the dissemination of CA-MRSA in rural population.

Pseudomonas lurida strain M2RH3 (MTCC 9245) is a Gram negative, non spore forming, fluorescent bacterium isolated from a high altitude rhizospheric soil from the Uttarakhand Himalayas. The identity of the bacterium was arrived by sequencing of the 16S rRNA gene and subsequent phylogenetic analysis. It grew and exhibited plant growth promoting traits at 4, 15 and 30°C, under in vitro conditions. The expression of plant growth promoting (PGP) traits by the bacterium was highest at 30°C, with a proportionate reduction in PGP activity at lower temperatures. Determination of phosphate solubilization by the bacterium at three incubation temperatures revealed a steady increase in the soluble P levels across the incubation temperatures, coupled with a concomitant drop in the pH levels of the culture supernatant, till the 14th day of incubation. Seed bacterization with the isolate positively influenced the growth and nutrient uptake parameters of wheat seedlings cv. VL 804 in pot culture conditions at controlled cold growing temperature. This is an early report on the phosphate solubilization and plant growth promotion by Pseudomonas lurida, which is a relatively new species of the genus Pseudomonas and opens up a hitherto unknown facet of this bacterium.  相似文献

Abstract

A study of the temporal synergism of serotonergic and dopaminergic activity in the regulation of reproductive seasonality of photosensitive and photorefractory migratory Red headed bunting was undertaken. In experiment 1, groups of birds kept in natural day length (NDL) in their photosensitive phase (April) received daily injections of L‐DOPA (L‐dihydroxyphenylalanine, a dopamine precursor) at 8 and 12 hr after 5‐HTP (5 hydroxytryptophan, a serotonin precursor) administration. One control group received two daily injections of saline. The injections with 12‐hr temporal relation induced early recrudescence and full gonadal growth was achieved much in advance to that of control birds. The applications with 8‐hr temporal relation suppressed the testicular growth initially but later on this effect was eliminated by NDL of May‐June. In Experiment 2, photorefractory birds also received the treatments but the temporal relation of 5‐HTP and L‐DOPA had no effect on the regressed gonads of these birds.

This study indicates that injections with specific phase relations of dopamine and serotonin precursors may alter seasonal reproductive conditions only in the photosensitive phase but had no effect in photorefractory birds.  相似文献
