Functional domains in introns: trans-acting and cis-acting regions of intron 4 of the cob gene 总被引:21,自引:0,他引:21
We have sequenced the mutational changes in eight mutants in the open reading frame of intron 4 of the cob gene on yeast mitochondrial DNA. Three have a cis-acting splicing defect, while the other inactivate a trans-recessive intron domain that specifies a trans-acting splicing factor. From phenotypic evidence, including analyses of the allele-specific extra proteins, we have identified a protein (P27) encoded wholly within the intron that appears to be the intron 4 splicing factor (maturase). The evidence suggests that P27 is a secondary translation product resulting from the proteolytic cleavage of a larger precursor encoded by exon and intron sequences, but an alternative model, in which P27 is a primary translation product, has not been ruled out. 相似文献

Agro-ecosystems have recently experienced dramatic losses of biodiversity due to more intensive production methods. In order
to increase species diversity, agri-environment schemes provide subsidies to farmers who devote a fraction of their land to
ecological compensation areas (ECAs). Several studies have shown that invertebrate biodiversity is actually higher in ECAs
than in nearby intensively cultivated farmland. It remains poorly understood, however, to what extent ECAs also favour vertebrates,
such as small mammals and their predators, which would contribute to restoring functional food chains within revitalised agricultural
matrices. We studied small mammal populations among eight habitat types—including wildflower areas, a specific ECA in Switzerland—and
habitat selection (radiotracking) by the Barn Owl Tyto alba, one of their principal predators. Our prediction was that habitats with higher abundances of small mammals would be more
visited by foraging Barn Owls during the period of chicks’ provisioning. Small mammal abundance tended to be higher in wildflower
areas than in any other habitat type. Barn Owls, however, preferred to forage in cereal fields and grassland. They avoided
all types of crops other than cereals, as well as wildflower areas, which suggests that they do not select their hunting habitat
primarily with respect to prey density. Instead of prey abundance, prey accessibility may play a more crucial role: wildflower
areas have a dense vegetation cover, which may impede access to prey for foraging owls. The exploitation of wildflower areas
by the owls might be enhanced by creating open foraging corridors within or around wildflower areas. Wildflower areas managed
in that way might contribute to restore functional links in food webs within agro-ecosystems. 相似文献

Background
Yeast surface display is a technique, where the proteins of interest are expressed as fusions with yeast surface proteins and thus remain attached to the yeast cell wall after expression. Our purpose was to study whether allergens expressed on the cell surface of baker's yeast Saccharomyces cerevisiae preserve their native allergenic properties and whether the yeast native surface glycoproteins interfere with IgE binding. Multicomponent signals consist of several traits that are perceived as a whole. Although many animals rely on multicomponent signals to communicate, the selective pressures shaping these signals are still poorly understood. Previous work has mainly investigated the evolution of multicomponent signals by studying each trait individually, which may not accurately reflect the selective pressures exerted by the holistic perception of signal receivers. Here, we study the design of the multicoloured face of an Old World primate, the mandrill (Mandrillus sphinx), in relation to two aspects of signalling that are expected to be selected by receivers: conspicuousness and information. Using reflectance data on the blue and red colours of the faces of 34 males and a new method of hue vectorisation in a perceptual space of colour vision, we show that the blue hue maximises contrasts to both the red hue and the foliage background colouration, thereby increasing the conspicuousness of the whole display. We further show that although blue saturation, red saturation and the contrast between blue and red colours are all correlated with dominance, dominance is most accurately indicated by the blue-red contrast. Taken together our results suggest that the evolution of blue and red facial colours in male mandrills are not independent and are likely driven by the holistic perception of conspecifics. In this view, we propose that the multicoloured face of mandrills acts as a multicomponent signal. Last, we show that information accuracy increases with the conspicuousness of the whole display, indicating that both aspects of signalling can evolve in concert. 相似文献

This study reports a phytoremediation pot experiment using an open-source program. Unsaturated water flow was described by the Richards' equation and solute transport by the advection-dispersion equation. Sink terms in the governing flow and transport equations accounted for root water and solute uptake, respectively. Experimental data were related to application of Vetiver grass to soil contaminated by metal ions. Sensitivity analysis revealed that due to the specific experimental set-up (bottom flux not allowed), hydraulic model parameters did not influence root water (and contaminant) uptake. In contrast, the results were highly correlated with plant solar radiation interception efficiency (leaf area index). The amounts of metals accumulated in the plant tissue were compared to numerical values of cumulative uptake. Pb2+ and Zn2+ uptake was satisfactorily described using a passive model. However, for Ni2+ and Cd2+, a specific calibration of the active uptake model was necessary. Calibrated MM parameters for Ni2+, Cd2+, and Pb2+ were compared to values in the literature, generally suggesting lower rates and saturation advance. A parameter (saturation ratio) was introduced to assess the efficiency of contaminant uptake. Numerical analysis, applying actual field conditions, showed the limitation of the active model for being independent of the transpiration rate. 相似文献

Nucleocytoplasmic transport occurs through nuclear pore complexes (NPCs) embedded in the nuclear envelope. Here, we discovered an unexpected role for yeast dynein light chain (Dyn2) in the NPC. Dyn2 is a previously undescribed nucleoporin that functions as molecular glue to dimerize and stabilize the Nup82-Nsp1-Nup159 complex, a module of the cytoplasmic pore filaments. Biochemical analyses showed that Dyn2 binds to a linear motif (termed DID(Nup159)) inserted between the Phe-Gly repeat and coiled-coil domain of Nup159. Electron microscopy revealed that the reconstituted Dyn2-DID(Nup159) complex forms a rigid rod-like structure, in which five Dyn2 homodimers align like 'pearls on a string' between two extented DID(Nup159) strands. These findings imply that the rigid 20 nm long Dyn2-DID(Nup159) filament projects the Nup159 Phe-Gly repeats from the Nup82 module. Thus, it is possible that dynein light chain plays a role in organizing natively unfolded Phe-Gly repeats within the NPC scaffold to facilitate nucleocytoplasmic transport. 相似文献

This study tested for behavioral and hormonal reactions of young men to brief social encounters with potential mating partners. Male college students were randomly assigned to engage in a short conversation with either a young man (male condition) or a young woman (female condition). Participants provided saliva samples before and after the conversation, completed a battery of psychological measures after the interaction, and had their behavior rated by their conversation partners. Salivary testosterone (T) increased significantly over baseline levels in the female condition only, though differences between conditions were not significant. In addition, change in T was significantly correlated with the degree to which the female confederates thought the male participants were trying to impress them. These behavioral ratings, in turn, were correlated with the participants' ratings of the female confederates as potential romantic partners. Results were generally consistent with the hypothesis that human males may exhibit a behavioral and endocrine courtship response that is similar to that observed in males of many nonhuman vertebrate species. 相似文献

Otolith morphology is a widely accepted tool for species identification in teleost fish, but whether this holds true for very small species remains to be explored. Here, the saccular otoliths of the cryptobenthic Mediterranean clingfish Gouania (Gobiesocidae) are described for the first time. The new data, although preliminary, indicate that otolith morphology and morphometry support the recognition of the recently differentiated five species of Gouania in the Mediterranean Sea. Furthermore, otoliths of phylogenetically closely related Gouania species resemble each other more than do those of the more distantly related species. 相似文献

In humans, eight types of histone H1 exist (H1.1-H1.5, H1 degrees , H1t and H1oo), all consisting of a highly conserved globular domain and less conserved N- and C-terminal tails. Although the precise functions of these isoforms are not yet understood, and H1 subtypes have been found to be dispensable for mammalian development, it is now clear that specific functions may be assigned to certain individual H1 subtypes. Moreover, microsequence variations within the isoforms, such as polymorphisms or mutations, may have biological significance because of the high degree of sequence conservation of these proteins. This study used a hydrophilic interaction liquid chromatographic method to detect sequence variants within the subtypes. Two deviations from wild-type H1 sequences were found. In K562 erythroleukemic cells, alanine at position 17 in H1.2 was replaced by valine, and, in Raji B lymphoblastoid cells, lysine at position 173 in H1.4 was replaced by arginine. We confirmed these findings by DNA sequencing of the corresponding gene segments. In K562 cells, a homozygous GCC-->GTC shift was found at codon 18, giving rise to H1.2 Ala17Val because the initial methionine is removed in H1 histones. Raji cells showed a heterozygous AAA-->AGA codon change at position 174 in H1.4, corresponding to the Lys173Arg substitution. The allele frequency of these sequence variants in a normal Swedish population was found to be 6.8% for the H1.2 GCC-->GTC shift, indicating that this is a relatively frequent polymorphism. The AAA-->AGA codon change in H1.4 was detected only in Raji cells and was not present in a normal population or in six other cell lines derived from individuals suffering from Burkitt's lymphoma. The significance of these sequence variants is unclear, but increasing evidence indicates that minor sequence variations in linker histones may change their binding characteristics, influence chromatin remodeling, and specifically affect important cellular functions. 相似文献
