We describe in this paper the preparation and characterization of semicarbazide glass slides and their use for the fabrication of microarrays using site-specific alpha-oxo semicarbazone ligation. The functional density and homogeneity of the semicarbazide glass slides were optimized by analyzing the reactivity of the layer toward a synthetic glyoxylyl fluorescent probe. Oligonucleotide microarrays were prepared by site-specific immobilization of glyoxylyl oligodeoxynucleotides. The slides were directly used in the hybridization assays using fluorescence detection and displayed a significant gain in sensibility as compared to the aldehyde glass slide/amino oligodeoxynucleotide chemistry. Semicarbazide slides were also used for the immobilization of a biotinylated peptide alpha-oxo aldehyde. The peptide microarrays allowed model interaction studies with streptavidin or an anti-biotin antibody. 相似文献

Naked mole-rats are highly social and strictly subterranean rodents that live in large communal colonies in sealed and chronically oxygen-depleted burrows. Brain slices from naked mole-rats show extreme tolerance to hypoxia compared to slices from other mammals, as indicated by maintenance of synaptic transmission under more hypoxic conditions and three fold longer latency to anoxic depolarization. A key factor in determining whether or not the cellular response to hypoxia is reversible or leads to cell death may be the elevation of intracellular calcium concentration. In the present study, we used fluorescent imaging techniques to measure relative intracellular calcium changes in CA1 pyramidal cells of hippocampal slices during hypoxia. We found that calcium accumulation during hypoxia was significantly and substantially attenuated in slices from naked mole-rats compared to slices from laboratory mice. This was the case for both neonatal (postnatal day 6) and older (postnatal day 20) age groups. Furthermore, while both species demonstrated more calcium accumulation at older ages, the older naked mole-rats showed a smaller calcium accumulation response than even the younger mice. A blunted intracellular calcium response to hypoxia may contribute to the extreme hypoxia tolerance of naked mole-rat neurons. The results are discussed in terms of a general hypothesis that a very prolonged or arrested developmental process may allow adult naked mole-rat brain to retain the hypoxia tolerance normally only seen in neonatal mammals. 相似文献

Aim Increasing geographical range and density of conifers is a major form of land‐cover change in the western United States, affecting fire frequency, biogeochemistry and possibly biodiversity. However, the extent and magnitude of the change are uncertain. This study aimed to quantify the relationship between changing conifer cover and topography. Location The central Great Basin in the state of Nevada, USA. Methods We used a series of Landsat Thematic Mapper satellite images from 1986, 1995 and 2005 to map change in pinyon–juniper woodlands (Pinus monophylla, Juniperus spp.) in the montane central Great Basin of Nevada. We derived fractional greenness for each year using spectral mixture analysis and identified all areas with an above average increase in greenness from 1986 to 1995 and 1995 to 2005. Results Areas with high fractional greenness in 2005 were most likely to occur at elevations between 2200 and 2600 m a.s.l. Increases in fractional greenness between 1986 and 2005 were most likely to occur at elevations below 2000 m a.s.l. and on south‐facing slopes. However, relationships between elevation and increasing greenness for individual mountain ranges varied considerably from the average trend. Fractional greenness values measured by Landsat suggest that the majority of pinyon–juniper woodlands have not reached their maximum potential tree cover. Main conclusions Expansion of pinyon–juniper at low elevations and on south‐facing slopes probably reflects increasing precipitation in the 20th century, higher water use efficiency caused by increasing atmospheric CO2 in the late 20th century and livestock grazing at the interface between shrubland and woodland. Identification of the spatial relationships between changing fractional greenness of pinyon–juniper woodland and topography can inform regional land management and improve projections of long‐term ecosystem change. 相似文献

In Saccharomyces cerevisiae, the sequence-specific binding of the negative regulator Rap1p provides a mechanism to measure telomere length: as the telomere length increases, the binding of additional Rap1p inhibits telomerase activity in cis. We provide evidence that the association of Rap1p with telomeric DNA in vivo occurs in part by sequence-independent mechanisms. Specific mutations in EST2 (est2-LT) reduce the association of Rap1p with telomeric DNA in vivo. As a result, telomeres are abnormally long yet bind an amount of Rap1p equivalent to that observed at wild-type telomeres. This behavior contrasts with that of a second mutation in EST2 (est2-up34) that increases bound Rap1p as expected for a strain with long telomeres. Telomere sequences are subtly altered in est2-LT strains, but similar changes in est2-up34 telomeres suggest that sequence abnormalities are a consequence, not a cause, of overelongation. Indeed, est2-LT telomeres bind Rap1p indistinguishably from the wild type in vitro. Taken together, these results suggest that Est2p can directly or indirectly influence the binding of Rap1p to telomeric DNA, implicating telomerase in roles both upstream and downstream of Rap1p in telomere length homeostasis. 相似文献

Understanding the mechanisms limiting the distributions of organisms is necessary for predicting changes in community composition
along habitat gradients. In many areas of the USA, land originally cleared for agriculture has been undergoing a process of
reforestation, creating a gradient of canopy cover. For small temporary wetlands, this gradient can alter abiotic conditions
and influence the resource base of wetland food webs by affecting litter inputs. As distributions of amphibians and many other
temporary wetland taxa correlate with canopy cover, we experimentally manipulated shade levels and litter types in pond mesocosms
to explore mechanisms limiting species performance in wetlands with canopy cover. Most differences between ponds were mediated
by litter type rather than direct effects of shading. Although all three amphibian species tested are open-canopy specialists,
spring peepers were the only species to show decreased survival in shaded ponds. Pond litter type generally had strong effects
on growth and development rates, with tadpoles of two species in grass litter ponds growing to twice the size of, and metamorphosing
7 days earlier than, those in leaf litter ponds. Contrary to our initial hypothesis, shade level and litter type showed very
few significant interactions. Our results indicate that the effects of shading cannot be considered in isolation of vegetation
changes in pond basins when evaluating the effects of forest succession on temporary pond communities. 相似文献

Mek is a dual-specificity kinase that activates the extracellular-signal-regulated (Erk) mitogen-activated protein (MAP) kinases upon agonist binding to receptors. The Erk MAP kinase cascade is involved in cell-fate determination in many organisms. In mammals, this pathway is proposed to regulate cell growth and differentiation. Genetic studies have shown that although a single mek gene is present in Caenorhabditis elegans, Drosophila and Xenopus, two mek homologs, Mek1 and Mek2, are present in the mammalian cascade. In the present study, we describe a mutant mouse line in which the mek1 gene has been disrupted by insertional mutagenesis. The null mutation was recessive lethal, as the homozygous mutant embryos died at 10.5 days of gestation. Histopathological analyses revealed a reduction in vascularization of the placenta that was due to a marked decrease of vascular endothelial cells in the labyrinthine region. The failure to establish a functional placenta probably explains the death of the mek1-/- embryos. Cell-migration assays indicated that mek1-/- fibroblasts could not be induced to migrate by fibronectin, although the levels of Mek2 protein and Erk activation were normal. Re-expression of Mek1 in the mutant mouse embryonic fibroblasts (MEFs) restored their ability to migrate. Our findings provide genetic evidence that establishes the unique role played by Mek1 in signal transduction. They also suggest that mek1 function is required for normal response to angiogenic signals that might promote vascularization of the labyrinthine region of the placenta. 相似文献

A link between leptin resistance, obesity, and salt sensitivity has been suggested. SHHF/Mcc-fa
cp rats (SHHF) were used to study the effect of gene dosage of a null mutation of the leptin receptor (cp) on salt sensitivity and response to a combined endothelin A and B receptor antagonist (bosentan). Obese (cp/cp), heterozygous (+/cp), and homozygous lean (+/+) male SHHF were fed a low salt diet (0.3% NaCl) for 7 days, followed by a high salt diet (8.0% NaCl) for 7 days. There were no significant differences in systolic blood pressure between genotypes on low salt. In response to high salt, cp/cp had significantly greater systolic pressure than +/cp and +/+. On high salt diet, cp/cp showed a significant increase in 24 h urinary endothelin excretion and increased renal expression of preproendothelin mRNA. There was no effect of high salt diet on renal excretion of nitric oxide (NOx) or on gene expression of endothelial, neuronal, or cytokine-induced nitric oxide synthase isoforms (eNOS, nNOS, iNOS, respectively). Treatment with bosentan prevented the high salt-induced increment in systolic blood pressure in cp/cp. This was associated with a doubling of renal NOx excretion, but without changes in eNOS, nNOS, or iNOS expression. Endothelin receptor antagonism did not normalize systolic pressure in any of the genotypes. Our studies indicate that obesity secondary to leptin resistance (cp/cp) results in increased salt sensitivity that is mediated by endothelin in the SHHF rat. 相似文献
