The effects of changes in photoperiod length upon body weight; spleen, thymus, and testis weights; testis protein content; testis cation pump enzyme activities; and plasma testosterone were studied in the developing Siberian hamster, Phodopus sungorus. Male hamsters were exposed to a cycle of 16L:8D (long-day), until Day 18 when half were switched to a 10L:14D (short-day) cycle, until killed 0, 2, 4, 7, 10, 12, or 15 days later. Body weight and relative testis weight (expressed as percentage of body weight) increased steadily during the first week of exposure. After 10 days, the long-day hamsters consistently weighed more (p less than 0.05). Relative testis weights in the short-day group began to decrease (p less than 0.005) within 10 days and continued to decline. Testis homogenate K(+)-pNPPase- (as a measure of Na+,K(+)-ATPase) and Mg(2+)-pNPPase-specific activities closely paralleled testis weight, with the short-day animals (p less than 0.05) differing after 10 days. Plasma testosterone levels remained below adult levels through exposure Day 15, but were relatively lower (p less than 0.05) in the short-day group after 10 days. Spleen weights were similar for the long- and short-day groups. The short-day group had larger thymus weights after 12 days (p less than 0.05), but thymus enzyme activities did not differ between the two groups. We conclude that cation pump activities in the Siberian hamster testis are significantly affected by changes in photoperiod length.  相似文献

Abstract: We have compared the competitive inhibitory effects of 2-deoxyglucose, glucosamine, N -acetylglucosamine, N -benzoylglucosamine, and the commonly used radiographic and density gradient agent metrizamide (2-[3 - acetamido - 2,4,6 - triiodo -5-( N - methylacetamido) benzamido]-2-deoxyglucose) on the mitochondrial and soluble forms of human brain hexokinase. Metrizamide produces a classical competitive inhibition with glucose for human brain hexokinase, with K is of 2.8 and 2.5 m M , respectively, for the mitochondrial and soluble forms. Glucosamine exhibited K is of 0.58 and 0.29 m M , while 2-deoxyglucose exhibited K is of 0.074 and 0.15 m M and N -acetylglucosamine 0.098 and 0.092 m M for these two forms, respectively. N -Benzoylglucosamine was by far the most effective inhibitor tested, with K i values of 0.0086 and 0.022 m M , respectively. In order of increasing potency as a competitive inhibitor for mitochondrial hexokinase are metrizamide, glucosamine, N -acetylglucosamine, 2-deoxyglucose, and N -benzoylglucosamine. For the soluble form of the enzyme in increasing potency are metrizamide, glucosamine, 2-deoxyglucose, N -acetylglucosamine, and N -benzoylglucosamine. Since N -benzoylglucosamine was over 100 times more potent than metrizamide, some of the effects of metrizamide could be due to contamination by N -benzoylglucosamine. However, gas chromatography-mass spectrometry analysis of metrizamide did not indicate the presence of N -benzoylglucosamine. In addition, column chromatographic separation of commercially available metrizamide and reconstitution of freeze-dried eluate fractions localized the inhibitory effect to the metrizamide peak.  相似文献

Cells of Mikania glomerata, Cephaelis ipecacuanha and Maytenus aquifolia were co-cultured in a two-phase system using filter paper as a solid support. The species were co-cultured in all possible paired combinations. Interaction between Mikania and Maytenus cells resulted in increased biomass production of Maytenus cells, but the friedelin content was reduced. Co-cultivation of Cephaelis and Mikania cells enhanced coumarin content, but inhibited the growth of Mikania cells. However, yield of emetine as well as Cephaelis biomass accumulation were positively stimulated by the co-cultivation. Results indicate a possible occurrence of allelopathy in such a system. Pseudomonas aeruginosa is an opportunistic pathogen associated with severe diseases, such as cystic fibrosis. During an extensive search for novel essential genes, we identified tgpA (locus PA2873) in P. aeruginosa PAO1, as a gene playing a critical role in bacterial viability. TgpA, the translated protein, is an internal membrane protein with a periplasmic soluble domain, predicted to be endowed with a transglutaminase-like fold, hosting the Cys404, His448, and Asp464 triad. We report here that Cys404 mutation hampers the essential role of TgpA in granting P. aeruginosa viability. Moreover, we present the crystal structure of the TgpA periplasmic domain at 1.6?Å resolution as a first step towards structure–activity analysis of a new potential target for the discovery of antibacterial compounds.  相似文献

The climatic cycles with subsequent glacial and intergalcial periods have had a great impact on the distribution and evolution of species. Using genetic analytical tools considerably increased our understanding of these processes. In this review I therefore give an overview of the molecular biogeography of Europe. For means of simplification, I distinguish between three major biogeographical entities: (i) "Mediterranean" with Mediterranean differentiation and dispersal centres, (ii) "Continental" with extra-Mediterranean centres and (iii) "Alpine" and/or "Arctic" with recent alpine and/or arctic distribution patterns. These different molecular biogeographical patterns are presented using actual examples. Many "Mediterranean" species are differentiated into three major European genetic lineages, which are due to glacial isolation in the three major Mediterranean peninsulas. Postglacial expansion in this group of species is mostly influenced by the barriers of the Pyrenees and the Alps with four resulting main patterns of postglacial range expansions. However, some cases are known with less than one genetic lineage per Mediterranean peninsula on the one hand, and others with a considerable genetic substructure within each of the Mediterranean peninsulas, Asia Minor and the Maghreb. These structures within the Mediterranean sub-centres are often rather strong and in several cases even predate the Pleistocene. For the "Continental" species, it could be shown that the formerly supposed postglacial spread from eastern Palearctic expansion centres is mostly not applicable. Quite the contrary, most of these species apparently had extra-Mediterranean centres of survival in Europe with special importance of the perialpine regions, the Carpathian Basin and parts of the Balkan Peninsula. In the group of "Alpine" and/or "Arctic" species, several molecular biogeographical patterns have been found, which support and improve the postulates based on distribution patterns and pollen records. Thus, genetic studies support the strong linkage between southwestern Alps and Pyrenees, northeastern Alps and Carpathians as well as southeastern Alps and the Dinaric mountain systems, hereby allowing conclusions on the glacial distribution patterns of these species. Furthermore, genetic analyses of arctic-alpine disjunct species support their broad distribution in the periglacial areas at least during the last glacial period. The detailed understanding of the different phylogeographical structures is essential for the management of the different evolutionary significant units of species and the conservation of their entire genetic diversity. Furthermore, the distribution of genetic diversity due to biogeographical reasons helps understanding the differing regional vulnerabilities of extant populations.  相似文献

AimsThe effects of chronic administration of Ferutinin (phytoestrogen found in the plants of genus Ferula), compared with those elicited by estradiol benzoate, were evaluated, following ovariectomy, on the uterus of ovariectomized rats as regard weight, size, structure and histomorphometry.Main methodsThe experimental study included 40 female Sprague–Dawley rats, assigned to two different protocols, i.e. preventive and recovering. In the preventive protocol, ferutinin (2 mg/kg/day) was orally administered for 30 days, starting from the day after ovariectomy; in the recovering protocol, ferutinin was administered, at the same dosage, for 30 days starting from the 60th day after ovariectomy, when osteoporosis was clearly established. Its effects were compared with those of estradiol benzoate (1.5 μg per rat twice a week, subcutaneously injected) vs. vehicle-treated ovariectomized controls and vehicle-treated sham-operated controls. Uteri were removed, weighed and analysed under both the structural and histomorphometrical points of view.Key findingsOur data show that ferutinin acts, similarly to estradiol benzoate, on the uterus stimulating endometrial and myometrial hypertrophy; this notwithstanding, the phytoestrogen ferutinin, in contrast to estrogen treatment, appears to increase apoptosis in uterine luminal and glandular epithelia.SignificanceFerutinin, used in osteoporosis treatment primarily for bone mass recovering, seems in line with an eventual protective function against uterine carcinoma, unlike estrogens so far employed in hormone replacement therapy (HRT).  相似文献

In a deep, subalpine holo-oligomictic lake, the relative abundance of Archaea and Crenarchaeota, but not that of Bacteria, increases significantly with depth and varies seasonally. Cell-specific prokaryotic productivity is homogeneous along the water column. The concept of active Archaea observed in the deep ocean can therefore be extended to a deep oxic lake.The abundance, activity, and community composition of epilimnetic and hypolimnetic prokaryotes have been less thoroughly investigated in deep lakes than in oceans. Strong evidence that the depth gradient plays a role in modulating the balance between the domains of Bacteria and Archaea has been found in various marine systems (8, 12, 13, 20). It has been shown that the percentage of Bacteria in the deep marine hypolimnion decreases (up to 5,000 m) while, conversely, the percentage of Archaea increases. The percentage of Crenarchaeota is also higher in the mesopelagic zone than in surface waters (10).Although Archaea have been found in a variety of freshwater habitats (3), little has thus far been published on differentiating between Bacteria, Archaea, and Crenarchaeota in the hypolimnion of deep lakes. An exception is a study of the high-altitude ultraoligotrophic Crater Lake (21, 22), where group I marine Crenarchaeota were observed in deep-water populations (22). This study and another study of various lakes from three continents (9) are based on summer sampling, making it impossible to ascertain the effects of temporal variability on the vertical distribution of Archaea and Crenarchaeota, as has been done for marine systems and shallow lakes (for examples, see references 8 and 11).Our primary objective was to follow variations in the relative abundance of Bacteria, Archaea, and Crenarchaeota found in the hypolimnetic waters of a deep holo-oligomictic lake with a permanent oxic hypolimnion and compare them with those in the epilimnetic assemblages. We used the catalyzed reporter deposition fluorescence in situ hybridization (CARD-FISH) technique and compared the data thus obtained with prokaryotic productivity.  相似文献

In this study we analyzed a sample of the urban population of La Plata, Argentina, using 17 mtDNA haplogroups, the DYS 199 Y-chromosome polymorphism, and 5 autosomal population-associated alleles (PAAs). The contribution of native American maternal lineages to the population of La Plata was estimated as 45.6%, whereas the paternal contribution was much lower (10.6%), clearly indicating directional mating. Regarding autosomal evidence of admixture, the relative European, native American, and West African genetic contributions to the gene pool of La Plata were estimated to be 67.55% (+/-2.7), 25.9% (+/-4.3), and 6.5% (+/-6.4), respectively. When admixture was calculated at the individual level, we found a low correlation between the ancestral contribution estimated with uniparental lineages and autosomal markers. Most of the individuals from La Plata with a native American mtDNA haplogroup or the DYS199*T native American allele show a genetic contribution at the autosomal level that can be traced primarily to Europe. The results of this study emphasize the need to use both uniparentally and biparentally inherited genetic markers to understand the history of admixed populations.  相似文献

The aim of this study was to evaluate the possibility to use automated EcoRI ribotyping to address, during the same analysis, both identification and genetic characterisation of 38 Staphylococcus aureus and 64 coagulase-negative staphylococci collected from surgical injuries. The ribotyping identification results confirmed those obtained using the API Staph system for 96% of the isolates. All strains were successfully genotyped and the ribotyping discriminatory power, calculated using the Simpson's index of discrimination, was very high for both groups of staphylococci tested. The same, as well as different biotypes, were identified among isolates with the identical ribotyping profile.  相似文献
