Uptake of GABA was demonstrated in rat brain synaptic vesicles which were prepared by a new and efficient procedure. The uptake activity co-purified with the synaptic vesicles during the isolation procedure. The purity of the vesicle fraction was rigorously examined by analysis of marker enzymes and marker proteins and also by immunogold electron microscopy using antibodies against p38 (synaptophysin). Contamination by other cellular components was negligible, indicating that GABA uptake by the synaptic vesicle fraction is specific for synaptic vesicles and not due to the presence of other structure possessing GABA uptake or binding activities. GABA uptake was ATP dependent and similar to the uptake of glutamate, which was assayed for a comparison. Both uptake activities were independent of sodium. They were inhibited by the uncoupler carbonyl cyanide 4-(trifluoromethoxy)phenylhydrazone, indicating that the energy for the uptake is provided by an electrochemical proton gradient. This gradient is generated by a proton ATPase of the vacuolar type as suggested by the effects of various ATPase inhibitors on neurotransmitter uptake and proton pumping. Competition experiments revealed that the transporters for GABA and glutamate are selective for the respective neurotransmitters.  相似文献

Summary In females of the acridid grasshopperChorthippus biguttulus, thoracic auditory interneurons were investigated with respect to their selectivity for temporal parameters of the conspecific song. Special attention was given to the detection of small gaps in the syllables of the song, since behavioural experiments have shown that the presence or absence of gaps is critical for the female's Innate Releasing Mechanism (cf. Fig. 1).The spiking response of one ascending interneuron, the AN4, shows filtering properties which closely resemble the behavioural reactions (cf. Figs. 1, 3 and 5b). The difference in the AN4's reaction to stimuli with gaps and uninterrupted stimuli is maintained over the behaviourally relevant intensity range (Fig. 4). This reaction is reliable enough that the stimulus type could be inferred by higher centres even from single stimulus presentations. Hence, this neuron is likely to participate in the task of gap detection and probably is a part of the neuronal filter network which determines the characteristics of the Innate Releasing Mechanism of this species. However, this interneuron is not species-specific: A homologue exists in other acridids as well and, inLocusta migratoria, has similar response characteristics (Fig. 6). Water culture, growth chamber, greenhouse and field experiments were conducted to compare the effect of NH4−N and NO3−N on yield and N uptake of rapeseed (Brassica campestris L.). In water culture, the yields of 28-day old rapeseed plants grown at 14 μg N ml−1 were double with NO3 compared to NH4, but N uptake was little affected. There was no such effect when concentration was reduced to 3.5 or 7 μg N ml−1. The yield and N uptake of 26-day old rapeseed grown on six soils (pH 4.6 to 6.5) in pots in a growth chamber were much greater with NO3 than with NH4, although N concentration was more in the NH4- than the NO3-grown plants. In a greenhouse experiment with rapeseed grown on 12 potted soils, the N uptake of applied N was greater with NO3 than with NH4 on all soils. Averages were 63% with NH4 and 78% with NO3. However, NH4-fixation capacities of the soils were only weakly correlated with yield from the two sources of N (r=0.48) and the relation was similar with N uptake. In contrast to the behavior of water culture, growth chamber and greenhouse experiments, the 33 field experiments did not show consistent difference in seed yield with NH4 and NO3 applied at time of seeding. In nine field experiments where band application was used for Ca(NO3)2, (NH4)2 SO4, NH4 NO3, yield tended to be greatest for (NH4)2SO4. However, in 19 experiments on acid soils with and without lime, yields in most cases were similar with (NH4)2SO4 and NH4 NO3. Nitrification inhibitors were added to spring banded NH4-based fertilizers in five experiments, but the yields were not influenced. Abstract Poly(3-hydroxybutyric acid) granules, which harbored only four major granule-associated proteins as revealed by SDS polyacrylamide gel electrophoresis, were isolated from crude cellular extracts of Chromatium vinosum D by centrifugation in a linear sucrose gradient. N-Terminal amino acid sequence determination identified two proteins of M r 41 000 and M r 40 000 as the phaE Cv and phaC Cv translational products, respectively, of C. vinosum D. In a previous study it was shown that both proteins are required for the expression opf poly(3-hydroxyalkanoic acid) synthase activity. The N-terminus of the third protein ( M r 17 000) exhibited no homology to other proteins. Lysozyme, which was during purification of the granules, exhibited a strong affinity to PHB granules and was identified as the fourth protein enriched with the granules.  相似文献

Summary In bacteria 5-aminolevulinate, the universal precursor in the biosynthesis of the porphyrin nucleus of hemes, chlorophylls and bilins is synthesised by two different pathways: in non-sulphur purple bacteria (Rhodobacter) or Rhizobium 5-aminolevulinate synthase condenses glycine and succinyl-CoA into 5-aminolevulinate as is the case in mammalian cells and yeast. In cyanobacteria, green and purple sulphur bacteria, as in chloroplasts of higher plants and algae a three step pathway converts glutamate into 5-aminolevulinate. The last step is the conversion of glutamate 1-semialdehyde into 5-aminolevulinate. Using a cDNA clone encoding glutamate 1-semialdehyde aminotransferase from barley, genes for this enzyme were cloned from Synechococcus PCC6301 and Escherichia coli and sequenced. The popC gene of E. coli, previously considered to encode 5-aminolevulinate synthase, appears to be a structural gene for glutamate 1-semialdehyde aminotransferase. Domains with identical amino acid sequences comprise 48% of the primary structure of the barley, cyanobacterial and putative E. coli glutamate 1-semialdehyde aminotransferases. The cyanobacterial and barley enzymes share 72% identical residues. The peptide containing a likely pyridoxamine phosphate binding lysine is conserved in all three protein sequences.  相似文献

In this study we compared the properties of cytochrome-c oxidase (COX) in cultured fibroblasts from two patients with Leigh Syndrome with COX from control fibroblasts. The fibroblasts from patients showed decreased growth reates and elevated lactate production. COX activity of patients fibroblasts was about 25% of control. Kinetic studies with isolated mitochondria showed a higher Km for cytochrome c and a markedly reduced molecular turnover of COX from patients, indicating a different structure of the enzyme. A biphasic change of COX activity was obtained by titration of dodecylmaltoside solubilized mitochondria from control fibroblasts with increasing concentrations of anions. With patient mitochondria we found only the inhibiting phase of COX activity and, in contrast to control mitochondria, irreversible inhibition of COX activity by guanidinium chloride. ELISA titrations with monoclonal antibodies to subunit II, IV, Vab, VIac and VIIab indicated a normal amount of mitochondrial coded subunit II, but a reduced amound of nuclear coded subunits. The data indicate incompletely assembled nuclear coded subunits of COX from patient fibroblasts.  相似文献

Influences of the salt concentration on the fatty acid composition of Ectothiorhodospira species and other phototrophic purple bacteria have been analysed. Major fatty acids in bacteria of the genera Rhodobacter, Rhodopseudomonas, Chromatium, and Ectothiorhodospira were straight chain saturated and monounsaturated C-16 and C-18 fatty acids. Salt-dependent responses of all investigated bacteria revealed relations to their salt optima. Minimum values of C-16 and saturated fatty acids and maximum values of C-18 and unsaturated fatty acids were found at or close to the salt optima. Responses of Ectothiorhodospira mobilis upon changes in salinity were nearly identical, whether cells were grown in batch culture or in continuous culture with identical dilution rates at all salt concentrations. With increasing temperature, the fatty acid composition of Ectothiorhodospira mobilis and Ectothiorhodospira halophila strains showed decreasing portions of C-18 and of unsaturated fatty acids, while the contents of C-16 and saturated fatty acids increased.
