RNAs are capable of modulating immune responses by binding to specific receptors. Neutrophils represent the major fraction of circulating immune cells, but receptors and mechanisms by which neutrophils sense RNA are poorly defined. Here, we analyzed the mRNA and protein expression patterns and the subcellular localization of the RNA receptors RIG-I, MDA-5, TLR3, TLR7, and TLR8 in primary neutrophils and immortalized neutrophil-like differentiated HL-60 cells. Our results demonstrate that both neutrophils and differentiated HL-60 cells express RIG-I, MDA-5, and TLR8 at the mRNA and protein levels, whereas TLR3 and TLR7 are not expressed at the protein level. Subcellular fractionation, flow cytometry, confocal laser scanning microscopy, and immuno-transmission electron microscopy provided evidence that, besides the cytoplasm, RIG-I and MDA-5 are stored in secretory vesicles of neutrophils and showed that RIG-I and its ligand, 3p-RNA, co-localize at the cell surface without triggering neutrophil activation. In summary, this study demonstrates that neutrophils express a distinct pattern of RNA recognition receptors in a non-canonical way, which could have essential implications for future RNA-based therapeutics.  相似文献

Uromyces umiamensis is described as a new rust fungus (Basidiomycota, Uredinales) of Momordica cochinchinensis (Cucurbitaceae) from Meghalaya, in northeastern India. The species differs from the known Uromyces species on members of Cucurbitaceae in terms of the character of the teliospores and aeciospores and its apparently demicyclic life cycle. Besides U. yakushimensis and the present species, all Uromyces species known from cucurbits occur in the New World. A key to known Uromyces species on Cucurbitaceae is provided.  相似文献

Introduction

Psoriasis is an inflammatory and immunological cutaneous disease. The high morbidity in patients with psoriasis results from severe clinical manifestations and/or adverse effects of treatment. The Undersea and Hyperbaric Medical Society and Federal Medicare and Medicaid Services have approved the use of hyperbaric oxygen (HBO2) for more than 15 indications, including wound healing, infections and late effects of radiation, which are largely unresponsive to conventional treatments. Accumulated data show that HBO2 has anti-inflammatory effects and other positive influences on the immune system, making it a rational treatment in the management of psoriasis plaques and arthritis.

Case presentation

We present the cases of two patients with long histories of psoriasis vulgarus who exhibited marked improvement with use of HBO2. The first patient was 40 years old and had pustular psoriasis and psoriatic arthritis. He was treated with six sessions of HBO2 (at 2.8 atmospheres of pressure for 60 minutes), which successfully controlled his symptoms. At the 18-month post-treatment follow up, the patient exhibited complete remission of psoriasis and marked improvement in psoriatic arthritis without medication. The second patient was 55 years old with extensive psoriatic lesions, and exhibited marked improvement within 15 sessions of HBO2. No adverse effects of HBO2 were identified.

Conclusions

HBO2 may possess potential therapeutic efficacy in the management of psoriasis. We outline the pathogenesis of psoriasis and the selective anti-inflammatory and immunosuppressive effects of HBO2. We hope that this will provide a basis for elucidating the mechanisms of action and consequently pave the way for further controlled studies.  相似文献

Localizing specific components in three-dimensional reconstructions of protein complexes visualized in an electron microscope increases the scientific value of those structures. Subunits are often identified within the complex by labeling; however, unless the label produces directly visible features, it must be detected by computational comparison with unlabeled complex. To bypass this step, we generated a cloneable tag from the actin-nucleating protein Spire that produces a directly visible “pointer” to the subunit after actin polymerization. We have used this new label to identify the intron of the C complex spliceosome to its small domain by fusing the 10 kDa Spire moiety to the affinity label that binds recombinant stem loops in the pre-mRNA substrate and assembling an actin filament on the particle.  相似文献

The spatial distribution of alien species richness often correlates positively with native species richness, and reflects the role of human density and activity, and primary productivity and habitat heterogeneity, in facilitating the establishment and spread of alien species. Here, we investigate the relationship between the spatial distribution of alien bird species, human density, and anthropogenic and natural environmental conditions. Next, we examined the relationship between the spatial distribution of alien bird species and native bird species richness. We examined alien species richness as a response variable, using correlative analyses that take spatial autocorrelation into account. Further, each alien bird species was examined as a response variable, using logistic regression procedures based on binary presence–absence data. A combination of human density and natural habitat heterogeneity best explained the spatial distribution of alien species richness. This contrasts with the results for individual alien species and with previous studies on other non-native taxa showing the importance of primary productivity and anthropogenic habitat modification as explanatory variables. In general, native species richness is an important correlate of the spatial distribution of alien species richness and individual alien species, with alien species being more similar to common species than to rare species.  相似文献

1 The dispersal abilities of agrioted beetles, serious pests on a variety of crops, are poorly known under natural conditions. This hampers their control. We used, for the first time, a stable isotope approach to assess dispersal of adult Agriotes obscurus in arable land.
2 After a diet switch from a C3- to a C4-plant, carbon isotope ratios of A. obscurus larvae significantly changed towards the isotopic signature of the new diet. Moreover, the larval δ13C signatures were transferred to the wing covers of the adult beetles with little distortion.
3 To assess the dispersal abilities under natural conditions, pheromone traps, lured for Agriotes sp., were installed at two study sites in Western Austria. Each site comprised a maize field (= C4-plant) and adjacent C3-grasslands with traps established along a transect of increasing distance to the maize.
4 δ13C signatures of wing covers revealed that adult male A. obscurus were able to migrate at least 80 m, which was the maximum distance that dispersal could be traced in the present study. The dispersal behaviour might have been influenced by site-specific factors.
5 The results obtained demonstrate a higher potential of adult male Agriotes to disperse than previously assumed. Moreover, the combination of pheromone trapping and stable isotope analysis proved to be an effective approach to study insect movement and dispersal in arable systems harbouring C3- and C4-crops.  相似文献

Kertesz et al. (Nature Genetics 2008) described PITA, a miRNA target prediction algorithm based on hybridization energy and site accessibility. In this note, we used a population genomics approach to reexamine their data and found that the PITA algorithm had lower specificity than methods based on evolutionary conservation at comparable levels of sensitivity.We also showed that deeply conserved miRNAs tend to have stronger hybridization energies to their targets than do other miRNAs. Although PITA had higher specificity in predicting targets than a naïve seed-match method, this signal was primarily due to the use of a single cutoff score for all miRNAs and to the observed correlation between conservation and hybridization energy. Overall, our results clarify the accuracy of different miRNA target prediction algorithms in Drosophila and the role of site accessibility in miRNA target prediction.  相似文献

Lignin is incorporated into plant cell walls to maintain plant architecture and to ensure long-distance water transport. Lignin composition affects the industrial value of plant material for forage, wood and paper production, and biofuel technologies. Industrial demands have resulted in an increase in the use of genetic engineering to modify lignified plant cell wall composition. However, the interaction of the resulting plants with the environment must be analyzed carefully to ensure that there are no undesirable side effects of lignin modification. We show here that Arabidopsis thaliana mutants with impaired 5-hydroxyguaiacyl O-methyltransferase (known as caffeate O-methyltransferase; COMT) function were more susceptible to various bacterial and fungal pathogens. Unexpectedly, asexual sporulation of the downy mildew pathogen, Hyaloperonospora arabidopsidis, was impaired on these mutants. Enhanced resistance to downy mildew was not correlated with increased plant defense responses in comt1 mutants but coincided with a higher frequency of oomycete sexual reproduction within mutant tissues. Comt1 mutants but not wild-type Arabidopsis accumulated soluble 2-O-5-hydroxyferuloyl-l-malate. The compound weakened mycelium vigor and promoted sexual oomycete reproduction when applied to a homothallic oomycete in vitro. These findings suggested that the accumulation of 2-O-5-hydroxyferuloyl-l-malate accounted for the observed comt1 mutant phenotypes during the interaction with H. arabidopsidis. Taken together, our study shows that an artificial downregulation of COMT can drastically alter the interaction of a plant with the biotic environment.  相似文献

The mitochondrial inner membrane contains numerous multispanning integral proteins. The precursors of these hydrophobic proteins are synthesized in the cytosol and therefore have to cross the mitochondrial outer membrane and intermembrane space to reach the inner membrane. While the import pathways of noncleavable multispanning proteins, such as the metabolite carriers, have been characterized in detail by the generation of translocation intermediates, little is known about the mechanism by which cleavable preproteins of multispanning proteins, such as Oxa1, are transferred from the outer membrane to the inner membrane. We have identified a translocation intermediate of the Oxa1 preprotein in the translocase of the outer membrane (TOM) and found that there are differences from the import mechanisms of carrier proteins. The intermembrane space domain of the receptor Tom22 supports the stabilization of the Oxa1 intermediate. Transfer of the Oxa1 preprotein to the inner membrane is not affected by inactivation of the soluble TIM complexes. Both the inner membrane potential and matrix heat shock protein 70 are essential to release the preprotein from the TOM complex, suggesting a close functional cooperation of the TOM complex and the presequence translocase of the inner membrane. We conclude that mitochondria employ different mechanisms for translocation of multispanning proteins across the aqueous intermembrane space.  相似文献
